Abstract
일광 차단제는 일광의 과다 노출시 피부와 면역계에 미치는 해로운 영향을 예방. 치료하기 위해 개발된 이래, 현재는 피부 미용학적 용도로도 광범위하게 사용되고 있다. 그러므로 일광과 일광 차단제가 피부뿐 아니라, 생체내 대사물질에 미치는 영향에 대하여 연구하는 것은 중요하다고 사료된다. 이에 저자는 자외선과 일반적으로 널리 사용되고 있는 세 종류의 일광 차단제가 생체내 주요 대사물질에 미치는 영향을 관찰하고자 자외선에만 1주, 3주간 노출시킨 군과 일광 차단제 도포후 자외선에 1주, 3주간 노출시킨 군으로 구분하여 이들 실험 동물의 hepatic microsomal cytochrome P - 450의 활성 변화, 간조직내 2 -acetylaminofluorene (이하 2- AAF 로 약기함) 화합물의 ring-및 N-hydroxylation 생성의 변화 및 간조직내 vitamin D의 함량변화등을 관찰한 바 다음과 같은 결과를 얻었기에 보고하는 바이다. 1. 신생쥐의 hepatic microsomal cytochrome P - 450의 함량은 자외선에 3주간 노출시킨 군에서 현저한 증가를 보였으나( P〈0.001) 일광 차단제 도포후 자외선에 3주간 노출시킨 군에서는 이러한 변화가 크게 억제되었다(P〈0.05, P < 0.001). 2. 자외선에 의해 유도된 cytochrome P - 450은 간에 대해 발암원으로 알려진 2- AAF의 N - hydroxylation을 의의있게 증가 시켰으며 (P〈0.05) , ring - hydroxylation은 더욱 더 의의있게 증가 시켰다 (P〈0.001). 일광 차단제 도포후 자외선에 3주간 노출시킨 군에서는 2-AAF의 ring-과 N-hydroxylation이 모두 의의있게 억제 되었다(P〈0.05, p〈0.001 ). 3. 신생쥐의 간조직내 vitaminD의 함량은 자외선에 1주, 3주간 노출시 점차 증가하였으며 (P〈0.001 ) , 일광 차단제 도포 후 자외선에 노출시킨 군에서는 이러한 변화가 현저히 억제 되었다. (P〈0.0001).;Today, it is current situation that sunscreen has widely been used even for cosmetic dermatological purposes, since it was developed for the purpose of preventing and curing over-exposure of skin to natural sunlight and its harmful effects on skin and immune system. In addition to simple purpose to prevent sunburn due to sunlight, it is considered that it is very important for us to study effects of sunscreen on living body by comparatively examining various changes on skin and biological metabolites which may occur in both cases of not using sunscreen and using sunscreen. Considering such importance of study, in order to observe effects of 3 kinds' sunscreens being used widely, on important biological metabolites, first, I classified it into 2 categories of group which was exposed by sunlight for one (1) week and three (3) weeks, without using sunscreen, and another Group which were exposed by sunlight for one (1) week and three (3) weeks, after being applied by sunscreen, and then, I tried to observe changes of activation of hepatic microsomal cytochrome P-450, changes of formation of ring-and N-hydroxylation of 2-acetylaminofluorene (Hereafter referred to as 2-AAF) in the liver, and changes of activation of vitamin D in the liver. The results of my study are as shown below : 1. The contents of hepatic microsomal cytochrome P-450 of newborn rats was found to be remarkably increased in the group exposed to UV light for 3 weeks (P<0.001), but, such changes were much reduced in the group exposed to UV light for 3 weeks after being applied by sunscreen (P<0.05, P<0.001). 2. Cytochrome P-450 induced by UV light was found significantly increase N-hydroxylation which is a process for inactivation of 2-AAf known as carcinogenic source for the liver, and then, to expedite normal metabolism (P<0.05, P<0.001). In the group exposed to UV light for 3 weeks after being applied by sunscreen, both of N-hydroxylation which is process for activation of 2-AAF and right-hydroxylation which is a process for inactivation of 2-AAf were significantly reduced (P<0.05, P<0.001). The activation of vitamin D in the liver of newborn rats was found to be gradually increased when it is exposed to UV light for 1 week or 3 weeks (P<0.001), and in the Group exposed to UV light after being applied by sunscreen, such changes were reduced remarkably (P<0.001).
