Abstract
식물상 복원공법을 이용하여 TNT로 오염된 지역을 정화하기 위해 TNT 제거능을 가진 내성종을 선정하였다. 8종의 대표적인 야초류를 이용하여 TNT 농도 변화에 따른 발아율, 유식물의 뿌리와 지상부의 생장을, 생체량 및 수분 흡수 능력 등의 TNT에 대한 독성 임계점(PT)을 측정하였다. 그 결과 TNT가 식물에게 미치는 독성효과는 오염물의 농도와 식물종에 따라 다르게 나타났다. 60 ~ 80 mgTNT/L 농도는 어저귀, 식용피, 자귀풀 종자의 발아을에 영향을 미치지 못하였다 뿌리와 지상부의 길이, 생체량은 TNT농도가 증가함에 따라 감소하였으며 수분 흡수량도 감소하였다. 발아 후 14일 동안 어저귀, 식용피, 자귀풀의 뿌리 생장에 있어 식물 독성 임계점(50%) 농도는 5 ~ 40 mgTNT/L 이며 지상부 생장 독성 임계농도는 50 ~ 73 mgTNT/L, 생체량 독성 임계농도는 68 ~ 99 mgTNT/L로 뿌리 길이 > 지상부 길이 > 생체량 > 발아율 순으로 TNT에 애해 민감하게 반응하였다. 실험 결과, 8종의 식물 중에서 어저귀와 자귀풀은 TNT에 대해 내성을 지닌 종으로 판명되었다. 식물체에 의한 TNT의 흡수율 및 전환 산물을 연구하기 위해 토착식물인 어처귀와 자귀풀을 20 ~ 80 mgTNT/L의 여러 가지 농도에서 수경 재배하였다. 액체 배지 내에서의 TNT는 유사 1차 반응으로 감소하였다. 80 mgTNT/L에 노출된 어저귀를 제외할 경우, TNT의 제거 기작은 Michaelis-Menten 식으로 표현 가능하였다. 두 식물 종은 배지에서 TNT 농도가 높을수록 TNT 제거 속도 상수가 감소하는 경향을 보였으나, 식물체의 생체 중량에 대하여 일반화할 경우, 오염물 제거는 60 mgTNT/L에 노출한 배지에서 가장 많았다. TNT 환원물질은 모든 수경재배 배지에서 발견되지 않았으나, 노출 7일 후에 수거한 식물의 추출액에서는 몇 종의 TNT 전환 산물이 검출되었다. 어저귀와 자귀풀의 주된 TNT 전환물은 2ADNT과 4ADNT이었으며 자귀풀의 경우 2,6DANT과 2,4DANT가 미량 검출되어 자귀풀이 어저귀보다 TNT를 다양하게 환원시킬 수 있는 종으로 판별되었다. 이상의 결과를 통해 어저귀와 자귀풀은 지금까지 보고된 식물보다 더욱 우수한 TNT 흡수 및 변환 능력이 있음을 확인하였다. 군사지역은 TNT와 같은 화약물질이 중금속과 복합으로 오염되어 있다. 따라서 TNT 제거를 위한 식물상 복원공법을 적용할 때 다른 오염물질이 식물체의 TNT 제거, 변환, 축적 능력에 영향을 줄 것으로 예상된다. 토착 야초류인 어저귀(Abutilion avicennae)를 TNT와 Cd으로 복합 오염된 배지에서 무근 수경재배하여 중금속이 TNT 제거 및 변환에 미치는 영향을 조사한 결과 20 mgTNT/L와 1.3 mgCd/L는 잎의 탄력 감소, 말림 현상(Leaf roll), 백화 현상(Chlorosis), 낙엽형성, 생체량 감소 등과 같은 식물독상효과를 나타내었고, TNT와 Cd에 복합으로 존재할 때 식물체이 미치는 독성효과는 상승되었다. 어저귀는 카드뮴 차단종으로 배지 내 카드뮴이 1.3 mg/L 존재하더라도 어저귀에 의한 TNT 제거, 변환, 이동, 분배에 영향을 미치지 않았다. 카드뮴은 식물 반응조 안에서의 TNT amination 위치에 영향을 미치지 못했다. 따라서 TNT를 제거하기 위해 어저귀를 식생할 경우 토양 용액 중 카드뮴이 어저귀에 의한 TNT 제거 및 전환에 영향을 미치지 않을 것으로 사료된다.;To select a suitable plant for the phytoremediation of TNT contaminated soil, TNT toxicity threshold of eight grasses were identified. It was determined based on various factors Including cumlulative seed germination, root and shoot length, fresh biomass, and the amount of water uptake under various TNT concentrations. Phytotoxic effect of TNT was enlarged as TNT concentration increasing but the toxic intensity was varied between grass species. Concentrations up to 60 ~ 80 mgTNT/L did not affect germination of Abutilion avicennae, Echinochioa crusgallivar , and frumentacea, and Aeschynomene indica. Length of root and shoot and fresh biomass decreased as TNT concentration increased. Phytotoxicity threshold(PT_50) of A. avicennae, E. crusgalli var. frumentacea, and A. indica were 5 ~ 40 mgTNT/L for root length, 50 ~ 73 mgTNT/L for shoot length and 68 ~ 99 mgTNT/L for fresh biomass after 14days of seedling exposure. PT_50 showed that sensitivity of TNT to plant was in the order of root length > shoot length > fresh biomass > germination rate. In addition, the amount of water uptake decreased with increasing TNT concentration in A. avicennae and A. indica. From these results, we concluded that A. avicennae and A. indica had tolerance to TNT anions tile species tested. The abilities of plants, A. avicennae and A. indica, on the uptake and transformation of TNT were quantified at various initial TNT concentrations ranging from 20 to 80 mgTNT/L using a hydroponic culture. The TNT concentration decreased following the pseudo-first order kinetic in the liquid media. In all plant batches, the higher TNT concentrations resulted in lower removal rate constants. However, TNT removal mass was the highest in the 60 mgTNT/L culture media. Reductive metabolites of TNT were not detected in hydroponic culture media, but transformation products were detected in the extracts of root, stem and leaves of the two plant species. Major TNT metabolites were 2-amino-4,6-dinitrotoluene and 4-amino-2,6-dinitrotoluene for the 2 species, however, 2,6-diamino-4-nitrotoluene and 2,4-diamino-6-nitrotoluene were detected in the extracts of A. indica, suggesting that both of plants could metabolize TNT and A. indica much further. Uptake and transformation of TNT by plants was described by the Michaelis-Menten equation, except for the 80 mgTNT/L in A. avicennae, which showed inhibition kinetics. From these results, we concluded that A. avicennae and A. indica had a better intrinsic capacity for uptake and transformation of TNT than other plants reported until now. Most of army depots contaminated with co-contaminants, TNT and heavy metals. In phytoremediation for the TNT, heavy metals may be inhibit mineralization, transformation and sequestration of TNT by the plant. We studied that effect of cadmium on TNT degradation and transformation by Abutilion avicennae in axenic hydroponic culture. When plant was cultured in 20 mgTNT/L and 1.3 mgCd/A, plant displayed phytotoxicities, reduction of leaf fresh, leaf roll, chlorosis, leaf loss and fresh weight loss. Phytotoxicity was more synergistic in the combined contaminants than single contaminant. It was not matter that TNT transformation, translocation and distribution by A. avicennae, although 1.3 mgCd/L existed in the TNT medium. Cadmium didn't affected that position, para-, of reductive transformation in TNT. For reason that A. avicennae did not uptake to cadmium. Therefore the soil solution containing 1.3 mgCd/L will not affected to TNT degradation by Abutilion avicennae in Army depots.
