Abstract
Focal adhesion kinase (FAK) 는 non-receptor cytoplasmic tyrosine kinase 로서 세포 골격 구성의 조절, migration, proliferation, survival 을 포함하는 여러가지 기능에 관여한다. FAK 의 발현 증가와 tyrosine 인산화의 증가는 암 발생 과정에서 중요한 역할을 하지만, FAK 이 암유전자로 인한 변형을 증가시키는 메커니즘은 아직까지 잘 알려져 있지 않다. 이에 본 논문에서는 암유전자로 인한 변형에서의 FAK 기능을 살펴보았고 FAK 의 861번 인산화가 ras 로 인한 섬유 세포의 변형에 관련이 있고, 이런 FAK 의 861번 인산화가 어떤 메커니즘을 조절하는지를 규명하였다. 먼저, histone deacetylase (HDAC) 억제제인 trichostatin A (TSA) 를 이용하여 변형된 섬유 세포를 정상화로 되돌렸을 때 FAK 의 역할을 조사하였다. 두 개의 다른 ras 암유전자로 인해 변형된 두 종류의 섬유 세포주에 TSA 로 HDAC 의 활성을 억제하였더니 세포의 모양이 평평해졌고 더 잘 펴진 모양으로 유도되었다. 이는 다시 정상화됨을 의미한다. 이러한 모양 변화는 fibronectin 위에서 변형된 NIH3T3 세포주의 펴지는 능력을 증가시키기도 하였다. FAK 에는 6개의 tyrosine 자리가 있는데, 이중에서 861번의 tyrosine 의 인산화가 TSA 에 인해 정상화로 되는 동안, 가장 확실히 감소하였다. FAK 과 상호작용하는 Src kinase 의 양과 활성화, 모두 감소 하였는데, 이는 Src kinase 가 FAK 861번의 인산화를 조절한다는 것을 의미한다. 이 연구로 FAK 861번의 증가된 인산화는 ras 로 인한 섬유 세포의 변형에 관련되어 있고, TSA 가 Src family kinase 를 통해FAK 861번 인산화를 조절함으로써 섬유 세포를 다시 정상화 시킬 수 있음을 제시하였다. 다음으로, tetracycline 으로 H-ras 암유전자를 조절할 수 있는 vector 를 도입한 NIH3T3 세포를 이용하여, 암유전자로 변형되는 과정에서 FAK 861번 인산화의 역할을 조사하였다. FAK 861번 인산화는 두 개의 다른 ras 암유전자로 변형시킨 두 종류의 섬유 세포와 H-ras 암유전자로 변형을 유도시킨 NIH3T3 세포에서 증가하였다. 그러나, 861번을 인산화가 될 수 없게 만든 돌연변이 FAK (Y861F) 를 도입시킨 세포는 정상 FAK 와 kinase 부분을 돌연변이 시킨 FAK 를 도입시킨 세포에 비해서, migration/invasion, focus-forming activity, anchorage-independent growth 가 감소되었다. FAK 과 Src kinase 의 활성이 변화되지 않는 것에 비해, FAK 과 p130CAS 와의 상호작용은 FAK Y861F 를 도입시킨 세포에서 감소 하였고, FAK 861번 인산화는 in vitro 에서 이러한 상호작용을 증가 시켰다. 또한, FAK Y861F 를 도입시킨 세포에서는 c-Jun NH2-terminal kinase 의 활성과 matrix metalloproteinase-9 의 발현이 암유전자로 변형을 유도시키는 동안에 감소 하였다. 이러한 결과들을 토대로, FAK 861번 인산화가 FAK 와 p130CAS 의 상호작용을 조절함으로써 H-ras 암유전자로 유도시킨 변형에서 결정적인 역할을 한다는 것을 제시하였다. 결론적으로, FAK 861번 인산화는 암유전자로 인한 변형에 중요한 역할을하고 FAK/p130CAS 신호전달을 조절한다는 것을 제시한다. ;Focal adhesion kinase (FAK) is a non-receptor cytoplasmic tyrosine kinase that modulates multiple cell functions, including the regulation of cytoskeletal organization, migration, proliferation, and survival. Although elevated expression and increased tyrosine phosphorylation of FAK are crucial for tumor progression, the mechanism by which FAK promotes oncogenic transformation is unclear. In this thesis, I have examined the role of FAK in transformation and found that phosphorylation of FAK at tyrosine 861 correlates with ras induced transformation of fibroblasts, and the regulatory mechanism of FAK phosphorylation at tyrosine 861. In the first part, I investigated the role of FAK in detransformation using the histone deacetylase (HDAC) inhibitor, trichostatin A (TSA). Inhibition of HDAC activity in two different ras-transformed fibroblast lines by TSA induced a morphological change into a flattened and more spread morphology, implying detransformation. These morphological changes included increased spreading ability of transformed NIH 3T3 cells on fibronectin. Of the 6 tyrosine phosphorylation sites in FAK, phosphorylation at tyrosine 861 was clearly decreased during detransformation by TSA. It resulted from both decreased activity of Src family tyrosine kinase and/or decreased amount of Src kinase interacting with FAK, implying that Src kinase regulates phosphorylation of FAK at tyrosine 861. All these findings suggest that increased phosphorylation of FAK at tyrosine 861 correlates with ras induced transformation of fibroblasts, and TSA is able to detransform them through regulation of FAK phosphorylation at tyrosine 861 by a Src family kinase. In the second part, I have determined the role of FAK phosphorylation at tyrosine 861 in transformational progression using NIH 3T3 cells transfected with a tetracycline-inducible H-Ras expression vector. FAK phosphorylation at tyrosine 861 was increased in both constitutively H-Ras-transformed and H-Ras-inducible NIH3T3 cells, in parallel with cell transformation. However, H-Ras-inducible cells transfected with the nonphosphorylatable mutant FAK Y861F showed decreased migration/invasion, focus-forming activity and anchorage-independent growth, compared with either wild-type or kinase-defective FAK. In contrast to unaltered FAK/Src activity, the association of FAK and p130CAS was decreased in FAK Y861F transfected cells and FAK phosphorylation at tyrosine 861 enhanced this association in vitro. Consistently, FAK Y861F transfected cells were defective in activation of c-Jun NH2-terminal kinase and in expression of matrix metalloproteinase-9 during transformation. Taken together, these results strongly suggest that FAK phosphorylation at Tyr-861 is crucial for H-Ras-induced transformation through regulation of the association of FAK with p130CAS. These findings indicate that FAK phosphorylation at tyrosine 861 plays a major role in transformation and regulates FAK/p130CAS pathway.
