Abstract
암세포의 항암제에 대한 약제 내성에는 drug efflux 혹은 radical scavenger에 의한 세포내 radical intermediates의 효율적 제거 등이 관여하는 것으로 알려져 있다. 본 연구에서는 항암제인 cisplatin에 대한 내성 형성 요인을 알아보기 위하여, small cell lung carcinoma인 NIH H69과 cisplatin 내성 세포주인 H69/CDDP에서 radical scavenger로 알려진 catalase, GST-L, GST_(π) 등의 발현 정도와 GST_(π)의 기질로서 작용하는 GSH의 세포내 수준 및 SOD 활성도를 비교 조사하였다. Catalase와 GST-L의 경우 모세포주와 cisplatin 내성 세포주에서 차이가 없었으나, GST_(π)의 경우 내성 세포주에서 현저한 증가를 보였다. SOD 활성도는 내성 세포주에서 1.7배가 높은 것으로 나타났다. H69/CDDP에서의 GSH의 세포내 수준은 모세포주에서와 동일하였으며, BSO 처리 후 GSH의 농도는 두 세포주에서 모두 급격하게 감소하였다. 이 밖에 두 종류의 non-small cell adenocarcinoma인 PC9과 PCl4의 모세포주와 cisplatin 내성 세포주인 PC9/CDDP, PC14/CDDP에서도 GST-L, GST_(π)의 발현 정도와 SOD 활성도를 측정하였다. PC9/CDDP와 PC14/CDDP에서 GST-L와 GST_(π)의 발현 정도는 모세포주에서와 동일하게 관찰되었다. 그러나 이 두 cisplatin 내성 세포주의 경우 SOD 활성도가 모세포주에 비해 각각 11.4배, 10.5배가 증가되었다. Apoptosis의 저해자인 Bcl-2의 세포내 수준도 NIH H69, PC9, PCl4, H69/CDDP, PC9/CDDP, PC14/CDDP에서 조사하였다. PC9/CDDP와 PC14/CDDP에서는 이들의 모세포주와 비교했을 때 Bcl-2 발현의 차이를 보이지 않았으나, H69/CDDP에서는 모세포주에 비해 현저히 증가된 것을 관찰할 수 있었다. 결과적으로 H69/CDDP에서의 cisplatin 내성 형성 요인으로는 GST_(π)와 Bcl-2의 과잉 발현이, PC9/CDDP와 PC14/CDDP의 cisplatin 내성 형성 요인으로는 SOD 활성도 증가가 중요한 요인의 하나로 제시되었다.;It has been known that drug efflux or depletion of cellular radical intermediates by radical scavengers are involved in the development of drug resistance in cancer cells. In these studies, to investigate which factors are responsible for drug resistance, we examined the cellular level or activity of radical scavengers; catalase, glutathione-S-transferase-L (GST-L), g1utathione-S- transferase-π (GST_(n)) and superoxide dismutase (SOD), and the level of glutathione (GSH), the substrate of GST, in the cisplatin-resistant subline of small cell lung carcinoma NIH H69 (H69/CDDP) and its parental cell line (NIH H69). There was no change in the cellular levels of catalase and GST-L in both cell lines. However, the SOD activity in H69/CDDP was 1.7 fold higher than that in NIH H69 and GST_(π) was overexpressed in H69/CDDP. The level of GSH in H69/CDDP was similar to that of GSH in NIH H69. Buthionine sulfoximine (BSO) treatment almost completely reduced GSH level in both cell lines. In addition to these two cell lines, we examined the levels of GST-L, GST_(π) and the activity of SOD in PC9 and PC14, non- small cell lung adenocarcinoma, and their cisplatin-resistant sublines, PCS/CDDP and PC14/CDDP. The expression levels of GST-L and GST_(π) were similar in parents and cisplatin-resistant sunlines. But the SOD activities in PC9/CDDP and PC14/CDDP were 11.4 and 10.5 fold higher than that of its parental cell lines, respectively. We also examined the level of Bcl-2, the inhibitor of apoptosis, in NIH H69, PC9, PC14, H69/CDDP, PC9/CDDP and PCl4/CDDP. In PC9, PC14 and their cisplatin-resistant sublines, Bcl-2 were similary expressed. On the other hand, Bcl-2 was overexpressed in H69/CDDP compared to NIH H69. The results in this study strongly suggest that overexpression of GST_(π), and Hcl-2 in H69/CDDP and the increased SOD activity in PC9/CDDP and PC14/CDDP may contribute to the development of cisplatin-resistance in these lung cancer cell lines.
