The regulating role of syntenin-1 in colon cancer cell migration

Abstract

Syntenin-1 is a PDZ domain-containing adapter protein which is involved in organization of protein complexes in the plasma membranes, regulation of intracellular trafficking and cell-surface targeting, cytoskeletal regulation, and cell adhesion. Recently, the role of syntenin in cancer metastasis is studied. For example, overexpression of syntenin-1 increased cancer cell migration, invasion, and pseudopodia formation. Moreover, syntenin-1 binds to syndecan-2, which serves highly metastatic potential in colon cancer cells, and helps the syndecan-2 mediated prometastatic downstream signaling. In this report, I overexpressed syntenin-1 in various colon cancer cells, and observed the cell migration was visibly increased. In this condition, both of FAK phosphorylation and Rac1 activation were increased in colon cancer cell lines. These changes were more activated when syntenin-1 was co-transfected with syndecan-2. In conclusion, syntenin-1 seems to help the colon cancer cell migration and metastasis as a scaffold protein of syndecan-2.;Syntenin-1은 세포막 주변의 단백질 복합체들을 조직화하는 역할을 하는 adapter 단백질로서, 세포 내부적인 단백질의 이동을 조절하고, 세포 표면으로 단백질들을 응집시키기도 하며, 세포 골격의 형성 및 세포 부착에도 중요한 역할을 하는 것으로 알려져 있다. 최근 들어, 암 전이에 있어서 syntenin-1의 역할이 연구되기 시작하였다. syntenin-1의 과발현은 암세포의 이동성과 침투성, 위족 형성을 촉진시킨다는 사실이 알려졌고, syntenin-1이 syndecan-2와 결합하여 세포내부로의 신호전달을 촉진함으로써 대장암 세포의 전이성을 높여준다는 것 또한 밝혀졌다. 이 논문에서, syntenin-1이 대장암 세포에서 과발현된 경우 세포의 이동성이 눈에 띄게 증가하였으며, FAK의 인산화 또한 증가하였다. 또한, syntenin-1이 과발현된 경우 small GTPase로서 actin filament의 형성을 조절하는 Rac1의 활성화가 증가된 것을 대장암 세포주에서 확인하였다. Syndecan-2와 syntenin-1이 함께 과발현된 경우, 이러한 변화들은 더욱 활성화되었다. 결론적으로, syntenin-1은 syndecan-2의 scaffold protein으로서 대장암 세포의 이동과 전이를 촉진하는 것으로 생각된다.
