Abstract
대맥종자의 Amylase생성에 미치는 GA와 IAA의 영향을 알아보기 위하여 GA(10-6M)와 IAA(10-5M)를 단독 또는 동시 투여하여 핵산생성과 아울러 Amylase의 생성 및 그 fraction의 변화를 조사 검토하였다. 실험결과 다음과 같은 사실들을 알 수 있었다. 1.GA와 IAA처리에 따른 DNA함량변화는 없었다. 2.RNA는 GA에 의하여 생성이 촉진되었고 IAA에 의하여서는 억제되었다. 3.Amylase는 배양8시간 후부터 생성되기 시작하였는데 그 생성은 GA에 의하여 촉진되었고 IAA에 의하여 억제되었다. 4.RNA생성은 배양 5시간에 이미 시작되어 Amylase생성의 시작보다 앞서고 있었다. 5. Amylase생성의 GA에 의한 촉진과 IAA에 의한 억제는 RNA생성의 촉진과 억제를 통하여 이루어지다고 생각되었다. 6.crude amylase는 다시 fraction I, II, III, IV로 분획할 수 있었으며 이들은 Km값, Ki값, 온도, pH, 금속이온 등에 대한 반응에서 효소화학적 성질을 달리하고 있었다. 그리고 이들의 효소화학적 성질로 미루어 FI는 b-Amylase, FIV는 a-Amylase인 것으로 생각되었다. 7.GA와 IAA에 의한 Amylase생성의 촉진과 억제는 모든 fraction에 걸쳐서 균등하게 있었는데 soluble protein당 Amylase양으로 보았을 때 IAA의 억제효과는 FIV에서만 있었고 FI에서는 뚜렷하지않았다. 8. Amylase생성에 있어 억제적인 IAA와 촉진적인 GA의 관계는 비경쟁적이어서 이를 hormone의 세포 내에서의 작용점이 다르다는 것으로 알 수 있었다. ; Using barley seed (Hordeum sativum Jess, var.), the influences of Gibberellic acid(GA) and Indole-3-acetic acid(IAA) on the Amylase synthesis and that of the Nucleic acid metabolism were investigated. 1.With the deembyronized barley seeds, the increase of amylase treated with a 10-6M concentration of GA and the decrease of Amylase treated with a 10-5M concentration of IAA were matched by a proportionate increase and decrease in the amount of RNA. The influence of the hormones on the RNA synthesis has appeared immediately after the treatment but on the amylase synthesis it has appeared 8 hours later. But no influence on the DNA synthesis was observed on both hormones. 2.The amylase from deembryonized barley seeds treated with GA and IAA have been fractionated by Gel filtration on Sephadex G-100. The amylase component showed four fractions on both enzyme treated with GA and IAA. Fraction I(FI) was differed from fraction IV(FIV) in Km value and the effects of temperature, pH and metal ions. On the basis of their enzymatic properties, it was considered that FI was b-amylase and FIV was a-amylase. The influences of GA and IAA on each fractions were appeared to be similar but on the Amylase units per soluble protein, IAA inhibited the production of amylase IV while it promoted that of amylase FI. 3.An experiment was conducted to determine whether IAA inhibits GA-promoted amylase competitively or non-competitively. Using a lineweaver-Burk plot, it was clear that IAA was acting in a non-competitive fashion. From this, IAA was probably not competing with GA at the same site, but it was acting at some other site which resulted in partial blocking of the action of GA the amylase synthesis.
