dc.description.abstractSpemann 과 Mangold 의 유명한 이식 실험과 Nieuwkoop 이 수행한 일련의 실험 내용이 발표된 이후, 배엽의 분화와 체축의 형성을 비롯한 초기 배아의 발생 과정에 필수적인 많은 세포 신호전달 체계가 발견되었고, 그에 대한 여러 연구가 진행되어 왔다. 이 중, Nodal signaling 은 중배엽/내배엽 형성과 초기 체축 분화에 대단히 중요하며, FGF signaling 은 중배엽/내배엽 형성과 낭배형성을 위한 세포 이동, 신경 유도, 그리고 전후축 형성에 중요한 역할을 수행한다. 본 논문에서는 Xenopus 배아 발생 과정 동안 이들 Nodal signaling 과 FGF signaling 의 메커니즘에 대해 연구하였다. Nodal signaling 은 Xenopus laevis 의 초기 배아 발생 과정 중 중배엽 유도와 체축 형성에 중심적인 역할을 수행한다. Nodal signaling 의 필수적인 보조 인자로 알려진 EGF-CFC 는 보존된 EGF-like motif 를 가지고 O-fucosylation 이 일어난다는 사실이 CHO 세포에서 보고된 바 있다. 생쥐와 초파리의 경우, Notch 단백질의 보존된 여러 EGF-like motif 역시 O-fucosylation 이 일어나며, 이는 POFUT1 에 의해 일어나는 것으로 알려져 있다. 여기서 나는 Xenopus laevis 의 이종상동 유전자 POFUT1 을 분리하고 이를 XPOFUT1 이라 명명하였다. XPOFUT1 은 수정 전 난자에서부터 발현하고 있으며 초기 발생 과정 동안 수정란에서도 발현한다. 또한 XPOFUT1 은 포배기에 수정란 전체에 걸쳐 발현하고 있다가 낭배기가 되면 원구순 주위에서 강하게 발현한다. 미뢰기와 올챙이 시기 배아에서, XPOFUT1 은 뇌와 눈, 아가미궁, 앞신장과 원체절에서 발현한다. XPOFUT1 발현의 억제는 축 형성과 원체절 형성, 심장 발생, 신경 발생에 결함을 일으킨다. 이들 결함은 XCR1/3 가 결핍된 배아에서 나타나는 결함과 유사하다. XPOFUT1 발현의 억제는 또한 늦은 포배 시기에 activin 반응 요소의 활성과 Smad2 인산화를 감소시키고, Nodal 의 타깃 유전자의 발현을 억제시킨다. 또한 나는 XPOFUT1 이 Xenopus 배아에 존재하는 다수의 Xnr signaling 을 발생 중 시공간적으로 조절한다는 사실을 발견하였다. 이러한 결과는 XPOFUT1 이 EGF-CFC 단백질의 발현 후 변형 과정을 통해 Nodal signaling 을 조절하여 초기 Xenopus 배아 발생 과정에서 중요한 역할을 수행한다는 사실을 보여준다. Frs2 (FGF receptor substrate 2) 는 FGF 수용체의 활성을 그 후속 인자들에게 전해주는 매개체로서 앞서 발견되었다. 여기서 나는 두 번째 Xenopus Frs 동족체, XFrs3 를 찾아, Xenopus 의 배아 발생 과정 동안의 역할에 대해서 연구하였다. XFrs3 는 수정 전 난자에서부터 발현하고 있으며 초기 발생 과정 동안 수정란에서는 특정한 발현 양상을 보인다. 미뢰기 배아에서, XFrs3 는 머리, 눈, 원체절에서 발현한다. XFrs3 의 기능 억제를 위해, 나는 먼저 tyrosine 의 인산화가 불가능한 XFrs3 를 사용하였고, 이의 과발현은 XFrs2 의 기능에 대한 이전 보고에서와 마찬가지로 FGF 에 의한 중배엽 형성을 방해한다. 하지만 XFrs3 mRNA 에만 특정하게 반응하도록 만들어진 morpholino 를 사용하면 눈과 머리 발생에 결함을 야기하고, 특히 렌즈 유도를 의미하는 표지 유전자 Pax6 의 발현을 감소시킨다. 이러한 결함은 XFrs3 mRNA 에 의해 회복되나 Shp2 결합 부위에 돌연변이가 일어난 XFrs3 (XFrs3-2YF) 에 의해서는 회복되지 않는다. 마지막으로, 나는 XFrs3 가 FGF 혹은 IGF 에 의해 유도되는 ERK 활성에 필요할 수 있다는 증거를 보였다. 이러한 결과는 XFrs3 의 Shp2 결합 부위를 통한 세포 내 신호전달이 Xenopus 배아의 눈 형성 과정에 필수적임을 보여준다.;Since the transplantation experiment of Spemann and Mangold and a series of experiments by Nieuwkoop were published, a number of signaling pathways which are essential for early embryonic development including the specification of germ layers and axial patterning were identified. Among these signaling pathways, Nodal signaling is critical for the formation of mesoderm and endoderm and the specification of the primary body axes, and FGF signaling plays important roles in mesoderm and endoderm formation, gastrulation movements, neural induction, and anterio-posterior patterning. In the present thesis, I studied about the mechanism of the Nodal and FGF signaling during Xenopus embryonic development. Nodal signaling plays pivotal roles for mesoderm induction and axis formation during early embryogenesis of Xenopus laevis. It has been previously reported that EGF-CFC factor, an essential co-factor of Nodal signaling, is O-fucosylated at the conserved EGF-like motif in CHO cells. In mouse and Drosophila, the conserved EGF-like motives of Notch proteins are O-fucosylated by POFUT1. I isolated an ortholog of POFUT1 from Xenopus laevis and named it XPOFUT1. XPOFUT1 is expressed maternally and zygotically through the early development. XPOFUT1 is expressed ubiquitously in blastula and around the blastopore lip in gastrula. In tailbud and tadpole stage embryos, XPOFUT1 is expressed in the brain, eyes, branchial arches, pronephros, and somites. Knockdown of XPOFUT1 causes defects in axis formation, somitogenesis, cardiogenesis, and neurogenesis. These defects are similar to those of XCR1/3-deficient embryos. XPOFUT1 knockdown also reduces the activation of an activin-responsive element and Smad2 phosphorylation in late blastula, and suppresses the expression of Nodal target genes. In addition, I found that XPOFUT1 regulates multiple Xnr signaling spatio-temporally. My results suggest that XPOFUT1 plays important roles during early Xenopus embryogenesis, at least in part, by regulating Nodal signaling possibly through the post-translational modification of EGF-CFC factors. Frs2 (FGF receptor substrate 2) has been previously identified as a mediator that links the activation of FGF receptor to its downstream effectors. In this study, I identified the second Xenopus Frs homolog, Xenopus Frs3 (XFrs3), and investigated its roles during Xenopus embryogenesis. XFrs3 is expressed maternally and zygotically with specific expression patterns throughout the early development. XFrs3 is expressed in the head, eyes, and somites in tailbud stage embryos. For knockdown of XFrs3 function I first used a tyrosine unphosphorylatable form of XFrs3, and its overexpression interferes with FGF-dependent mesoderm formation like XFrs2. However, a specific morpholino of XFrs3 causes defects in the development of eye and head, particularly the expression of a specific eye marker gene (Pax6). These defects are rescued by XFrs3 mRNA, but not by a mutant of the Shp2-binding sites (XFrs3-2YF). Finally, I present evidence that XFrs3 can be required for FGF or IGF induced ERK activation. My results suggest that intracellular signaling through Shp2-binding sites in XFrs3 is essential for eye formation of Xenopus embryos.-
