Abstract
포유동물의 수정란(zygote)을 채외 배양시 포배기(blastocyte)까지 발생하지 못하고 중도에서 반드시 발생이 정지되는 시기가 있어 이런 현상을 in vitro culture block이라 하며 포유 동물 종류에 따라 그 정지하는 시기가 어느 일정한 난할 시기에 정해져 있다(사람과 토끼는 제외). 생쥐의 경우 초기 2-세포기에 이 시기가 있어 이를 2-cell block이라 한다. 이런 block 현상이 생기는 것은, 생체에서 환경변화 및 생리적 변화가 심하게 일어나지만, 체외 배양 조건을 체회배양에서 100% 다 맞추어 주지 못하는데서 기인하며, 근본적인 원인은 핵내에 cdc유전자(cell division control유전자)의 비활성화에서 기인한다. 본 연구에서는 이런 유전자에 영향을 미치는 여러가지 화학 물질을 처리하여 유전자 활성을 유도함으로써, 사람의 IVF-ET 과정에 대한 대응 실험으로 많이 사용되고 있는 생쥐에서 수정란을 체외 배양시 반드시 일어나는 이러한 2-cell block현상의 원인을 규명하고자 하였다. ICR계의 생쥐 암컷(4-6주)의 복강에다 PMSG 5IU를 처리하고 48시간후 다시 HCG 5IU를 주사하여 과배란(superovulation)을 유도하며 바로 생식력이 있는 수컷과 합사시켜 mating시켰다. HCG 주사후 18-30시간 사이에서 난관을 세척하여 수정난 및 초기 2-세포기배를 얻었다. 배양액은 M-16을 기본 배양액으로 사용하였고 여기에서 인산, 포도당, 비타민, 철분, insulin, 아미노산 및 EDTA등을 첨가 또는 제거하여 24,72 및 96시간 배양후 저자는 이들 성분이 2-Cell block 극복에 어떤 영향이 있는지를 관찰하였다. 본 연구의 결론은 다음과 같다. 1. 배양액의 성분중 인산은 2-세포 발생에 억제적으로 작용하여 2-cell block의 원인이 되었다. 인산 성분을 제거했을 때 33%-38%의 극복(overcome)효과가 있었다. 2. 배양액 성분중 포도당은 햄스터의 경우와 달라 억제적인 작용을 하지 않으며 또한 촉진의 효과가 없었다. 3. 비타민중 panthotheic acid와 inositol은 2-cell block을 극복시키는 작용은 없으나 일단 극복된 배의 발생을 촉진시켜 배발생속도를 빠르게 하였다. 4. Fe++(Fe2(NO3) )성분은 Ca++-inhibitor로 작용하며 8-세포기에 일어나는 compaction과정을 방해하여 상실기 이상의 발생을 억제시켰다. 5. EDTA 성분 추가로 2-cell block이 67% 극복되었다. EDTA의 chelating 효과로 인한 Ca++의 기능을 조절함으로써 야기되는 것으로 추정되어 이에 대한 연구가 절실히 요망되고 있다. ; In most mammals, there is a block stage when fertilized ova(zygotes) are cultured and these zygotes can not develop to the blastocyst in vitro system except when cultured in ampulla portion of the oviduct. In mouse, this block stage is in a early 2-cell embryo. Its reason is that in vitro culture system can not mimic the in vivo system and can not satisfy all the conditions of dynamic environment and all the physiological conditions of embryos development of the in vivo system. Most basic reason of this 2-cell block is the inactivation of cdc genes of the nuclei, which is caused by unknown factor(s) of the in vitro system. In the present study, it is aimed to investigate the causing factor of mouse 2-cell block by the addition or removal of the components of the culture medium.. Female mice of ICR strain(4-6 weeks old) were used. Superovulation was induced with PMSG 5IU and HCG 5IU at 48-hour interval. Then, immediately the female mice were mated with fertile males. Zygotes or early 2-cell embryos were collected by flushing the oviducts 18-30 hours after HCG injection. M16 was the basic medium used in the present study from which phosphate or glucose was omitted and vitamins(inositol, panthotheic acid), Fe+++, insulin, amino acids(glutamine, methionine, phenylalanine and proline) and EDTA were added depending on the experimental groups. In these different medium components, zygotes or 2-cell embryos were cultured for 24, 72 and 96 hours. Results as followings were obtained from this study. 1. Phosphate of the medium was strongly inhibitory to 2-cell embryo development, and 33-38% of 2-cell block were overcome by emission of phosphate from the culture medium. 2. Glucose seems to have neutral effect differently from those of hamster. It did not show any inhibition or stimulation of 2-cell embryo development. 3. Panthotheic acid and inosital show some stimulatory effect of development of 2- cell embryos which were already overcome by the emission of phosphate from the culture mediun. 4. Fe+++ seems to be inhibitory to the compaction of 8-cell embryos by replacing Ca++ of the culture medium. In the presence of Fe+++, all the embryos could not develop to 8-cell stage. 5. Overcoming effect of EDTA of 2-cell block was 67%. This seems to be caused by regulation of Ca++ function which is the major divalention in the culture medium. It is further needed for the study of Ca++ functions in mouse 2-cell block.
