dc.description.abstract본 연구에서는 Bacteria의 서로 다른 비생장속도(specific growth rate) 특징을 이용하여 환산저해모델(reduced inhibition model)을 개발하였고 이 모델을 이용하여 MTBE와 그의 대사산물(its intermediates)의 독성에 대한 정성/정량적인 영향 평가 가능성을 검증하였다. 또한 오염되지 않은 다양한 토양 환경이 MTBE와 그의 대사산물(its intermediate)에 노출되었을 때 토착 미생물 군집에 미치는 영향을 평가하기 위해 물리 화학적 요인들에 대한 조사를 비롯한 탈수소효소(dehydrogenase activity) 활성 측정, 미생물 수(biomass, CFU: colony forming units) 측정 등 다각적인 생물학적 특성을 평가하고 이들간의 상관관계를 살펴보았다. 분자 생물학적 기법인 PCR-DGGE(Polymerase chain reaction - Denaturing gradient gel electrophoresis)를 이용하여 미생물 군집 구조 변화 및 특성을 관찰하였으며, Shannon Index지수(다양성) 와 균등성 지수를 이용하여 미생물 군집의 종 다양성 조사하였다. MTBE와 그의 대사산물(its intermediate)에 노출 되었을 때 Escherichia coli, Pseudomonas aeruginosa, Bacillus amylo-liquefaci의 성장 속도 패턴(growth rate pattern)이 서로 다른 것으로 나타났으며, 독성 물질의 노출 강도가 강할수록 성장이 농도에 비례하여 저해 되었다. 특히 FA 노출 조건에서는 세 균주 모두 정체기를 가진 후 지수 성장 하는 것으로 나타났다. 또한 기질의 농도(TOC: 24,500 mg/L of LB medium 100%~5%)에 따라 독성 물질에 대한 반응 속도가 달라지는 것을 관찰 할 수 있었다. 세균의 성장 특징을 파악 한 후 각각의 비생장속도(specific growth rate)를 이용하여 저해상수(inhibition constant, KI)를 구하고 환산 저해상수(reduced inhibition constant, αi)와 환산 저해농도 (reduced inhibition concentration) 개념을 도입하였다. 대장균(E. coli)의 경우 경쟁적 저해(competitive), 비경쟁적 저해(Non-competitive) 모델 방법 모두 적용 가능했다. 특히 비경쟁적 저해 모델로 적용 시 저해상수 KI보다는 μmax,app의미를 내포한 K’I 적용 해야 했다. 환산저해상수를 이용했을 때의 EC50과 환산저해상수를 이용하지 않았을 때 EC50는 MTBE, TBA, FA 각각 75, 150, 0.8 mM로 같은 것을 확인하였다. P. aeruginosa의 경우 MTBE, TBA에 대한 내성이 비교적 강한 것으로 나타났으며 FA 노출 시 긴 정체기를 가졌다가도 시간이 지나면(1.5 mM FA 7시간) 적응 후 최대 세포 농도에 이르는 것을 관찰 하였으며 기질(substrate)의 농도가 높은 경우에 경쟁적 저해 모델을 따르다 기질 농도가 낮아지면 비경쟁 저해모델을 따르는 mixed type의 모델을 따르는 것으로 나타났으며, 대장균의 경우와 마찬가지로 저해상수 KI보다는 K’I 적용 해야 한다. 각 물질에 대한 EC50 예측할 수 있었으며, MTBE, TBA, FA 각각 250, 100, 1 mM였다. B. amylo-liquefaci의 경우 MTBE와 그의 대사산물에 대해 대장균과 P. aeruginosa에 비해 내성이 강한 것으로 나타났으며 FA 노출 조건에서는 정체기를 보인 후 지수 성장 하는 것을 관찰 할 수 있었다. MTBE 노출 조건에서는 경쟁적 조건 모델을 수렴하다가 기질 농도가 낮아지면 비경쟁저해 모델을 따랐으며 TBA와 FA 노출 조건에서는 비경쟁적 저해 모델을 따랐고 이 경우에도 저해상수 KI보다는 K’I 적용 해야 했다. 환산저해상수 적용 시 EC50은 MTBE, TBE, FA 각각 200, 400, 0.8 mM었다. 세 균주 모두 MTBE와 그의 대사산물의 내성은 TBA, MTBE, FA 순이었고 FA의 독성 강도가 센 것을 관찰 할 수 있었다. MTBE, TBA, FA의 2, 3성분 혼합계의 경우 저해상수 K’I 적용 시 각각의 농도 조건이 달라도 기준물질의 기준선과 비교적 일치하는 것으로 나타났으며 적은 농도 범위에서도 성장 저해가 쉽게 일어나는 것을 관찰 할 수 있었다. 이를 통해 어떠한 경우에라도 비생장속도를 파악 한 후 기준 물질을 정하고 환산저해농도 상수를 이용하면 쉽게 독성도를 파악 할 수 있었다. 토양 환경에 독성물질에 노출되었을 때, pH, 탈수소 효소 활성, 미생물 수, 미생물 군집구조는 독성 물질의 첨가량에 따라 직접적으로 영향 받는 것으로 나타났다. 독성물질의 첨가량과 pH 변화 사이에는 상관관계가 없는 것으로 나타났으며, 탈수소효소의 경우 MTBE는 논토양에, TBA는 논과 산림토양에, FA는 밭토양에 존재하는 미생물 탈수소효소에 민감하게 작용하는 것으로 나타났다. 또한 탈수소효소 활성 저해 관계지수 비교 시, 저농도(0~250 mg/L)로 MTBE, TBA, FA에 노출되었을 때 논토양은 다른 토양 조건과 달리 활성이 촉진되는 것으로 관찰되었다. 미생물 수의 경우 독성물질 첨가 농도가 증가 할수록 감소하였다. 순수 균주 조건과 마찬가지로 MTBE와 그의 대사산물 노출 시 독성 물질의 농도가 증가함에 따라 미생물 활성이 감소 하였으며, 논, 밭, 갯벌, 산림 토양 모두 TBA, MTBE, FA순으로 내성을 가지고 있었으며, FA에 대한 독성 강도가 센 것을 관찰 할 수 있었다. 또한 독성물질의 첨가량이 증가 될수록 이들 물질에 내성을 가지는 균주들이 발견되는 것이 관찰할 수 있었으며, 군집구조 분석 결과 독성물질 첨가량과 시간에 따라 확실히 구분 되는 것을 관찰 할 수 있었으나, Shannon-Wiener 지수를 이용하여 미생물 군집의 다양성과 균등도 분석 결과, MTBE, TBA, FA의 주입 농도가 높은 농도로 주입되었음에도 차이가 거의 없었으며 토양 미생물 군집에 적용시키는데 어려움이 있는 것으로 판단되었다. 따라서 본 연구는 국내에서 연구가 미흡한 이들 물질에 대한 잔류 독성 영향에 대한 독성도 평가를 종합적으로 예측하고 평가할 수 있는 방법을 실질적으로 활용 할 수 있도록 연구한 것으로 향후 위해성 평가를 하는데 있어 기초 자료로 활용될 수 있을 것으로 기대된다.;For this study, a reduced inhibition model was developed, using the characteristics of different specific growth rates of bacteria. Using the model, a study to test the possibility of qualitative and quantitative evaluations on the effects of the MTBE and its intermediates’ toxicity was conducted. In order to evaluate how MTBE and its intermediates affect the indigenous microorganism community in non-polluted soil environments, various biological characteristics were assessed through studies of physical and chemical factors, of dehydrogenase activities (DHA) and biomass in colony forming units (CFU), and of their correlations. Molecular biology technique, PCR-DGGE (Polymerase chain reaction - Denaturing gradient gel electrophoresis) was used to observe the structure and the changes in the microorganism community, and Shannon Index (richness) and the evenness index were utilized to study the variety of species in the microorganism community. When soil was exposed to MTBE and its intermediates, the growth rate patterns of Escherichia coli, Pseudomonas aeruginosa, and Bacillus amylo-liquefaci were changed. Higher levels of exposure to toxic substance produced more significant inhibition of their growth rates. Under FA exposure conditions, all three strains had stagnation periods, and then showed exponential growths. It was also observed that their response speed to toxic substance changed respond to the concentration of the substrate (TOC: 24,500 mg/L of LB medium 100%~5%). After the growth characteristics study of the germs, their inhibition constant (KI) was obtained using the specific growth rate of each germ, and the reduced inhibition constant (αi) and reduced inhibition concentration were introduced. With E.coli, competitive inhibition and non-competitive inhibition model methods were all applicable. When the non-competitive inhibition model was applied, the inhibition constant K’I, which contained the meaning of μmax,app, was applied rather than KI. Both cases with and without the reduced inhibition constant, EC50 was 75, 150, and 0.8 mM for MTBE, TBA, and FA, respectively. With P.aeruginosa, its tolerance for MTBE and TBA was found to be comparatively strong. When it was exposed to FA, it had a long stagnation period (7hours with 1.5mM of FA) and then it achieved its maximum cell concentration. It was also observed that study with P.aeruginosa suited a mixed type model, as its change showed the same pattern as the competitive inhibition model when the substrate concentration was high, and it fit on the non-competitive inhibition model when the substrate concentration became low. With Bacillus amylo-liquefaci, the inhibition constant K’I, rather than KI, was applied. EC50 of each substance were 250, 100, 1 mM for MTBE, TBA, FA, respectively, as predicted. With B.amylo-liquefaci, it showed strong tolerance to MTBE and its intermediates in comparison to E.coli and P.aeruginosa. Under exposure to FA, the community with B.amylo-liquefaci showed a stagnation period followed by an exponential growth. Under exposure to MTBE, it was consistent with the competitive inhibition model, but as the substrate concentration level dropped, it was consistent with the non-competitive inhibition model. Under exposure to TBA or FA, it followed the non-competitive inhibition model, and in all these situations, the inhibition constant K’I, rather than KI, was applied. When the reduced inhibition constant was applied, EC50 was 200, 400, and 0.8 mM for MTBE, TBA, and FA, respectively. The tolerance of the three strains for MTBE and its intermediates were in the order of TBA, MTBE, and FA, and it was observed that the toxic intensity of FA was the strongest. In the case of mixed systems containing two or three of the MTBE, TBA, and FA, it was found that growth inhibition occurred even in a small concentration range and the results were roughly in agreement with the standards of the basic substances despite of different concentration conditions. Through this it was concluded that when the specific growth rate was discovered, toxicity level could easily be identified using reduced inhibition concentration constant with chosen basic substance. In this study, it was observed that PH, dehydrogenase activation, number of microorganisms, and microorganism community were directly affected by the amount of added toxic substance in soil environment. No correlation was found between the volume of toxic substance and pH changes. In the case of dehydrogenase activity, it was found that MTBE significantly affected the dehydrogenase activities of microorganisms in rice paddy soil, while TBA significantly affected those in rice paddy and forest soils, and FA significantly affected those in farm field soil. Through comparison of indices related to the inhibition of dehydrogenase activities, accelerated activity in rice paddy soil was found in contrast to those in other soils, when the soil was exposed to MTBE, TBA, and FA occurred in low concentrations (0~250 mg/L). As for the number of microorganisms, decrease was observed when the concentration toxic level was increased. Like pure strains, the activity of microorganisms was decreased as the level of MTBE and its intermediates was increased. It was observed that the tolerance level to TBA, MTBE, and FA was stronger in order of rice rice field, leek patch, tidal mud flat, and forest soils and all of them had strongest tolerance level to the toxic intensity of FA. It was also observed that as the volume of added toxic substance increased, more strains with tolerance were found. The analysis of the community structure showed that such strains became more noticeable as the concentration of added toxic substance and exposed time were increased. The richness and evenness of the community decreased as the toxic level was increased. This study examined the comprehensive method of predicting and evaluating the toxicity assessment on the effect of remaining toxic substance to create the foundation of basic data for the future risk evaluations on toxic exposure.-
