일산화탄소 급성중독에 의한 십자매 혈장단백질의 변화에 관한 연구

CO gas 濃度 800 ppm의 環境下에서 십자매를 中毒시켰던 바 2~7時間 정도의 심한 中毒狀態(中毒群)를 계속하다가 점차 回復되어 24時間이 지나면 완전히 回復(回復群)되며 이 回復群을 공기중에 내놓으면 완전히 原狀으로 回復(原狀回復群)되는 것을 觀察하였다. 그런데 계속되는 CO 環境下에서 中毒狀態가 계속되다가 다시 回復되는 現狀을 규명하기 위하여 십자매의 血液 成分을 分析하여 보았다. 그 結果 total hemoglobin의 量은 對照群에 比해 모든 實驗群에서 增加되었고 oxyhemoglobin과 carboxyhemoglobin의 量은 中毒群보다 回復群에서 다소 減少되었으며 methemoglobin의 量은 回復群에서만 다소 增加되었다. 한편 plasma protein을 SDS-polyacrylamide vertical slab gel electrophoresis를 한 結果 對照群에서는 prealbumin region에 band II만이 나타났으나 中毒群과 回復群에서는 prealbumin region에서 band I, II, III이 나타났으며 그 量은 回復群에서 가장 많았다. 그리고 plasma protein을 cellulose acetate electrophoresis한 結果 A/G ratio가 對照群에 比해 中毒群, 回復群의 順으로 減少되었으며 특히 α-globulin이 回復群에서 현저하게 增加되었다. 한편 CO 中毒症狀과 밀접한 관계가 있는 hepatic microsomal cytochrome P-450의 量은 모든 群에서 變化가 없었다. ; It was observed that a Society Finch recovered after the poisoned state during exposure to an atmosphere contaminated 800 ppm of CO. To examine closely this phenomenon, the variations of plasma protein were investigated. The Society Finch were divided into control group(normal birds), poisoned group, recovering group and completely-recovered group. The poisoned group was exposed for 3hrs at 800 ppm of CO and the recovering group was exposed for 24 hrs at the same CO concentration and completely-recovered group was exposed for 6 hrs in air after recovered at 800 ppm of Co. The contents of total hemoglobin in the experimental groups inreased more than in the control group. The contents of carboxyhemoglobin and oxyhemoglobin in the recovering group slightly decreased more than in the poisoned group but the content of methemoglobin in the recovering group more slightly increased than in the control group. In the control group only the prealbumin region band II appeared and in the poisoned group the prealbumin region band I, II, III to some extent appeared and to large extent in the recovering group on SDS-polyacrylamide vertical slab gel electrophoretic patterns of plasma protein. By means of cellulose acetate electrophoresis of plama protein, A/G ratios in the poisoned group and the recovering group decreased much more than in the control group and especially α-globulins in the recovering group increased in great quantity. The contents of hepatic microsomal cytochrome P-450 in the control group and the experimental groups were not changed.
