TGFβ1 gene delivery to adipose-derived stem cells cultivated in collagen scaffolds for effective chondrogenesis

Abstract

연골은 혈관, 신경, 임파관이 없기 때문에 손상을 받은 후에 연골이 다시 정상적인 기능과 구조를 가진 조직으로 재생되는 데에는 많은 한계를 지니고 있다. 결손부위를 치료하기 위해서 외부로부터 세포를 주입하는 방법이 있는데 이를 더 효과적으로 하기 위해 콜라겐 페이스트를 이용해 삼차원 지지체를 제조하고 전환성장인자 (TGFβ1) 유전자 전달을 통한 지방 유래 줄기 세포의 연골 세포로의 분화 연구를 진행하였다. 지방 유래 줄기 세포의 다분화성에 의해 다양한 세포가 함께 생성되는 것을 억제하고 보다 균일한 연골 세포를 공급하기 위해 특이적으로 전환성장인자 유전자 도입을 통해 분화시키는 방법을 시도하였다. 지방 유래 줄기세포는 많은 양을 용이하게 얻을 수 있으며 지지체로 사용된 콜라겐 페이스트는 3D구조로 주입 가능하며 연골 조직 재생에 유용함을 확인하였다. 지방 유래 줄기 세포의 연골 세포로의 분화 정도의 차이와 증식 속도를 알아보는 실험을 통해 그 우수성을 입증하였다. 또한 유전자 전달 콜라겐 페이스트가 연골 손상 부위에 이식이 편리하고, 천연 소재를 사용함으로써 이후 관리가 유용한 이점 또한 겸비함으로써 연골 재생에 유용함을 확인하였다.;Articular cartilage, as an avascular tissue, has significant limitations with respect to healing and repair due to lack of blood supply, nutrients and available factors for regeneration. Recently, the combined techniques of gene transfer and tissue engineering have gained considerable attention for the practical application of articular cartilage repair. Transforming growth factor β1 (TGFβ1) is multifunctional molecule that can play a main role in promotion of cartilage repair, and inhibition of inflammatory and alloreactive immune response. In that respect, TGFβ1 transfected Adipose-derived cells (ASCs) suspended in collagen paste (collagen-TGFβ1-ASC) and normal ASCs suspended in collage paste (collagen-ASC) were prepared to culture in chondrogenic media or control growth media. TGFβ1 release from transfected ASCs showed extended gene expression and provided superior biological effect on the differentiation of ASCs into chondrocytes compared to other groups.
