Abstract
파골 세포는 조혈모세포에서 파생한 단핵구 및 대식세포계열의 다핵세포로 조골세포와 기질세포로부터 분비되는 RANKL, M-CSF에 의해 활성이 유도된다. 파골 세포는 뼈의 파괴를 담당하여, 뼈의 합성을 유도하는 조골세포와 더불어 뼈 리모델링의 주축을 이룬다. 본 연구에서는 이러한 파골 세포의 뼈 파괴 기능 외에 골수에서의 조혈모세포 가동화에 미치는 중요한 역할을 연구하였다. 스트레스 상황의 신호반응에 의해 활성화된 파골 세포는 단백질 분해효소인 MMP-9과 CTK를 다량으로 분비함을 확인하였다. 또한 이러한 효소들이 함유된 활성화 상태의 파골 세포 배양액으로 조골세포와 골수세포를 각각 배양한 결과 조골세포의 표면에 발현하는 SDF-1α이 유의하게 감소되었고 골수세포의 표면 CXCR4는 증가하였다. 또한 CCl4를 투여하여 간 손상을 유도한 마우스와 파골 세포의 분화 유도제인 RANKL을 투여한 마우스는 정상 마우스보다 말초혈로 가동화된 골수전구세포가 유의하게 증가하였으며 CCl4와 RANKL을 함께 투여한 경우 가장 큰 가동률을 보였다. 이러한 결과는 활성화된 파골 세포가 골수 내 조혈모세포 미세환경의 주요한 구성원인 조골세포의 표면 SDF-1α의 발현을 감소시켜 조골세포와 조혈모세포간의 부착력을 약화시키고, 더불어 조혈모세포 표면의 CXCR4를 증가시켜 말초혈로의 가동을 촉진한다는 것을 시사한다.;Osteoclasts (OCLs) are multinucleated cells that derived from the monocyte/macrophage hematopoietic lineage in response to receptor activator of NF-κB ligand (RANKL). They are specialized cells responsible for physiological bone resorption and as well as pathologic bone loss, playing a central role in the regulation of bone mass with bone forming cells, osteoblasts. In addition to unique ability to resorb bone, OCLs also play potential role in the mobilization of hematopoietic progenitor cells from bone marrow. Various stress stimuli (e.g. hypoxia, injury and inflammation) induced activation of OCLs along the stem cell-rich endosteum region of bone, secretion of proteolytic enzymes and mobilization of progenitors. Here we investigated effect of activated OCLs on the stem niche whether lead to mobilization of hematopoietic progenitors in vitro. As a result, RANKL-stimulated RAW 264.7 mouse monocyte/macrophage cell line cleaved SDF-1α on the osteoblast and increased CXCR4 expression on the bone marrow cell (BMC), thus it might be involved in alteration of stem cell niche components and mobilization of hematopoietic progenitor cells.
