- Hypercholesterolemia characterized by an increase in plasma low density lipoprotein (LDL) cholesterol and total cholesterol (TC) is the leading cause of non-alcoholic fatty liver disease (NAFLD), the most common liver disease worldwide. The present thesis evaluated the effects of fermented tea (FGT) extract on high cholesterol diet (HCD)-induced hypercholesterolemia and liver injury in hamster, an animal having similar lipid metabolism of human. Ten-week-old golden syrian hamsters were given normal diet (ND) or HCD containing 0.2% cholesterol and 10% lard. FGT (200 and 500 mg/kg/d) was orally administrated from the initiation of HCD for 12 weeks. FGT did not affect body weight, food intake, and calorie intake, but effectively inhibited HCD-induced increases in plasma LDL, TC, and triglyceride (TG). HDL was also significantly increased by HCD, but there was no additional increase by FGT administration. FGT also inhibited HCD-induced liver oxidative stress, fibrosis, and lipid accumulation. Interestingly, FGT significantly increased liver protein level of cholesterol 7 alpha-hydroxylase (CYP7A1), the rate-limiting enzyme for bile acid synthesis, together with increase in fecal excretion of TC. Taken together, the present data suggest that FGT may become a valuable health functional food preventing diet-induced hypercholesterolemia and NAFLD possibly through increasing fecal cholesterol excretion. ;혈장 저밀도지단백(low density lipoprotein: LDL) 콜레스테롤과 총콜레스테롤(total cholesterol: TC)의 증가를 특징으로 하는 고콜레스테롤혈증은 비알코올성지방간질환(non-alcoholic fatty liver disease: NAFLD)의 주 원인이다. 본 논문에서는 인간과 유사한 지질대사를 가진 햄스터에서 고콜레스테롤식이(high cholesterol diet: HCD)로 유발된 고콜레스테롤혈증과 간손상에 대한 발효녹차추출물(fermented green tea: FGT)의 효과를 평가하였다. 10 주령의 golden syrian 햄스터를 활용하여 일반식이(normal diet: ND)와 0.2% 콜레스테롤과 10% lard가 포함된 HCD를 12주간 제공하였으며, FGT(200과 500 mg/kg/day)를 12 주간 경구 투여하였다. FGT는 체중증가, 음식섭취 및 칼로리섭취에 영향을 주지 않았지만, HCD에 의하여 증가한 혈장 LDL, TC, 중성지방(triglyceride: TG)을 유의하게 억제하였다. HDL은 HCD에 의해 현저하게 증가하였고 FGT 투여에 의한 추가적인 변화는 없었다. 또한 FGT는 HCD에 의한 간 조직내 산화성스트레스, 섬유화 및 지질축적을 억제하였다. 흥미롭게도, FGT는 담즙산합성의 속도 제한 효소인 콜레스테롤 7-α hydroxylase(CYP7A1)의 단백질 수준을 크게 증가시켰으며, 대변을 통한 TC 배설도 증가시켰다. 정리하면, 본 연구에 활용한 FGT는 아마도 CYP7A1발현을 증가시켜 대변을 통한 콜레스테롤의 배설을 증가시킴으로써 식이로 유발된 고콜레스테롤혈증 및 NAFLD 발생을 억제 할 수 있는 건강 기능 식품이 될 수 있음을 시사하였다.
