Abstract
INO80 는 ATP 를 가수분해 할 때 나오는 에너지를 이용해서 nucleosome 구조를 바꿀 수 있는 swi/snf based chromatin remodeling complex 의 family 중 하나이다. INO80 은 최근 효모에서 밝혀진 것으로 여러 개의 단백질로 이루어진 complex 를 이루고 있다. 효모에서 밝혀진 INO80 은 다른 ATP dependent chromatin remodeling complex 와 마찬가지로 전사 조절에 관계될 뿐 아니라, DNA 이중 나선 절단을 복구하는데 있어서도 직접적으로 관련 있고 최근에선 DNA의 복제에도 직접적으로 관계한다는 것이 밝혀졌다. 포유류 genome 은 효모 INO80 유전자의 homolog (mammalian INO80) 를 가지고 있지만 mammalian INO80 의 기능에 대해서는 알려진 보고가 없다. 본 논문에서는 인간의 INO80 complex가 세포 주기 진행에 있어서 크로마틴에 직접 결합을 함으로써 기능을 한다는 것을 밝혔다. 먼저 human INO80이 세포질에 주로 존재해 있다가 G1기에서 S기로 진행되면서 핵 안으로 들어가 결합을 한다는 것을 보았다. 좀 더 자세히 세포 주기를 나눠 분석한 결과 S기 뿐만 아니라 late G2기에서 early M기로 들어갈 때에도 크로마틴에 결합하는 것을 보았다. 이를 통해 hINO80이 S기 뿐만 아니라 G2와 M기에도 다이나믹하게 크로마틴에 직접적으로 결합을 하여 세포 주기 진행에 있어 기능을 한다는 것을 알게 되었다. 또한 hINO80이 크로마틴에 직접 결합을 할 때 인산화가 일어난 다는 것을 보았다. 이를 통해 hINO80이 크로마틴에 직접 결합을 하여 기능을 할 때 인산화가 중요하다는 것을 밝히게 되었다. 이는 앞으로 hINO80의 기능 연구에 중요한 단서 역할을 할 것이라 여겨진다. 마지막으로 hINO80의 발현이 억제되었을 경우 M기 자체의 진행은 이상이 없으나 M기로의 진입이 지연되는 것을 밝혔다. 이를 통해 hINO80이 세포 주기 진행의 조절에 중요한 역할을 한다는 것을 밝혔으며 앞으로 좀 더 면밀한 연구를 통해 자세한 기능을 밝혀야 할 것이다.;ATP-dependent chromatin remodeling complexes are capable of utilizing the energy of ATP hydrolysis to alter the structure of nucleosomes in a way to facilitate the access of cellular proteins to their target DNA. Among these complexes is INO80 most recently identified from yeast and its homologue exists in higher eukaryotes including human cells. Although yeast INO80 (yINO80) has been well established to play important role in DNA repair as well as in transcription, the cellular function of mammalian INO80 has remained largely unexplored. Our laboratory previously found that human INO80 (hINO80) deficient cells exhibit defects in DNA replication and S phase progression as well as genome instability. In an effort to understand the underlying mechanisms, I investigated the subcellular localization and chromatin association of hINO80 during the cell cycle progression. First, using biochemical fractionation experiments, I found that hINO80 exists predominantly within the cytoplasm at G1 phase, and is translocated into the nucleus as cells enter S phase. I then found that hINO80 dynamically associates with and dissociates from chromatin during progression from S to G2 to mitotic phases. The chromatin association of hINO80 increases as cells progress from G1/S border to mid S phase, in which hINO80 and PCNA are colocalized at replication forks. hINO80 then dissociates from chromatin after mid S phase and re-associates with chromatin when cells progress to late G2 and prophase. Interestingly, my data suggest that hINO80 is modified by phosphorylation when it enters the nucleus and associates with chromatin. Finally, I found that siRNA knockdown of hINO80 leads to a defect in chromosome condensation and delay of mitotic entry, consistent with the fact that hINO80 re-associates with chromatin at late G2 and prophase. These results suggest that hINO80 exerts its function via dynamic interaction with chromatin and that the modification of hINO80 by phosphorylation is potentially important for its function.
