Caspase에 의해 유도되는 nucleosomal DNA 절편화에 대한 연구

Abstract

nucleosomal DNA 절편화는 caspase가 활성화되면서 일어나는 세포사멸시 일어나는 대표적인 현상이다. nucleosomal DNA 절편화가 일어나기 위해서는 DNase complex에서 저해제로 작용하는 ICAD/DFF45가 잘라지면서 CAD/DFF40이 활성화되는 것이 필요하다. ICAD/CAD-DFF45/DFF40 protein complex는 핵안에 대부분 존재한다. 하지만 이와는 일치하지 않게 분리된 핵에 어떠한 세포 추출물이나 정제된 단백질을 추가로 넣어주지 않고 caspase단독에 의해서 DNA 절편화가 유도되지 않는다고 알려져 있다. 그러나 본 실험에서는 50-75mM KCl 또는 NaCl의 조건에서 caspase-3와 -7에 의해서 다른 추가적인 요소 없이 단독으로 DNA 절편화를 일으켰다(caspase-6와 -8에서는 일어나지 않는다.). ICAD에 의해 조절되는 이 DNase의 활성은 핵 추출물을 첨가할 때 뚜렷하게 나타난다. 이는 ICAD/CAD-DFF45/DFF40 protein complex가 핵 안에 존재한다는 것을 기능적으로 증명하는 것이다. CAD/DFF40가 유도한다고 알려져 있는chromatin condensation은 caspase-3를 처리한 핵에서는 일어나지 않았다.;For nucleosomal DNA fragmentation (DF), one of hallmarks of apoptosis, activated caspase, an apoptosis specific cysteine protease, is required to cleave ICAD/DFF45 that releases its complexed DNase, CAD/DFF40. The ICAD/CAD-DFF45/DFF40 protein complex was immunodetected predominantly in the nuclei. Inconsistently, caspase alone was known to be unable to induce DF in the isolated nuclei without the addition of cell extract or purified CAD/DFF40. In this study, however, it is demonstrated that under selected conditions with 50-75 mM of KCl or NaCl caspase-3 and -7 (but not -6 and -8) could induce the DF without the additional factor(s). The DNase activities sensitive to ICAD were recovered predominantly in the nuclear extract, consistent with the predominant nuclear localization of ICAD/CAD-DFF45/DFF40, providing a functional identification of the protein complex in the nuclei. Chromatin condensation which CAD/DFF40 was also known to induce was not observed in the caspase-3 treated nuclei, contradicting the DNase as a chromatin condensation factor.
