Abstract
본 연구에서는 고지방식이를 먹인 쥐에서 capsaicin이 비만 억제에 효과를 나타내는지 알아보기 위하여 실시되었다. 45%의 고지방식이에 각각 0% (대조군), 0.01%, 0.02% 용량의 capsaicin이 첨가하여 8주간에 걸쳐 실험동물에게 공급한 뒤 식이 섭취량과 체중, 식이 효율 등을 조사하였고, 부고환 지방과 신장주변 지방의 무게를 측정하였다. Capsaicin이 지질대사에 미치는 영향을 알아보기 위하여 혈청 내의 중성지방, 총 콜레스테롤, HDL-콜레스테롤 농도가 분석되었으며, 간의 총 지질, 중성지방, 총 콜레스테롤, HDL-콜레스테롤 농도가 분석되었다. 또한 분변으로 배설된 총 지질, 중성지방, 총 콜레스테롤 농도가 분석되었다. 체내 에너지 대사의 대표적인 조절인자이며 열발생(thermogenesis)에 중요한 역할을 하는 UCP2의 유전자 발현에 capsaicin이 미치는 영향을 미치는지 알아보기 위하여 real time PCR을 통해 부고환 지방 조직 내의 UCP2의 mRNA 발현 정도가 정량 되었다. 8주간의 실험기간 동안 각 그룹별 식이섭취량을 살펴본 결과 0.01% capsaicin 첨가 군의 에너지 섭취량은 대조군의 에너지 섭취량과 유의적인 차이를 보이지 나타내지 않았고 0.02% capsaicin 첨가군의 에너지 섭취량은 대조군에 비해 유의적으로 감소한 것으로 나타났다. 그러나 에너지 섭취효율은 capsaicin 첨가군들과 대조군간에 유의적인 차이를 나타내지 않았다. 실험기간 종료 후 동물들의 체중을 조사한 결과 대조군에 비해 capsaicin을 고지방식이와 함께 먹인 그룹의 체중이 용량 의존적인 방법으로 감소한 것으로 나타났으며 이 중 식이의 0.02% capsaicin을 공급한 그룹의 체중은 대조군에 비해 유의적으로 작은 것으로 나타났다. 또한 0.02% capsaicin 첨가군은 대조군에 비해 부고환지방 무게가 유의적으로 작았으며 신장주변 지방 무게도 감소하였으나 통계적으로 유의한 차이를 나타내지는 않았다. 혈중 중성지방을 분석한 결과 대조군에 비해 0.01% capsaicin 첨가군은 25%, 0.02% capsaicin 첨가군은 29% 감소한 것으로 나타났다. UCP2의 유전자 발현의 상대적 정량 결과는 대조군에 비해 0.02% capsaicin 첨가군은 2.22배 유의적으로 증가한 것으로 나타났다. 위의 결과들을 종합하여 볼 때, 식이로 섭취한 capsaicin은 고지방식이로 유도되는 체내 지방 축적 및 혈중 중성지방을 억제시켜 비만을 개선시킬 수 있으며, 이러한 capsaicin의 항비만 효과에 UCP2의 유전자 발현 증가가 일부 관여하는 것으로 생각된다.;Capsaicin is a pungent agent of red peppers, which is widely consumed as food adjuncts in Korea and other countries. The present study was performed to investigate anti-obesity effects of capsaicin via increase in mRNA level of UCP2. Male Sprague-Dawley rats (n=21) were fed with 45% high-fat diet (control), high-fat diets containing 0.01% or 0.02% capsaicin for 8 weeks. The final body weight and the body weight gain were significantly lower in 0.02% capsaicin treated diet group (p<0.05) compared to control. Epididymal adipose tissue mass in the group supplemented with 0.02% capsaicin was significantly lower than that of control (p<0.05). The diets with 0.01% or 0.02% capsaicin significantly decreased the level of serum triglyceride (p<0.05) by 25% or 29%, respectively. However, there was no significant difference in the level of serum cholesterol among the groups. Also, capsaicin supplementation didn’t affect the levels of hepatic and fecal cholesterol. Result from real-time PCR showed that the expression level of UCP2 gene in rats supplemented with 0.02% capsaicin was 2.22-fold (p<0.05) of the level expressed in control. In conclusion, dietary capsaicin reduced adipose tissue mass and serum triglyceride in rats fed with high-fat diet. These effects were supposed to be at least partially mediated by up-regulation of UCP2 gene expression which is related to thermogenesis.
