- 1 장. 항암효과가 기대되는 새로운 apio-carbocyclic 뉴클레오사이드 유도체의 비대칭적 합성 neplanocin A와 apio-dideoxyadenosine (apio-ddA)의 생리활성을 바탕으로 이들의 특성을 결합시킨 이상적인 apio-neplanocin A 유도체 (5-12)를 입체 선택적으로 합성하였다.
최종 뉴클레오사이드 (5-12)의 apio 부분은 lactol인 17번 물질에 potassium carbonate와 37% formaldehyde를 가하여 입체 선택적으로 합성하였다. 또한, Neplanocon A의 carbasugar 부분은 diene인 19번 물질을 methylene chloride에 녹인후 Grubbs’ catalyst를 가하여 합성할 수 있었다. 최종 물질인 5-12번 뉴클레오사이드는 glycosyl donor인 21번 물질을 Mitsunobu 상태 하에서 핵산 염기와 함께 축합하였다.
이와 같은 방법으로 합성한 중요 중간체(20)를 가지고 주요 합성 단계로 각각 catalytic hydrogenation과 Simmons-Simith cyclopropanation을 통해 apio-aristeromycin (13)과 (N)-apio-methanocarbaadenosine (14)를 합성하였다.
합성된 5, 13, 14번 물질로 S-adenosylhomocysteine hydrolase (SAH) 효소에 대한 저해 활성을 측정해 보았지만 측정치가 100 μM 이상으로 나타나 유의할만한 효소 저해 활성을 나타내지 않았다. 이는 작용 기전상 cofactor-bound NAD+ 에 의해 산화될 C3′ 위치에 secondary hydroxyl group 이 없어서 효소 저해 활성이 나타나지 않은 것으로 사료된다. 그러나 apio-neplanocin A (5)는 A1 and A2A 아데노신 수용체에서는 결합 친화력을 나타내지 않고 선택적으로 A3 아데노신 수용체에만 강한 결합력 (Ki = 628 ± 69 nM)을 나타내었다.
이 장에서는 입체선택적인 hydroxymethylation과 RCM 반응을 통해 apio-carbasugar를 가진 이상적인 carbocyclic 뉴클레오사이드를 최초로 합성하였다. 또한 human A3 아데노신 수용체의 효현제로 작용하는 새로운 모형을 발견하였다. 이를 통해 A3 아데노신 수용체의 새로운 효현제의 개발과 수용체 결합 부위를 알아내는 데 큰 도움이 될 것이다.
2 장. C형 간염 바이러스에 대한 항바이러스 효과가 기대되는 2′-C-methyl-cyclopropyl-fused carba-nucleoside의 입체 선택적 합성 C 형 간염은 치명적이면서도 아직 효과적인 치료제가 개발되지 않았으며 항 C형 간염 바이러스제로 많은 뉴클레오사이드와 비 뉴클레오사이드 유도체가 합성되었다. 이들 가운데 2′-C-methyladenosine (2)과 2′-C-methylguanosine (3)이 cell-based HCV replicon assay에서 강력한 C형 간염 바이러스 저해제 (각각 EC50 = 0.26과 3.5 μM)로 알려져 있다. 이들 2번과 3번 화합물은 입체적으로 northern C3′-endo의 입체배치를 한다고 알려져 있다.
이러한 물질들의 입체학적 자료를 바탕으로 cyclopropyl-fused carbanucleoside 유도체 4-8번 화합물을 합성하였다. 이 장에서 합성한 cyclopropyl-fused nucleoside 화합물들은 2번과 3번 화합물과 유사한 northern C3′-endo의 입체배치를 나타내기 때문이다. 2′-C-methyl-cyclopropyl-fused carbanucleoside는 주요 반응 단계로 입체 선택적 cyclopropanation, isopropylidene 기의 선택적 탈리, 입체 선택적 Grignard reaction 그리고 cyclic sulfate chemistry를 사용하여 합성하였다.
합성된 화합물들은 C형 간염 바이러스에 대해 항 바이러스 활성을 나타내지 않았지만 이 장에서 시도된 모든 합성은 유기 화학자와 의약 화학자들이 새로운 carbasugar를 연구하는데 크게 기여할 것이다.;Part I. Asymmetric Synthesis of Novel Apio Carbocyclic Nucleoside Analogues as Potential Antitumor Agents
On the basis of the biological activity of neplanocin A and apio-dideoxyadenosine (apio-ddA), novel apio-neplanocin A analogues 5-12, combining the properties of two nucleosides, were stereoselectively synthesized. The apio moiety of the target nucleosides 5-12 was stereoselectively introduced by treating lactol 17 with 37% formaldehyde in the presence of potassium carbonate. The carbasugar moiety of neplanocin A was successively built by exposing diene 19 on a Grubbs’ catalyst in methylene chloride. The final nucleosides 5-12 were synthesized from the condensation of the glycosyl donor 21 with nucleic bases under the standard Mitsunobu conditions. Similarly, apio-aristeromycin 13 and (N)-apio-methanocarbaadenosine 14 were derived from the common intermediate 20 using catalytic hydrogenation and Simmons-Simith cyclopropanation as key steps. The synthesized compounds 5, 13, 14 were subjected to enzyme inhibition assay. However they did not show any significant inhibitory activity against S-adenosylhomocysteine hydrolase (SAH) up to 100 μM, maybe due to the absence of the secondary hydroxyl group at the C3′-position, which should be oxidized by cofactor-bound NAD+. However, apio-neplanocin A (5) showed potent and highly selective biding affinity (Ki = 628 ± 69 nM) at the A3 adenosine receptor without any binding affinity at the A1 and A2A adenosine receptors.
In conclusion, novel carbocyclic nucleosides with unnatural apio-carbasugars were synthesized first time using stereoselective hydroxymethylation and RCM reaction as key steps. A new template of human A3 adenosine receptor agonist, which plays a great role in developing new A3 adenosine receptor agonist as well as in identifying the binding site of the receptor was also discovered.
Part II. Stereoselective Synthesis of 2′-C-Methyl-cyclopropyl-Fused Carbanucleosides as Potential Anti-HCV Agents
2′-C-Methyladenosine (2) and 2′-C-methylguanosine (3) were discovered as potent HCV inhibitors (EC50 = 0.26 and 3.5 μM, respectively) in a cell-based HCV replicon assay. Compounds 2 and 3 were reported to display a strong preference for a Northern C3′-endo conformation. On the basis of this conformational information, the cyclopropyl-fused analogues 4-8 were designed because these compounds could take the Northern C3′-endo conformation similar to that for compounds 2 and 3. Stereoselective synthesis of 2′-C-methyl-cyclopropyl-fused carbanucleosides was accomplished via stereoselective cyclopropanation, regioselective cleavage of the isopropylidene group, stereoselective Grignard reaction, and cyclic sulfate chemistry. Although potent anti-HCV compounds were not discovered, all chemistries employed in this study will greatly contribute to the development of new carbasugar templates by organic and medicinal chemists.
