Abstract
TNF-related activation-induced cytokine (TRANCE), a TNF family member, and its receptor, TRANCE-R, mediates survival of dendritic cells in the immune system and is required for osteoclast differentiation and activation in the bone remodeling. The present study investigated the regulatory mechanism of TRANCE-induced osteoclastogenesis and cytokine expression, focusing especially on reactive oxygen species (ROS)-mediated signaling in osteoclast progenitor cell line, RAW264.7 cells and bone marrow-derived macrophage cells. TRANCE increased intracellular ROS and TRANCE-induced ROS generation was blocked by PI3-K inhibitor, LY294002 and wortmannin and c-Src inhibitor, PP1. These results showed that TRANCE generated ROS through PI3-K and c-Src. In addition, PI3-K inhibitor strongly blocked the TRANCE-induced activation of p38 mitogen-activated protein kinase (MAPK) and also attenuated the osteoclast differentiation by stimulation of TRANCE. Considering that TRANCE-induced osteoclastogenesis is triggered by p38MAPK activation, these results define a mechanism by which TRANCE activates p38MAPK via ROS generation, which may contribute to osteoclastogenesis. In RAW264.7 cells, TRANCE induces the expression of IL-1β through p38MAPK pathway in a ROS-sensitive manner in addition to enhancing osteoclast formation. TRANCE increased IL-1β gene expression, which was attenuated by LY294002, wortmannin, NAC, and p38MAPK inhibitor, SB203580. Collectively, these data indicate that TRANCE stimulates ROS production via PI3-K and c-Src, and that these ROS mediate activation of p38MAPK, which culminates in osteoclast differentiation and IL-1β gene expression in osteoclast progenitor cells.;TNF family에 속하는 TNF-related activation-induced cytokine (TRANCE)는 TRANCE receptor를 자극하여 면역세포인 dendritic cells의 생존을 매개하며 골밀도의 균형을 유지하도록 파골세포로의 분화를 자극한다. 본 연구에서는 파골세포의 전구세포인 RAW264.7 세포와 골수에서 직접 분리해 낸 파골 전구세포에서 TRANCE에 의해 유도되는 파골세포 분화와 cytokine의 발현 조절 메카니즘을 H₂O₂와 같은 활성세포종 (ROS)에 주안점을 두고 조사해 보았다. TRANCE는 활성세포종 생성을 증가시켰고 이는 PI3-K 억제제인 LY294002와 wortmannin 그리고 c-Src 억제제인 PP1에 의해 감소되었다. 또한 Rac1의 Dominant negative mutant인 RacN17를 발현시킨 경우와 NADPH oxidase억제제인 DPI를 전처리한 경우에 TRANCE 자극을 가해도 ROS level에는 변화가 없었다. 이 결과로 TRANCE는 PI3-K, c-Src, Rac1/NADPH oxidase를 통해 활성세포종을 생성시킴을 확인할 수 있었다. TRANCE는 extracellular signal-regulated kinase (ERK), c-Jun NH2-terminal kinase (JNK), 그리고 p38MAPK와 관련된 세포 내 신호를 전달한다고 알려져 있다. 이러한 세가지 MAPK는 항산화제인 N-acetylcysteine (NAC)에 의해 부분적으로 저해됐다. 반면, PI3-K 억제제에 의해서는 p38MAPK만 강하게 감소했다. 이로서 TRANCE자극 후, PI3-K를 통해 생성된 ROS는 p38MAPK를, PI3-K를 경유하지 않고 생성된 ROS는 ERK와 JNK를 활성화시킬 것이라 예상할 수 있었다. 한편, PI3-K억제제는 TRANCE에 의한 파골세포 분화를 억제시켰다. TRANCE에 의해 유도되는 파골세포 형성이 p38MAPK 신호를 받아 활성화된다는 보고에 근거해 TRANCE는 ROS를 매개로 p38MAPK를 자극해 결국 파골세포를 형성시킬 거라 추측할 수 있었다. 면역세포인 dendritic cells에서 TRANCE는 염증유도 cytokine인 IL-1, IL-6와 T cell 성장과 분화를 유도하는 cytokine IL-12, IL-15를 발현시킨다는 보고가 있다. 이를 토대로 본 실험에서는 파골세포의 전구세포인 RAW264.7를 써서 TRANCE에 의해 유도되는 cytokine과 이러한 cytokine 발현에 ROS가 관여하는지를 알아 보았다. 항산화제인 NAC과 RAW264.7 세포의 ROS 생성에 관여할 거라 예상되는 PI3-K 억제제에 의해서 또한 p38MAPK 신호전달을 제어하는 SB203580에 의해서도 IL-1β 발현이 감소되었다. 그 결과, TRANCE는 ROS를 매개로 p38MAPK를 활성화시켜 IL-1β 유전자발현을 유도함을 확인할 수 있었다. 요약하면, 본 연구는 파골 전구세포주인 RAW264.7 세포와 골수에서 직접 분리한 파골 전구세포에서, TRANCE는 PI3-K, c-Src, Rac1/NADPH oxidase를 경유해 ROS를 생성시키고 이 때 생성된 ROS는 p38MAPK를 활성화시켜 결국 파골세포 분화와 IL-1β 발현을 유도함을 알 수 있었다.
