Abstract
Streptomyces tenebrarius에 의해 생산되는 토브라마이신 (tobramycin)은 카나마이신(kanamycin)과 함께 4,6-disubstituted aminoglycoside 계열에 속하는 항생제로써, 다양한 세균성 감염 치료에 사용된다. 최근에는 β-lactam 계열의 항생제와 병용할 경우 다재저항성 (multi-resistance)을 가지는 Acinetobacter 세균과 열병 호중구 감소성 암환자에게도 효과적인 것으로 보고되고 있다. 이처럼 tobramycin의 임상학적 가치는 꾸준히 증가하고 있는 반면, tobramycin의 생합성 과정에 대해서는 밝혀진 바가 많지 않으며, 생산균주인 S. tenebrarius에서는 유전자 조작이 어려운 문제점이 있다. 따라서 본 연구에서는 tobramycin과 구조적으로 유사한 kanamycin B의 생합성 경로를 규명한 선행연구 결과를 바탕으로 이종숙주인 Streptomyces venezuelae에서 tobramycin을 생합성하고, 더불어 tobramycin의 생합성에 관여하는 일부 유전자들의 기능을 규명하고자 하였다. 우선 다양한 아미노글리코사이드 유전자군의 생물정보학적 분석을 통해 3'-deoxygenation에 관여할 것으로 예상되는 유전자 세트 aprD3-aprD4와 tobramycin 유래의 두 번째 당전이 효소 코딩 유전자인 tobM2를 선정하였다. 이후, kanamycin B를 생산하는 발현벡터와 추가적인 클로닝을 통하여 재조합 균주를 제작하고 분석한 결과, 3'-deoxykanamycin A, 3’-deoxykanamycin C와 더불어 0.4mg/L의 tobramycin (3’-deoxykanamycin B)이 생산되었다. 최종 생산물인 tobramycin을 포함한 3'-deoxykanamycin들은 모두 ESI-MS/MS를 통해 확인하였다. 또한, S. tenebrarius 및 S. venezuelae의 세포외추출물 (cell-free extracts)을 이용해, 3’-deoxygenation에 관여하는 AprD3-AprD4의 기능을 규명하였으며, TobM2의 경우 아미노글리코사이드 생합성 중간체인 파로마민 (paromamine)과 2’-디아미노-2’-하이드록시니아민 (2’-deamino-2'-hydroxyneamine)의 3’-deoxy anolog 및 네브라민 (nebramine)을 당수용체로 사용할 수 있다는 것을 확인할 수 있었다. 결과적으로, 다양한 아미노글리코사이드 유전자들의 조합생합성 및 이종숙주 발현을 통해 tobramycin 생산에 성공하였으며, AprD3-AprD4 및 TobM2의 기능을 규명하였다. 이를 바탕으로 추후 연구에서도 조합생합성기법을 이용하여 다양한 구조의 아미노글리코사이드를 생합성하고 각 유전자의 기능을 규명함으로써, 내성균의 증가로 인해 그 요구가 증대되고 있는 신규 항생제 개발에 유용하게 활용될 수 있을 것으로 기대된다.;Tobramycin produced by Streptomyces tenebrarius belongs to 4,6-disubstituted aminoglycoside together with kanamycin. It is clinically important antibiotics with a broad antibacterial spectrum, particularly against Gram-negative bacteria. Recently, it is reported that tobramycin is effective even against the Acinetobacter bacteria which cause multi-drug resistance and for treatment of cancer patients with febrile neutropenia. However, the biochemistry and genetics underlying the biosynthesis of tobramycin have still remained unclear, due to difficulties in genetically manipulating the producing strain and obtaining soluble functional enzymes. In this study, we produced tobramycin and its analogs through the heterologous expression of combinations of putative biosynthetic genes from Streptomyces tenebrarius and Streptomyces kanamyceticus in the Streptomyces venezuelae lacking the genes for biosynthesis of the native deoxysugar (thymidine 5’-diphopho-desosamne). In addition, we determined the function of TobM2 and AprD3-AprD4 using the cell-free extracts of S. tenebrarius and S. venezuelae expressing the corresponding genes. The gene products of aprD3-aprD4 from S. tenebrarius are responsible for the 3’-deoxygenation of pseudodisaccharides, paromamine, 2’-deamino-2’-hydroxyneamine and neamine. Besides, the second glycosyltransferase TobM2 from S. tenebrarius transfer kanosamine to the resulting 3’-deoxygenated psudodisaccharides, leading to the production of 0.4mg/L (0.8μM) tobramycin, along with 3’-deoxykanamycin A and 3’-deoxykanamycin C. The structure of 3’-deoxykanamycins including tobramycin was confirmed by their retention behaviors and ESI-MS/MS fragmentation patterns. This work demonstrates the potential for direct in vivo production of clinically useful antibiotics using combinatorial biosynthesis.
