- 고온성 방선균을 분리하여 동정하고 그 균주에 의해 생성되는 당화형 α-amylase를 정제하여 그 특성을 조사하고 분자량을 측정하였다.
고온성 방선균의 분리는 서울, 정기 일원의 퇴비를 채취해서 소량을 멸균 생리 식염수에 현탁하고 중성 배지(pH 7.0)에 접종하여 55℃에서 48시간 배양하였다. 분리된 균주들은 액체 배지(pH 7.0)에 접종하여 50℃에서 3일간 배양한 후 amylase 활성을 조사하여 활성이 높은 균주를 선별하였다.
이 균주의 형태적 배양학적 생리적 특징을 조사한 결과 Thermomonospora속으로 추정하였다.
Amylase 생성은 중성 액체 배지에서 48℃, 60시간 진탕 배양하였을 때, 가장 많았다.
이 amylase를 ammonium sulfate 침전, DEAE-cellulose column chromatography, Sephadex G-100 gel filtration을 통해 정제하였고 회수율은 19.4%였다.또한 SDS-polyacrylamide 전기영동을 하여 분자랑을 측정한 결과는 51,000으로 나타났다.
정제 효소에 의한 전분과 amylose azure의 가수분해 산물을 Thin-layer chromatography로 분석한 것과 Iodine-staining power 측정을 종합하여 본 결과 균주7-1이 생성하는 amylase는 당화형 α-amylase임을 확인할 수 있었다.
이 당화형 α-amylase의 전분에 대한 ㎞치는 6.25㎎/㎖이었고 최적 pH는 6이었으며 pH 5~9사이에서 안정하였고, 최적 온도는 50℃였으며 55℃에서 1시간 안정하였고 CaCl_(2) 첨가시에서 60℃까지 안정하였다. 이 효소는 또한 Li^(+), Ca^(2+), Mg^(2+), Ba^(2+)에 의해서는 효소 활성이 촉진되었고 Hg^(2+), Pb^(2+), Cu^(2+), Ag^(+) 의해서는 현저히 억제되었으며, EDTA를 제외한 다양한 범위의 detergent에 대해서 안정하였다.;The saccharifying α-amylase produced by Themzomonospora sp . (No. 7-1) isolated from composts was purified, and its enzymatic properties were investigated.
Incubation in neutral media with shaking at 48℃ for 60 hours gave optimal cultural condition for the production of amylase.
The amylase was purified by ammonium sulfate precipitation. DEAE-cellulose column chromatography and Sephadex G-100 gel filtration serially. The enzyme was purified to homogeneity and its yield was 19.4%. The molecular weight was estimated by SDS polyacryiamide gel electrophoresis to be 51,000.
The purified enzyme was confirmed to be saccharifying α-amylase by thin-layer chromatography and iodine staining power measurement with hydrolytic product of starch and amylase azure. Characteristics of purified α-amylase were as follows ;
Km value was 6.25 mg/ml
Optimal pH and temperature were 6.0 and 50℃.
The enzyme was stable in the range of pH 5.0 to 9.0.
It was stable for 1 hour at 55℃, with CaCl_(2) up to 60℃.
When Li^(+), Ca^(2+), Mg^(2+), Ba^(2+) were treated, the enzymatic activity was increased, while with Hg^(2+), Pb^(2+), Cu^(2+), Ag^(+) it was decreased.
