Abstract
과거 단순히 세포막의 구성성분으로만 인식되어왔던 스핑고지질은 생체기능을 조절하는 생체내 활성분자이며 세포사멸과 세포생존의 신호전달 물질로서 최근 그 생리활성 기능이 연구 되어 지고있다. 스핑고신의 신규합성은 serine에 palmitoryl기가 붙으면서 시작되는데, 이 과정에서 스핑고신이 스핑가닌이되고 디하이드로세라마이드가 합성된다. 여기에 스핑고신 구조의 4,5번 탄소 사이에 이중결합이 형성되면서 세라마이드의 합성이 일어난다. 이 세라마이드가 다시 거꾸로 스핑고신으로 생성이 되고 이 스핑고신에 인산기가 붙으면 스핑고신-1인산 (S1P) 이 된다. 이렇게 생성된 스핑고신 1-인산은 세포막 수용체를 가지는 세포간 신호전달 물질에 관여하는 커다란 메신저로 그 역할을 담당하고 있다. 스핑고신에 인산이 붙어 있는 구조인 스핑고신 1-인산은 세라마이드와 스핑고신과는 다르게 세포보호작용을 가지고 있으며, 세포생존인자로 인식된다. 이후 세포막에 G단백질에 커플링하는 S1P수용체가 S1P에 대해 특이성을 갖는 세포막수용체로 보고되었다. 지금까지 5종류의 S1P 수용체가 발견되었으며, 각각의 수용체를 통한 다양한 기능에 대한 연구와 그 선택적인 작용에 대해서도 꾸준하게 연구 되고 있다. 이처럼 스핑고지질에 대한 연구가 활발하게 진행되고 있으나 그 중에서 스핑고지질의 대사체 중의 하나인 파이토세라마이드와 파이토스핑고신에 대해서는 세포 생존 또는 세포보호작용에 대해서는 아직 정확하게 보고 되어진 것이 없다. 따라서 이를 타겟으로 알츠하이머질병에 원인이 되는 다양한 인자들인 산화적스트레스와 신경세포독성, 염증, 베타 아밀로이드 생성 등에서 그 효과가 나타나는지를 세포와 동물모델에서 실험을 진행해 보게 되었다. 신경세포의 활성화는 알츠하이머와 같은 퇴행성신경질환의 진행에 있어서 중요한 역할을 하게 된다. 따라서 그 활성을 억제하는 것이 퇴행성 신경질환의 치료에 중요하다고 사료된다. 본 연구에서는 L-glutamate, KCl, β-amyloid 에 의해 활성화된 cortical cell이 두 가지의 약물 중에서 파이토세라마이드에 의해서 세포독성유발을 억제하는 것을 확인하였고, scopolamine에 의해 기억력기능을 저하시킨 동물모델에서 Passive avoidance test, Y-maze, Morris water maze를 통한 행동능력실험을 통해서 그 효과를 다시 한번 확인 할 수 있었다. 이후 더 나아가β-amyloid를 임의적으로 주입한 알츠하이머질환 동물모델에서도 passive avoidance test를 통해 기억력 감소에 파이토세라마이드가 긍정적인 효과를 나타낸다는 것을 다시 한번 확인하였다. 그 기전을 규명하기 위한 mRNA 레벨 테스트에서 약물과 S1P 수용체가 관계가 있는지를 PCR을 통해 알아본 결과 어느정도 관계가 있음을 확인할 수 있었다. 또한 염증반응을 조절하는데 관여하는 것으로 알려진 사이토카인인 TNF-α 와 IL-6, IL-1β의 발현을 감소시키는 효과를 볼 수 있었다. 결론적으로, 본 연구에서는 파이토세라마이드가 다양한 인자에 의해 활성화된 신경세포에서의 세포독성유발을 감소시킨다는 것과 항염증 작용도 함께 나타낸다는 것을 확인하였고, 여기에 S1P 수용체가 관련 되어 있음을 확인하였다. 아직 파이토세라마이드와 파이토스핑고신이 다른 실험결과를 나타내는 이유와 그 기전을 구체적으로 확인하지는 못했지만 이러한 연구결과는 스핑고지질의 유도체인 파이토세라마이드가 퇴행성 뇌신경질환의 잠재적인 치료제로 사용될 수 있음을 보여준다.;Sphingolipids contain an important group of biomolecules, some of which have been shown to play pivotal roles as messenger in life and death of cells on the central nervous system. Especially, sphingosine 1- phosphate (S1P) is an influential biomediator that can act as either an intracellular or an intercellular messenger. In the central nervous system, the specific membrane receptors for S1P have been identified in various brain regions and are involved in the regulation of multiple biological processes such as cell growth, survival, angiogenesis, migration, differentiation and immune function both in neurons and glial cells. Extracellular actions of S1P are mediated with a family of five specific G-protein-coupled receptors, S1P1-S1P5. The biosynthetic pathway and biological functions of sphingosine metabolites are studied but the role of sphingolipid metabolites has been focused mainly in non-neuronal tissues. Furthermore the study of phytoceramide (Pcer) and phytosphingosine (Pso) which are metabolites from plant sphingolipid are unknown. The pathophysiological events involved in the neuronal dysfunction and degeneration in AD are considered to be the increased oxidative stress and the production of neurotoxic forms of amyloid-β (Aβ) peptide. Aβ peptide has been shown to have the potential to induce the oxidative stress and the inflammation, which consider play important roles in the pathogenesis of AD. And also the scopolamine-induced amnesic animal models have been used to screen for potential treatments for cognitive dysfunction. This study was aimed at investigating the possible effects of Pcer and Pso on neuronal cell and neurodegenerative disease. First, Pcer and Pso on memory functions were measured and its biological effects on neurotoxicity in hippocampal and cortical neurons. This present study suggests that Pcer contribute about memory recovery and enhancing with inhibitory action on neurotoxicity. Pcer was developed for the prevention of neurodegenerative disorder such as Alzheimer’s disease and vascular dementia.
