Abstract
Histamine Releasing Factor (HRF) is divided into two types, IgE-dependent HRF and IgE-independent HRF. IgE-HRf, soluble mediators that can release histamine from basophils and mast cells is an identical molecule with p21, p23, TCTP (translationally controlled tumor protein). In our laboratory, we are studying on IgE-dependent HBF that is the major factor to activate basophils in patients with chronic inflammation and allergic bronchial asthma. It was reported that HRF was significantly reduced in brain of Alzheimer's Disease (AD) and Down Syndrome (DS) as compared to normal person. This decreased level of HRF in the brain of AD and DS suggests that HRF might be involved in cognitive function. The aim of this study was to investigate the role of mitogen-activated protein kinase (MAPK) in IgE-HRF-induced dopamine release using neuronal PC12 cell line. I found that JNK, but not ERK and P38, was activated by HPLC-purified HRF although it was not activated by the endotoxin and the vector protein itself. However, HPLC-purified HRF failed to have biological activities, such as histamine-releasing and neurotransmitter-releasing activity, suggesting that functional HBF might need to be modified. To test this possibility, I investigated the interaction between HRF and peroxiredoxin I since it was found as a HRF - interacting protein by yeast-two hybrid screening with human brain cDNA library. I performed pull-down assay to see the interaction and the data suggest that Prx I interacts with HRF. Therefore, it is possible that HRF protein should be modified to be functional, presumably through the process of oxidation and reduction.;Histamine Releasing Factor (HRF)는 IgE가 있을 때 histamine 유리를 촉진시키는 IgE-dependent HRF와 IgE와는 무관하게 histamine 유리를 촉진시키는 IgE-independent HRF로 나눌 수 있다. IgE-dependent HRF는 기존에 알려진 P2l, P23, TCTP (translationally controlled Tumor protein)와 동일한 단백질로 mast cell 및 basophil에서 histamine을 유리 시키는 인자이다. 본 실험실에서 주로 연구하는 단백질은 IgE-dependent HRF로 만성적 염증 반응이나 알러지성 천식환자에게서 basophil을 활성화시키는 주요 인자로 작용한다. 또한 다운증후군이나 알쯔하이머 질환을 가진 환자의 뇌에서 HRF의 농도가 정상인에 비해 낮게 나타난다는 점을 볼 때 HRF가 인식, 지각 능력의 저하를 설명할 수 있는 가능성을 제기하기도 한다. 본 논문에서는 neuronal cell인 PCl2 cell에 IgE-dependent HRF를 처리했을 때 dopamine이 분비된다는 사실을 바탕으로 MAPK중 어떤 signal이 관여하는 지를 알아보고, in vitro상에서 histamine을 분비하는 활성을 가진 HRF 단백질을 분리, 정제하고자 하였다. HPLC를 통과해 gradient별로 분리된 fraction 각각을 실험에 사용해 보았고 단백질을 재조합하는 과정에서 혼입된 endotoxin이 실험에 영향을 미치는지를 살펴보았다. 또한 단백질의 활성이 HRF cDNA가 들어있는 백터의 영향을 받는지를 알아보기 위해 pET32a(+), pRSET, pGEX-4T-3에 들어있는 각각의 HRF를 정제해 실험에 사용해 보았다. 그리고 본 실험실에서 pEG202/rat recombinant HRF (rRHBF)를 bait로 human brain cDNA library로부터 yeast two hybrid assay를 실시한 결과 peroxiredoxinⅠ과 rRHRF가 결합할 수 있다는 사실을 알게 되었다. 이 사실을 바탕으로 HRF 단백질의 활성이 산화, 환원의 정도와 상관성이 있는지를 알아보기 위해 항 산화제인 peroxiredoxinⅠ과의 pull down assay 결과 HRF와 PrxⅠ이 결합함을 확인하였다.
