dc.description.abstract펩타이드는 여러 생물학적 현상에서 다양한 역할을 하기 때문에 흥미 있는 물질로 여겨진다. 하지만 생체 내 효소에 의한 분해, 낮은 세포투과성 등의 제한성을 가지기 때문에 연구와 응용에 한계가 있다. 따라서, 의학, 생물학, 약학 등의 연구에서 이러한 단점을 극복할 수 있는 펩타이드 모방체의 개발은 매우 중요한 주제가 되었다. 펩타이드 모방체의 하나인 펩토이드는 펩타이드의 곁가지가 펩타이드 결합의 질소로 이동된 형태로 펩타이드에 비해 세포투과성이 좋고, 세포 내 가수분해에 안정하며, 고체 상에서 합성이 쉽고, 수 많은 다양성을 부여할 수 있다. 본 연구에서는 펩토이드를 기본골격으로 하여 생물학적으로 응용 가능한 화학적 도구 개발에 대한 두 가지 연구를 보고한다. 첫 번째 장에서는 유전자-기반 분석법을 이용하여 체계적으로 선형 및 고리형 화합물의 세포투과성을 비교하기 위하여 일련의 선형, 고리형 펩토이드-덱사메사손 유도체들을 합성하였다. 이 유도체들은 구조적 견고함과 유연성이 세포투과성에 어떤 영향을 미치는지 알아 볼 수 있으며, 선형 펩토이드의 길이와 고리 펩토이드의 고리 크기에 따른 세포투과성의 영향에 대해서도 중요한 정보를 제공해 주리라 기대한다. 두 번째 장에서는 세포 내 당단백질이나 당펩타이드의 역할 연구에 유용한 화학도구로서 당펩타이드 모방체인 당펩토이드의 고체상 합성에 대해 보고하였다. 부분입체이성질체인 글루코오스, 갈락토오스, 만노오스에 서로 다른 분자량을 가지는 이음이를 연결하여 O-linked 또는 N-linked 탄수화물-아민 유도체들을 효율적으로 합성하였다. 이를 이용하여 당펩토이드를 고체상에서 합성한 후 질량분석으로 효과적으로 배열을 확인할 수 있음을 보였다.;Over the years peptides have been an interesting class of molecules owing to their crucial bioactivities in many biological events. However, they often have several limitations including the susceptible enzymatic degradation and the poor cell permeability. Thus, the development of peptidomimetics has been an important issue in medicinal, biological and chemical area. Peptoids (N-substituted oligoglycines) have been extremely interesting molecules as peptidomimetics, having high structural diversity, high conformational flexibility, improved cell permeability and easy synthesis by 'esubmonomer strategy' on solid-phase. Herein two kinds of studies were reported using peptoids as chemical tools for biological applications. In chapter I, the rational design and efficient solid-phase synthesis of a series of cyclic and linear peptoid-dexamethasone conjugates are reported for their systematic cell permeability studies by using reporter gene-based assays. These model compounds may reveal how the cell permeability of cyclic molecules is affected by several physicochemical properties, especially, intramolecular or internal hydrogen bond potential, reduced conformational flexibility and ring size. In addition, the synthetic strategy that was adopted in this study can also provide a robust platform for postchemical modifications of various molecular scaffolds in solid-phase or solution-phase syntheses. In chapter II, O-linked and N-linked glycopeptoids were developed on the solid-phase using commercially available primary amines and synthetically prepared sugar amines, resulting in the construction of the glycopeptoid library. Diastereomeric sugar monomers were differentiated by using different amine linkers. Thus glycopeptoids could be successfully sequenced by tendem mass spectrometry (MS/MS).-
