dc.description.abstractRehmannia glutinosa Libosch. ex Fischer & C. Meyer. (Family: Orobanchaceae) is a perennial plant, a traditional Chinese herb that used as medicinal substance. It has been reported that the roots of R. glutinosa has various pharmacological activities and has been used in oriental medicine for the treatment of hematic, tonic, and antipyretic. Although many studies have been conducted on the major compounds and activities of Rehmannia, there are not much reports related to structural analysis of other constituent compounds. In this study, the dried roots of R. glutinosa were extracted with MeOH, and the extract was fractionated in the order of n-Hexane, EtOAc, and n-BuOH. Two compounds, martynoside (20), and leucosceptoside A (21), which are phenylethanoid glycosides, were isolated from the EtOAc fraction by open column chromatography MPLC, HPLC and Sephadex. The structure of the compounds was analyzed using various physicochemical and spectroscopic techniques and confirmed by comparing them with existing literature of other plants. In addition, through in silico docking simulations, the possibility of whether these compounds could serve as PPAR activator in diseases related to lipid metabolism, was checked and also checked their potential as a tyrosinase inhibitor in related diseases including hyperpigmentation that may occur due to excessive melanin production. In previous studies, it was reported that the main component of R. glutinosa exhibits anticancer activity, so a cytotoxicity assays were requested for the extracts, but the results were not found in this study.;Rehmannia glutinosa Libosch. ex Fischer & C. Meyer. (열당과: Orobanchaceae)는 다년생 식물이고, 중국의 전통 허브로 약용으로 많이 사용된다. R. glutinosa 의 뿌리는 다양한 약리 활성 작용과 함께 한방에서 보혈제, 강장제, 해열제로써의 사용이 보고되어 있다. 지황의 주성분과 활성에 대한 많은 연구가 진행되었지만, 그에 비해 다른 구성 물질의 구조 분석과 관련된 보고는 많지 않다. 이번 연구에서는 지황 뿌리를 MeOH로 추출하고, 그 추출물을 n-Hexane, EtOAc, BuOH의 순서로 분획하였다. 그리고 EtOAc 분획물로 open column choromatography, MPLC, HPLC, Sephadex의 방법을 통해 phenylethanoid glycoside 종류인 martynoside (20), leucosceptoside A (21) 2개의 단일물질을 분리하였다. 화합물의 구조는 각종 이화학적 및 분광학적 기법을 사용하여 분석하였고, 기존의 문헌과 각각 비교하여 확인하였다. 또한 in silico docking simulation을 통하여 이 물질들이 지질대사 관련 질환에서 PPAR activator로서의 역할을 할 수 있을지를 확인하였다. 또한 과도한 멜라닌 생성으로 인해 발생할 수 있는 hyperpigmentation등 관련 질환에서 tyrosinase inhibitor로서의 잠재력을 확인하였다. 또한 과거 연구에서는 R. glutinosa의 주성분이 항암 활성을 보인다고 보고된 바 있었기에 상기 추출물들에 대해 세포독성 시험을 의뢰하였으나 결과는 아닌 것으로 나타났다.-
