당뇨병 환자의 혈청 Lipoprotein에 관한 연구

Abstract

혈청내 lipoprotein은 중성지방, 인지질, 콜레스테롤을 운반하는 화합물이다. 그러므로 lipoprotein내 이들 성분의 변화는 생체내 각종대사와 밀접한 관계가 있다는 것은 잘 알려진 사실이다. 저자는 당뇨병환자의 혈청내 lipoprotein변화의 기전을 살피고자 혈청 lipoprotein을 초원심침전법을 이용하여 VLDL, LDL 및 HDL로 분리하고 이들 lipoprotein의 성분인 단백질, 중성지방, 인지질 및 콜레스테롤 함량을 측정하여 정상인과 비교하고 혈청 lipoprotein의 지방성분조성변화를 살펴보았으며 한편 그 지방성분의 조성을 더욱 자세히 살피고자 SDS- polyacrylamide Gel (7.5%) 전기영동법을 이용하여 VLDL, LDL및 HDI의 electrophoretic pattern을 측정 관찰하여 다음과 같은 결과를 얻었다. 1.당뇨병환자의 혈청단백 및 혈청 각 lipoprotein fraction에서의 단백질 함량은 대조군에 비해 변화가 없었다. 2. 당뇨병환자의 혈청중에 총 콜fp스테롤 함량은 대조군에 비하여 현저히 증가하였고. 각 lipoprotein fraction 특히 LDL이 현저히 증가하였고 HDL은 약간 증가하는 경향이었다. 3. 당뇨병환자의 혈청중에 총 phospholipid 함량은 증가하였고 각 lipoprotein fraction인 VLDL과 HDL에서는 감소하였고 LDL에서는 현저히 증가하였다. 4. 당뇨병환자의 혈청중 총 triglyceride 함량은 대조군에 비하여 증가되었으며, lipoprotein fractien인 VLDL에서는 감소하고. LDL은 현저히 증가하였으며 HDL은 약간 증가하였다. 5. SDS-Polyacylamide Gel (7.5%) electrophoresis한 결과 대조군과는 다른 electrophoretic pattern을 나타내었다.;The protein, triglyceride and cholesterol concentations were measured and lipoprotein were analyzed by SDS-Polyacrylamide GeL Electrophoresis in whole serum and the 3 major lipoprotein classes of the diabetic patients and control. The results are as follows: 1. The protein concentrations of whole serum and3 major lipoprotein classes were not significantly different in normal and diabetes' sera. 2. The cholesterol concentrations were higher in diabetes' serum than normal. There was markedly increased cholesterol in the LDL of diabetess. 3. The phospholipid concentrations were higher in diabetes' serum, but lower in the VLDL & HDH of diabetes than normal. 4. The trighyceride concentrations were higher in diabetes' serum than normal, but Lower in the VLDL of diabetes. 5. The electrophoretic patterns of LDL & HDL by SDS-Polyacrylamide Gel Electrophoresis in the diabetes showed the abnormal pattern in comparison with normal.
