Abstract
금속 이중층 수산화물 (LDH)는 다양한 유기/무기 음이온들을 담지 할 수 있어 음이온 교환체, 열안정제, 약물 전달체, 촉매 등으로 널리 이용되고 있다. 따라서 본 연구에서는 새로운 금속 이중층 수산화물 (LDH)를 합성하여 그 물리적, 화학적 성질을 평가하고, 생체-나노 융합 기술을 통해 약물 전달 기능을 갖는 나노 혼성체를 형성하여 생체 상호작용들을 평가하였다. 본 연구의 제 1장에서는 알루미늄 (Al³^(+))이 일부 안티몬 (Sb³^(+))으로 치환된 새로운 금속 이중층 수산화물을 합성하였다. 수산화 안티몬 (Sb(OH)₃)은 열역학적으로 더 안정한 형태인 산화 안티몬 (Sb₂O₃)의 형성으로 고체상뿐만 아니라 수용액상에서도 보고된 바가 없다. 이러한 이유로 안티몬이 들어있는 금속 이중층 수산화물은 아직 보고된 바가 없다. 하지만 특정 합성조건에서 알루미늄의 6배위 자리에 안티몬을 약 10%까지 부분적으로 치환한 새로운 금속 이중층 수산화물을 합성하였다. X-선 회절 분석 및 에너지 분산 X-선 분광 분석을 통해 안티몬이 LDH의 격자 내에 안정적으로 화학적 결합을 이루었음을 확인하였다. 특히, 격자 상수를 통해 LDH의 이방성을 확인하고 (c ≫ a) 안티몬의 inert pair 효과를 증명하였다. 이렇게 합성된 안티몬 이중층 수산화물은 안티몬이 시너지 효과를 낼 수 있는 할로겐이 함유된 고분자와의 혼합을 통해 그 효과를 극대화 시킬 수 있다. 안티몬 이중층 수산화물과 PVC의 나노 혼합체를 형성하여 난연성을 평가한 결과 기존의 물질보다 그 특성이 향상되었음을 알 수 있었다. 제 2장에서는 항암제의 일종인 메소트렉세이트 (Methotrexate, MTX)를 공침반응을 통해 항암제/나노 캡슐 혼성체를 합성하고, 항암치료제로써의 활용 가능성을 평가하였다. X-선 회절 분석과 적외선 분광 분석을 통해 folate 계열의 약물인 MTX가 정전기적 상호작용을 통해 화학적 활성기를 유지하면서 금속 이중층 수산화물 층간에 삽입되어 있음을 확인하였다. 다음으로 동물실험을 통해MTX-LDH 나노 혼성체의 유효성을 확인하였다. 쥐의 면역반응 실험을 통해 나노 혼성체는 면역체계에 크게 영향을 주지 않는다는 것을 알 수 있었으며, 쥐의 독성실험을 통해 나노 혼성체의 복강주사 최적 농도는 60 mg/kg으로 결정하였다. 이와 같은 결과를 바탕으로 mouse의 C33A orthotopic model에서MTX-LDH의 유효성 및 암 선택성을 확인하였다. 연구 결과에 따르면, MTX만 주사 한 집단에 비해, MTX-LDH 를 주사할 경우 근육이나 혈액에 비해서 종양 조직에 고농도의 MTX가 유지됨을 알 수 있었다. 제 3장에서는 암세포 내에 높은 붕소농도를 필요로 하는 붕소 중성자 포획 치료에 매우 효과적으로 사용될 수 있는 나노 혼성체를 합성하고 그 유효성을 평가하였다. 공침방법을 통해sodium mercaptoundecahydro-closo-dodecaborate (BSH)가 금속 이중층 수산화물에 성공적으로 캡슐화되었으며, 이는 X-선 회절분석을 통해 확인하였다. 또한 실제 동물모델에서 나노 혼성체를 주사하고 붕소의 생체분포를 확인한 결과, 근육이나 혈액에 비해서 종양 조직에 상대적으로 매우 높은 붕소 농도가 유지됨을 알 수 있었다. 따라서 나노 혼성체를 약물전달시스템에 응용하면, 암세포에 대한 선택성이 현저히 증가되어 정상 세포에 대한 세포 독성을 현저히 감소시키면서 암 치료에 적은 양으로도 효과적으로 이용될 수 있다는 것을 알 수 있었다.;The nanohybrid exhibits extraordinarily high synergetic and complementary behavior. In particular, the intercalation compound of layered double hydroxide (LDH) offers new synergetic properties. Under various synthetic methods and conditions, various kinds of inorganic or organic anions (A^(n-)) can introduced into the interlayer space of the LDHs. Due to this property, LDHs have a wide range of applications such as anion exchangers, thermal stabilizers, drug or biofunctional molecule delivery vectors, absorbents, and ionic conductor catalysts. Therefore, the aim of this study is to synthesize new LDH and to determine the crystal structure and chemical bonding nature of LDHs and to develop drug delivery system for anticancer chemotherapy. In first study, to obtain improved flame-retardancy of LDH, an attempt has been made to prepare new Sb³^(+)-containing Mg-Al LDH by substituting the Al³^(+) in octahedral site partially with Sb³^(+) upto approximately 10%. To dates, no antimony hydroxide has been reported not only in solid state but also in aqueous solution, surely due to the fact that the formation of antimony oxide, Sb₂O₃, is thermodynamically more favorable than that of the hydroxide phase, Sb(OH)₃. From the XRD analysis, we found that the lattice constants (a = 3.075 Å, c = 23.788 Å) of the pristine, Mg-Al LDH, increased gradually upto those of a = 3.087 Å, c = 24.167 Å, when the Sb substitution rate reached to 8%. However, the lattice constants remained unchanged along with the formation of Sb₂O₃ as an impurity if the substitution rate exceeded more than 10%. It is, therefore, concluded that the solubility limit of Sb³^(+) in LDH would be around 10% under the present synthetic condition (25 ℃, 1 atm, pH 12). In addition, we were able to determine the chemical formula of Sb-substituted LDHs as Mg₄Al₁-_(x)Sb_(x)OH_(10)(CO₃)_(1/2).H₂O (x = 0~0.08) on the basis of energy dispersive X-ray spectroscopy analysis. Also, in our present work, we studied the flame-retardant behavior of Sb-substituted LDHs in the PVC, and compared the flame-retardant efficiency with that of Mg-Al LDH. From the thermal degradation results, we confirmed that the Sb-substituted LDH has an enhanced flame retarding effect and thermal stability. In second study, an anticancer drug, methotrexate (MTX), have been successfully encapsulated into LDH through conventional co-precipitation method to facilitate drug transport into tumor cell. From the powder X-ray diffraction patterns and spectroscopy analysis, MTX molecules intercalated with monolayer structure maintaining their functional groups. According to the immune test, it was confirmed that MTX-LDH hybrid has hardly any toxicity in mice. We also investigated the tumor selectivity of MTX-LDH through bio-distribution test in C33A orthotopic model. From the results, the accumulated MTX at tumor was considerably higher in MTX-LDH treated mice than in free MTX treated mice. Furthermore, MTX-LDH had better tumor selectivity than free MTX. Therefore, MTX-LDH nanohybrid has enhanced drug efficacy. In addition, sodium mercaptoundecahydro-closo-dodecaborate (BSH, Na2B12H11SH) was successfully intercalated into LDHs eventually for efficient boron neutron capture therapy (BNCT). To enhance the tumor selectivity, we have attempted to modify the LDH surface with folic acid which is the cancer cell specific ligand. According to the powder X-ray diffraction patterns and spectroscopy analysis, it was confirmed that BSH molecules are stabilized in the interlayer of LDHs by strong hydrogen bonding, maintaining their chemical properties. From the bio-distribution results in xenografted nude mice, surface modified BSH-LDH had better tumor selectivity than free BSH. Therefore, BSH-LDH nanohybrid would be the most efficient in delivering rich boron to tumor cells.
