- Abstract
- DNA polymerase (pol) k is one of the human Y-family polymerases, which is involved in error-free translesion DNA synthesis (TLS) through minor-groove DNA lesions such as N2-benzo[a]pyrene diolepxide(BPDE)-G and thus can accurately and efficiently synthesize DNA against N2-BPDE-G. The genetic polymorphisms of pol k in individuals can be a genetic factor for determining the mutation or cancer susceptibility for minor-groove-DNA adduct-forming carcinogens such as polycyclic aromatic hydrocarbons (e.g. benzo[a]pyrene) and heterocyclic amines, but their roles are not well understood. To assess the effect of genetic variations in pol k on TLS, we analyzed the kinetic properties of six natural polymorphic variants, Leu21Phe, Asp189Gly, Arg219X, Arg219Ile, Ser423Arg, and Tyr432Arg variants to copy past G and a minor-groove adduct N2-CH2-9-anthracenyl(Anth)G, using the corresponding catalytic core enzymes purified after heterologous expression in Escherichia coli.
The truncating R219X variant was devoid of polymerase activity. In steady-state kinetic analyses, E189G displayed ~4-fold decrease in kcat/Km for dCTP insertion opposite N2-AnthG compared to wild-type enzyme, while S423R exhibited ~6-fold increase in kcat/Km for that. L21F, D189G, and Y432S may reduce their TLS abilities, and S423R may rather increase its TLS ability of pol κ upon carcinogen-bound minor-groove DNA lesions, which emphasizes the necessity of further evaluation for functional consequences of these genetic variations of pol κ in vivo, for example, the increase or decrease of individual cancer susceptibility to carcinogen exposure such as polycyclic aromatic hydrocarbons and heterocyclic amines;인체의 Y-family DNA polymerase들 중 하나인 DNA polymerase (pol) κ는 N2-benzo[a]pyrene diolepxide(BPDE)-G와 같은 minor-groove DNA lesion들을 통과하는 error-free translesion DNA synthesis (TLS)에 관여하며, N2-B[a]P-G및 N2-IQ-G에 대하여 정확하고 효율적인 DNA 합성을 한다고 알려져 있다.
따라서 인체에서 pol κ의 유전적 변이가 minor-groove DNA adduct를 형성하는 발암원들에 의한 돌연변이 또는 암 발생 감수성에 영향을 미치는 인자의 하나로 작용할 가능성을 예측하고 본 연구를 진행하였다.
Pol κ의 유전적 변이가 pol κ의 TLS 기능에 미치는 효과를 알아보고자, wild-type 효소와 Leu21Phe(L21F), Asp189Gly(D189G), Arg219X(R219X), Arg219Ile(R219I), Ser423Arg(S423R) 및 Tyr432Arg(Y432S)의 6종의 genetic variant의 catalytic core를 각각 Escherichia coli 에서 발현 후 분리한 다음 이들의 정상 G과 minor-groove adduct인 N2-CH2-anthracenyl(Anth)-G에 대한 DNA합성 시 효율성과 정확성을 측정하는 kinetic 분석 실험을 수행하였다.
그 중 절단된 형태의 R219X variant는 polymerase 활성이 전혀 없었고, steady-state kinetic 분석 결과 D189G는 N2-AnthG 에 대해 dCTP를 삽입할 시의 kcat/Km 값이 wild-type에 비하여 약 4배 정도 감소한 것을 보였으며, S423R은 약 6배 정도 증가한 것을 보였다.
Pol κ의 L21F, D189G및 Y432S 변이체들의 경우 각종 발암물질에 의해 형성되는 minor-groove DNA 손상들에 대한 TLS 기능이 감소하며, S423R변이체의 경우는 반대로 그 TLS 기능이 증가할 것으로 예측된다.
앞으로 이들 결과들을 바탕으로 pol κ의 유전적 변화들이 초래하는 in vivo 기능 변화 - 예를 들어 polycyclic aromatic hydrocarbon과 heterocyclic amines등의 발암물질 노출에 대한 개개인에서의 발암 감수성의 증감 관련성 평가 - 에 대해서도 보다 심층적으로 연구할 필요가 있을 것으로 기대된다.
