Abstract
A non-thermal sterilization technique has been considered as an alternative to heat sterilization techniques. Intense pulsed light (IPL) is a nonthermal sterilization technique that can effectively decontaminate bacteria. It uses a pulse in a wide, intense spectrum in a 100-1100 nm wavelength range. In the broad spectrum of light, UV-C light in the 100-280 nm wavelength range plays an important role in inactivating bacteria. E. coli O157:H7 is an important foodborne pathogen to be controlled. It causes severe illnesses and has high lethality. E. coli ATCC 25922 was commonly used as the surrogate of E. coli O157:H7. There are several factors that affect IPL decontamination efficiency such as biological, operational and external factors. In this study, IPL treatment effects depending on biological factors, incubation time and population density of E. coli ATCC 25922 were evaluated. As a result, the inactivation rate decreased as the incubation time increased. This suggests that IPL treatment is more effective when it is performed immediately after contamination has happened. Also, there was a difference in decontamination effect depending on the growth phase. According to the 4D value that represents resistance to IPL, bacteria in the exponential phase weremore susceptible to IPL and bacteria in the stationary phase were the most resistant. When IPL was treated on different population densities, the inactivation effect was significantly reduced above 109 CFU/mL. This same inactivation tendency appeared when the distance between the lamp and sample was changed. In the case of 109, 1010 CFU/mL, additional treatments at 1200 V were performed. 109 CFU/mL showed 3.4 log reductions at 12.5 J/cm2 applied. However, 1010 CFU/mL decreased less than 1 log even though 18.7 J/cm2 was applied. These results were attributed to the transmittance of the bacterial suspension and shading effect. Transmittance of the bacterial suspensions decreased as the cell density rose. However, there was a big gap between the inactivation level of 1010 and 109 CFU/mL, even though their transmittance was similar. This difference might be due to cell distribution and aggregation. In the confocal images of the 109 and 1010 CFU/mL samples, the 1010 sample had more cell aggregation and denser cell distribution. With these results, it was seen that when a liquid sample is treated, IPL has limited efficiency depending on the bacterial population density.;비가열 살균 기술은 가열 살균의 대체 기술로 고려되어 왔다. 광펄스는 비가열 살균 기술 중 하나로 박테리아 제어에 효율적임이 알려져 있다. E. coli O157:H7은 식품에서 중요하게 통제되어야 하는 병원성 균 중 하나로 감염 시 심각한 식중독 증상을 유발하고 높은 치사율을 나타내는 균이다. E. coli ATCC 25922는 E. coli O157:H7를 대체하여 실험에서 자주 사용되는 균으로 본 연구에서는 E. coli ATCC 25922의 배양 시간과 초기 균수에 따른 IPL 처리 효과를 알아보고자 하였다. 처리 결과, 배양 시간이 증가할수록 살균 속도가 감소하는 것으로 나타났다. 이를 통해 오염이 발생한 직후 처리를 할 때 IPL 효율이 높을 것임을 예상할 수 있었다. 또한 생장 단계에 따라 살균 효과에 차이가 있었다. IPL에 대한 저항성을 나타내는 4D 값을 비교했을 때, 그 값이 exponential phase (3, 4시간 배양)의 균이 0.54 J/cm2, stationary phase (11시간 배양)의 균이 0.68 J/cm2였다. 이를 통해 exponential phase의 균이 가장 민감하며 stationary phase의 균이 가장 저항성이 높은 것을 알 수 있었다. IPL을 다양한 초기 균수를 가진 샘플에 대해 처리했을 때, 104-108 CFU/mL에서는 높은 에너지의 빛을 처리할수록 균수가 더 많이 감소하는 경향이 나타났지만 109 CFU/mL 이상에서는 살균 효과가 현저하게 감소하였다. 광펄스 살균 효율에 영향을 주는 요소 중 하나인 램프와 샘플 사이의 거리가 변경되었을 때에도 이와 같은 경향이 나타났다. 109, 1010 CFU/mL의 경우, 1200 V로 전압을 조절하여 추가 실험을 진행했다. 그 결과, 109 CFU/mL에서는 12.5 J/cm2을 처리했을 때, 3.4 log 감소했지만 1010 CFU/mL는 18.7J/cm2를 처리해도 1 log 미만으로 감소한 것을 확인했다. 이러한 사멸율 차이의 원인을 알아보고자 균액의 투과도를 측정하였다. 균액의 균수가 높을수록 균액의 투과율은 감소했다. 다만 1010과 109 CFU/mL의 투과도는 차이가 근소한데에 비해, 살균 정도에는 차이가 컸다. 공초점 현미경 관찰 결과, 1010 및 109 CFU/mL 샘플의 공초점 현미경 사진에서 1010 CFU/mL 샘플은 세포의 분포 밀도가 더 높고 응집된 균이 더 많은 것을 확인했다. 따라서 1010과 109 CFU/mL에서의 살균 효과 차이는 shading effect와 관련 있음을 알 수 있었다. 이러한 결과를 통해 IPL 처리시 미생물의 생리적 상태가 살균 효율에 영향을 주며 액체 상태의 샘플의 경우, 초기 균수에 따른 빛의 투과도와 shading effect를 함께 고려해야 한다는 결론을 얻을 수 있었다. 따라서 실제 산업 현장에서 IPL을 적용할 시에는 효율을 증진시키기 위해 배양 시간과 초기 균수와 같은 생물학적 요인들이 고려되어야 한다.
