Abstract
ferritin H-chain은 Fe^(2+) 를 Fe^(3+) 로 산화 (ferroxidase 반응) 시키면서 iron 의 축적을 촉진하고 ferritin L-chain 은 수화 과정을 통해 ferritin 내부에 Fe^(3+) 을 축적시키는 역할을 한다. Ferritin 은 오래 전부터 연구되어 왔지만 ferroxidase 반응 동안에 흘러나오는 전자의 이동이 어떻게 되는지 그 경로는 아직 밝혀지지 않았다. 이 연구에서는, H-ferritin 의 cysteine 이 ferroxidase 반응을 나타내는 과정에서 중요한 역할을 한다는 것을 밝혔다. 인간의 ferritin H-chain 은 3개의 cysteine을 보유하고 있는데 각각 아미노산 103번과 131번, 그리고 91번이다. 이 중 103번과 131번 아미노산과는 달리 91번 아미노산은 오로지 인간의 H-ferritin 에만 존재한다. Mass spectrometric 분석을 통해 재조합한 H-ferritin 에 분자내 cysteine 이중결합이 존재함을 발견하였고 , 이 이중결합이 cysteine 91번과 103번으로 이루어져 있음을 알게 되었다. H-ferritin 의 AMS 표지 분석과 cysteine 돌연변이 단백질을 가지고 분석한 결과 cysteine 91번이 -SOH로 산화되어 cysteine 103번과 이중결합을 이루는 중요한 역할을 하는 cysteine임을 밝혔다. 분자내 disulfide 결합은 다양한 세포종을 이용한 실험에서도 확인되었다. 또한 철 수용 활성도 측정 실험을 통해 야생형 H-ferritin이 cysteine 돌연변이에 비해 활성도가 떨어짐을 확인하였다. 흥미롭게도, 높은 농도의 Fe^(2+) 하에서는 야생형과 돌연변이의 활성도 차이가 사라짐을 확인하였다. 2D 분석을 통한 실험으로 높은 농도의 Fe를 처리했을 경우 H-ferritin이 원래 위치보다 더 산성 쪽으로 옮겨가는 것을 확인하였고 이것이 cysteine의 산화 작용에 의한 현상이라고 추측하였다. 이 실험을 통해 또한 ferritin 의 ferroxidase 반응이 가역적인 cysteine 이중결합을 통한 반응임을 추측할 수 있었다.; Ferritin H-chain catalyzes iron uptake via oxidation of Fe^(2+) to Fe^(3+) (ferroxidase reaction) and ferritin L-chain facilitates accumulation of Fe^(3+) into the ferritin cavity via hydration. Although ferritin has long been studied, the fate of an electron released from ferroxidase reaction is still controversial. In this study, I demonstrate potential role of cysteine residues in H-ferritin in ferroxidase reaction. Human ferritin H-chain has three cysteine residues: two, Cys103 and Cys131, are highly conserved among mammalian proteins and the other one, Cys91, present only in human protein. Through mass spectrometric analysis of H-ferritin on non-reducing gel, I found that there is an intramolecular disulfide linkage between Cys91 and Cys103 residues of recombinant H-ferritin. Biochemical experiments with AMS-labeling system and cysteine mutants show that Cys91 residue of H-ferritin is a primary oxidation site, by which cysteine sulhydryl is converted to cysteine sulfenyl (-SOH), and forms disulfide with Cys103 residue. Intramolecular disulfide linkage is also observed in endogenous ferritin H-chain in various cell types. I then show that cysteine mutants have a defect in iron uptake activity as compared with that of WT at low concentration of iron. Interestingly, the difference of iron uptake activity between WT and cysteine mutants is disappeared at higher concentration of Fe^(2+). By 2-dimensional PAGE analysis, I show that new spots are appeared at more acidic position compared to that of original spot and this might be oxidation of cysteine residue of H-ferritin by high iron concentration. Taken together, the studies suggest that reversible formation of cysteine disulfide could be involved in ferroxidase reaction of ferritin.
