Abstract
비만은 전세계적으로 폭발적으로 증가하고 있는 추세이며, 만성비만으로 인한 대사증후군은 심장질환의 발생에 있어 강력한 위험인자로 작용하는 것으로 알려져 있다. 이 과정에서 복부지방을 구성하고 있는 지방세포에서의 산화성 스트레스의 증가와 미토콘드리아의 기능 장애로 인해 지방세포에서 분비하는 아디포카인에 이상이 생기며 그 결과 전신의 염증반응과 인슐린 저항성 등이 야기된다. 따라서, 지방세포에서의 미토콘드리아 산화-환원 조절과 그 역할에 대한 연구가 중요하게 여겨지고 있다. 본 논문에서는 3T3-L1의 지방 세포 분화과정에서 Peroxiredoxin 3 (Prx3)라고 하는 미토콘드리아에 특이적으로 존재하는 항산화효소가 크게 증가함을 관찰하였다. 지방세포에서의 Prx3의 역할에 대해서는 아직 연구된 바가 없으므로, 본 논문에서는 Prx3가 지방세포의 산화성 스트레스, 미토콘드리아 기능, 그리고 아디포카인 생산에 중요한 역할을 할 것이라는 가설을 세우고 이에 대해 연구하였다. Prx3가 결핍된 생쥐로 실험 한 결과, wild type (WT)에 비해 Prx3 knockout (KO) 생쥐에서 복부 지방의 축적이 증가하였으며 WT과 Prx3 KO 생쥐에서 얻은 지방유래 줄기세포를 분화시킨 결과 Prx3가 결핍된 경우 지방세포화 인자와 지방생성 유전자의 발현이 유의하게 증가하였다. 또한 프로테오믹스 기술과 유전자 발현 분석을 통해 Prx3가 결핍된 지방세포는 미토콘드리아 생합성과 당/지방의 대사, 그리고 항산화 조절 기전에 이상이 있음을 관찰하였다. 그리고 3T3-L1 세포에서Prx3의 발현을 siRNA로 억제시켰을 때 미토콘드리아 내 산화성 스트레스가 증가하였다. Prx3의 결핍이 지방세포의 기능에 어떠한 영향을 미치는지 알아보기 위해 대표적 아디포카인인 adiponectin과 plasminogen activator inhibitor-1 (PAI-1)의 발현을 측정한 결과 Prx3가 결핍된 지방세포에서 유전자와 단백 발현 모두에서 adiponectin이 감소하고 PAI-1이 증가함을 관찰하여 Prx3가 결핍된 상태에서 지방세포의 기능에 문제가 생김을 규명하였다. 나아가 Prx3 KO쥐에서 당불내성과 인슐린 저항성이 일어남으로 보아 전반적인 대사에 문제가 생겼음을 알 수 있었다. 특징적으로, 비만한 db/db 생쥐의 간과 신장에서는 db/m 생쥐에 비해 Prx3의 발현이 증가한 반면 지방조직에서는 Prx3의 발현이 유의하게 감소한 것을 통해 Prx3의 결핍이 비만과 밀접한 상관관계가 있음을 규명하였고 이러한 결과는 BMI 25이상의 과체중인 사람의 지방조직에서도 동일한 결과를 나타내었다. 8-dihydro-2-deoxyguanosine (8-OHdG)은 DNA 염기서열이 산화성 스트레스의 공격을 받아 생기는 부산물로서 최근 연구들에서 항산화, 항염증 작용을 지니는 것으로 알려졌다. 따라서 본 논문에서는 8-OHdG이 비만으로 인한 여러 유해한 반응을 치료할 수 있는 신약 후보 물질이 될 수 있는지 지방세포에서 검증하고자 하였다. 3T3-L1에 8-OHdG를 투여하자 지방세포로의 분화과정 중에 지방화 인자들이 감소하였으며 PAI-1의 투여로 인한 fatty acid synthase의 발현 증가가 8-OHdG의 전처리를 통해 억제되었다. 또한 8-OHdG가 Prx3 결핍으로 인해 증가된 미토콘드리아 내 산화성 스트레스를 감소시키는 것으로 보아 항산화 작용을 통해 지방세포의 기능을 원활하게 유지시킬 수 있다는 것을 규명하였다. 본 논문에서는 Prx3가 지방세포의 기능을 정상적으로 유지시키는 데에 중요한 역할을 한다는 것을 밝혔으며 Prx3가 결핍되었을 경우 항산화 조절과 미토콘드리아의 기능 그리고 아디포카인의 발현에 이상으로 이어져 비만을 야기하고 대사증후군으로 진행될 수 있음을 규명하였다. ;Obesity is one of the most prevalent chronic diseases worldwide. Increased oxidative stress and mitochondrial dysfunction in obese adipocytes contributes to adipokine dysregulation, inflammation, and insulin resistance. Therefore, understanding the mitochondrial redox state and its function in adipocytes is an important matter. Peroxiredoxin 3 (Prx3) is a thioredoxin-dependent mitochondrial peroxidase and its level dramatically increases during 3T3-L1 adipocyte differentiation. To date, the role of endogenous Prx3 in adipocyte metabolism has not been elucidated. In this thesis, I hypothesized that endogenous Prx3 plays an important role in regulation of adipocyte oxidative stress, mitochondrial biogenesis, and adipokine expression. The role of Prx3 in adipocytes was examined employing Prx3 knockout (KO) mouse and 3T3-L1 cells transfected with Prx3 siRNA. Compared to wild type mice, Prx3 KO mice had increased fat mass due to adipocyte hypertrophy. Differentiation of adipose tissue-derived stem cells from Prx3 KO mice showed increased adipogenic transcription factors and lipogenic gene expression. An advanced proteomic analysis and gene expression analysis of Prx3 KO mice adipocytes showed defect in mitochondria biogenesis along with enzymes involved in glucose/lipid metabolism and oxidative phosphorylation. Increased nitrotyrosine accumulation in Prx3 KO adipose tissue and increased mitochondrial superoxide level in Prx3 knockdown 3T3-L1 cells showed aberrant regulation of oxidative stress. In addition, expression level of adiponectin was downregulated and plasminogen activator inhibitor-1 (PAI-1) was upregulated in Prx3 KO adipocytes. Impaired glucose tolerance and insulin resistance in Prx3 KO mice further implied metabolic dysregulation. Interestingly, Prx3 levels were increased in liver and kidneys of obese db/db mice, but were significantly decreased in obese db/db mice and human subjects indicating that Prx3 levels are closely related with development of obesity. Exogenous treatment of 8-dihydro-2-deoxyguanosine (8-OHdG) has been recently rediscovered to possess anti-inflammatory and anti-oxidative effects. In an attempt to find the therapeutic target to ameliorate the detrimental effects of oxidative stress-induced diseases including obesity, the last part of the thesis was focused on the effect of exogenous 8-OHdG on adipocytes. 8-OHdG treatment significantly decreased adipogenic gene expression during 3T3-L1 adipogenesis. Also, increased mitochondrial oxidative stress in Prx3 knockdown adipocytes were ameliorated by 8-OHdG treatment. Suppression of PAI-1 induced fatty acid synthase expression by 8-OHdG suggests that 8-OHdG may regulate adipocyte lipid metabolism partly through anti-oxidative effect. In summary, these data suggest that endogenous Prx3 plays an essential role in maintaining normal characteristics of adipocytes and that defect in Prx3 alters antioxidant system, mitochondrial function, and adipokine expression in adipocytes leading to metabolic alteration.
