dc.description.abstractLife span has been increased worldwide due to the improvement of living standard and the advancement in medicine. Consequently, there is a great increase ofthe elderly population. The fast movement into an aging society is expected to entail lots of social problems, including a rapid increase in the number of patients with chronic diseases. We have been interested in the synthesis, characterization, and the behaviorof newly designed dendrimers to search for the uniquely functioning biomolecular beacons. We structurally designed functional dendrimers similar to Amyloid oligomers which are proposed to be responsible for the Alzheimer’s disease. One of the chronic diseases of elderly people is Alzheimer`s disease. We first began our research, starting from commercially available adamantane to synthesize desired tetra-substituted cores of dendrimers. We completed the synthesis of 1,3,5,7-tetrakis(aminomethyl)adamantane tetrahydrochloride of 48% yield, adamantane-1,3,5,7-tetracarboxylic acid of 51% yield, 1,3,5,7-Tetra(bromomethyl)adamantane of 13% yieldfrom adamantane. Adamantane-1,3,5,7-tetracarboxylic acid was served as a common tetra-substituted core of the 1st generation of adamantane-based dendrimer in our laboratories. The first generation dendrimer was successfully synthesized using HATU as coupling reagents and HOAt. In conclusion, the efficient synthesis of the first generation dendrimer was completed from adamantane as starting material in 5 steps and overall 13% yield.;우리나라는 비약적인 경제발전과 생활수준의 개선, 의학의 발달 등으로 평균수명이 연장되고 있으며 이에 따라 노인인구가 크게 증가하였다. 빠른 속도로 고령화 사회에 진입함에 따라 많은 사회문제가 생길 것으로 예상되며 그 중 하나가 만성질환자의 급증을 들 수 있다. 노인의 만성질환 중 하나인 알츠하이머 질환(Alzheimer’s Disease, AD)은 노년기에 발생하는 대표적인 질환이다. 현재 많은 나라에서 알츠하이머 환자의 치료제로 아세틸콜린 에스터라제 억제제를 사용하고 있다. 여기에는 Donepezil(Aricept), Galatamine(Reminyl), Rivastigmine (Exelon), Tacrine(Cognex) 등이 있다 그러나 이 치료제들의 효과는 경증에서 중증 정도의 알츠하이머 환자에게 효능이 있다. 그래서 생리적인 조건에서 분자 구조가 변함에 따라 형광 특성이 변하는 기능성 생물활성분자인 덴드리머를 합성하여 말단 작용기에 펩타이드를 연결한 펩타이드 덴드리머의 합성을 목표로 1세대 덴드리머를 합성하였다. 안정된 구조를 갖는 펩타이드 덴드리머는 구형의 아밀로이드 올리고머를 모방하는데 우수한 후보물질로 여겨지므로 AD연구에 이용 할 것이다. 덴드리머 합성을 위해 먼저 adamantane을 출발물질로 분자내 네 곳에 치환체가 있는 다양한 tetra substituted core의 형태를 가진 덴드리머의 중심부를 합성하였다. 그 중 tetra aminomethyl adamantane은 출발물질인 adamantane에 bromination과 cyanation을 시켜주어 네 곳 모두 하나의 새로운 탄소를 더 생성하였다. 그리고 마지막으로 reduction을 수행하여 총 48% 수율로 합성하였다. 또한, 동일한 방법으로 출발물질로 adamantane을 이용하여 bromination과 cyanation을 거친 후 생성된 탄소와 질소의 삼중 결합을 hydrolysis를 거쳐 tetra- carboxylic acid adamantane을 총 51% 수율로 합성하였다. 그리고 Tetra- carboxylic acid adamantane에 esterification, reduction, bromination을 진행시킨 결과 다른 친핵성 가지와 결합하기 좋은 leaving group인 tetra bromomethyl- adamantane을 출발물질인 adamantane으로부터 총 13% 수율로 합성할 수 있었다. 그리고 가지체로 tris(hydroxymethyl)-aminomethane를 출발물질로 cyanation, esterification시켜 62%의 수득률로 얻었다. 합성한 다양한 중심부 중 tetra carboxylic acid adamantane을 core로 선택하여 합성한 가지체와 coupling, hydrolysis하여 총 13% 수득률로 1세대 덴드리머를 합성할 수 있었다.-
