dc.description.abstractEGFR family member (EGFR, ERBB2, ERBB3, ERBB4) 중 하나인 ERBB3의 경우 receptor tyrosine kinase 활성이 자연적으로 소실되었음에도 불구하고 배아발생에 필수적인 역할을 수행하는 것으로 알려져 있다. 이는 EGFR family member에 의해 ERBB3의 인산화가 일어나고 여기에 다양한 adaptor 단백질들이 붙어 신호전달을 매개하기 때문이라고 알려져 있다. 이들 adaptor 중 세포 분열과 사멸에 중요한 PI3K-AKT pathway의 기능을 분석하기 위하여, human ERBB3에 PI3K가 결합하지 못하는 유전자 변형 마우스 (Erbb37A) 모델을 제작하였다. 이들 마우스 모델을 이용하여 배아 발생 과정에서 ERBB3-PI3K/AKT pathway의 기능을 규명하고, 기존의 보고된 Erbb3 knockout 표현형질과 비교 분석을 통해 ERBB3-PI3K signaling pathway의 in vivo 기능을 규명하고자 하였다. Nrg1, Erbb2/3 pathway signaling은 말초신경계의 초기 발달에 중요한 역할을 하는 것으로 알려져 있는데, 그 대표적인 phenotype으로는 Schwann 세포가 결핍 되거나, myelination이 일어나지 않는 것으로 알려져 있다. Mouse knockout 연구에 따르면 이러한 defect phenotype은 EGFR ligands의 하나인 Nrg1 및 Erbb2/3 receptor가 관련되어 있으며, 이는 주요 downstream signaling pathways인 PI3K나 MAPK pathway에 이상이 발생하여 나타나는 현상으로 파악되고 있다. 이와 관련해 최근 ERK/MAPK, Shp, FAK, Calcineurin, Rac/cdc42, MAPK pathway 등의 유전자들의 knockout 접근 방법을 통해 유사한 phenotype이 보고가 되어 지고 있다. 우리의 연구에서 Erbb37A/7A 배아는 착상 후 12.5에서 13.5 사이에 죽었고, defect로는 neural crest 세포 유래의 Schwann 세포 precursor가 DRG와 nerves에서 소실 되어져 있는 것을 확인 할 수 있었다. 또한, DRG에서의 전체 증식 세포의 수와 Schwann 세포 precursor의 증식하는 세포의 수가 현저히 감소하는 것을 확인 할 수 있었다. Erbb3-/- 배아에서도 위와 동일 한 현상이 관찰 되었다. 그러나 Erbb3-/- 배아에서는 착상 후 12.5에서의 DRG의 크기가 현저히 감소해 있었고, apoptosis 세포의 수도 크게 증가 되어 있었다. 또한, 이것은 Schwann 세포 precursor 또는 neuron에서의 apoptosis 세포의 수가 현저히 증가하는 결과와 동일한 것 이였다. 이러한 결과는 in vitro에서 Nrg1, Erbb2/3 signaling을 통해 확인 한 결과 ERBB37A의 경우 phospho-AKT와 phospho-ERK1/2이 partially 발현되었고, ERBB3-는 거의 발현 되지 않았다. In vivo에서도 phospho-AKT와 phospho-ERK1/2가 모두 줄어 드는 것을 확인 할 수 있었는데 이러한 이유 때문에 defect의 차이로 예상 되어 진다. 이런 결과를 토대로 종합해 보면, Erbb37A/7A의 줄어든 phospho-AKT와 phospho-ERK1/2 signal들에 의해 말초신경계의 neural crest 세포 유래의 Schwann 세포 precursor의 이동과 증식에 관여하지만, Erbb3-/-의 경우 Schwann 세포 precursor 이외에도 neuron의 발달에도 영향을 미치는 것으로 보여진다. ;ERBB3 is a member of the EGFR family (EGFR, ERBB2, ERBB3, and ERBB4) of receptor tyrosine kinases (RTKs), and is known as an inactive kinase. Upon binding a direct ligand, ERBB3 is activated through heterodimerization with ERBB2. The cytoplasmic tail of ERBB3 is transphosphorylated and these phosphorylation sites cause the binding of signaling adaptors, mediating signal transduction. Through 7 phosphoinositide 3-kinase (PI3K) phosphotyrosine-binding sites, ERBB3 is able to recruit PI3K and initiate the PI3K/AKT signaling pathway. Since the PI3K/AKT signaling pathway is the most important downstream signaling pathway mediated by ERBB3, we addressed the significance of PI3K binding sites in ERBB3 using the ERBB3-PI3K mutant knockin mouse (Erbb37A) model and analyzed early peripheral nervous system (PNS) development during embryogenesis. In this study, we mutated 7 YXXM sequences in ERBB3 to YXXA to block ERBB3-dependent PI3K signaling. We observed phenotypic differences between Erbb37A/7A and Erbb3-/- in the development of neuronal and Schwann cells in the PNS. Erbb37A/7A embryos died at the E12.5 to E13.5 stage and were devoid of Schwann cell precursors (SCPs). Our results show that ERBB3-dependent PI3K signaling is essential for Schwann cell development but not neurons in the dorsal root ganglia. Further, although partial phosphorylation of Akt and extracellular signal-regulated kinase (ERK1/2) is observed in ERBB37A, it is nearly not seen in ERBB3-/- in vitro. Overall, the reduced ERK1/2 and Akt signaling in ERBB37A does not influence the development of PNS, but is required for proper SCP development.-
