Abstract
A3 아데노신 수용체에 높은 선택성을 지닌 효능제인 IB-MECA와 Cl-IB-MECA의 4' 위치를 황으로 치환한 Thio-Cl-IB-MECA, Northern C3′-endo 입체배치로 제한되어있는 탄소뉴클레오사이드의 5'-uronamide 위치에 메칠기를 붙임으로써 효과적으로 A3 아데노신 길항제로 치환할 수 있었다. 또한 5' 위치의 치환기를 제거하고 N6 위치를 아민기로 치환한 물질의 경우 기존 어떤 길항제보다 A3 아데노신 수용체에 높은 친화성을 보였으며, 다른 아류형의 아데노신 수용체에 비해 A3 아데노신 수용체에 높은 선택성을 가진 것으로 나타났다. 이와 같은 선행 연구에 기초하여 이번 연구에서는 기존 5' 위치의 치환기를 제거하고 N6 위치를 아민기로 치환한 물질의 3' 위치 알코올 기를 같은 수소결합에 참여할 있는 아민기로 치환하는 연구를 수행하였다. 타겟 물질은 acetoxy bromide를 key step으로 하여 입체선택적으로 합성되었다. 이렇게 합성된 3'-아미노뉴클레오사이드들은 CHO 세포들을 이용하여 활성연구가 진행 되었으나, 안타깝게도 모든 탄소뉴클레오사이드의 최종 물질들은 A3 아데노신 수용체에 활성을 보이지 않았다.;On the basis of bioisosteric rationale, the 3'-hydroxyl group of the truncated carbocyclic nucleosides, 4'-thionucleosides and 4'-oxonucleosides acting as potent and selective A3 adenosine receptor (AR) antagonists was substituted with the 3'-amino group which may act as a hydrogen bonding donor. The 3'-amino group was stereoselectively introduced using the regioselective synthesis of the acetoxy bromide as a key step. In the case of carbocyclic nucleoside, NaN3 was used to stereoselective synthesis of azide. Treatment with 2-acetoxyisobutyryl bromide gave the bromoacetate which was then treated with NaN3 to give the azido derivatives and its regioisomer. On the other hand, LiN3 was used to give the azido derivatives instead of NaN3 in 4'-thionucleosides series. In the case of 4'-oxonucleosides, the bromo derivative was treated with ammonia solution in t-butanol to give the desired 3'-amino derivative directly. These analogues are expected to bind similarly to the 3'-hydroxyl derivatives in the binding site of A3 AR.
