Abstract
Traf2- and Nck-interacting kinase, TNIK, is known to act as an essential activator of canonical Wnt/-catenin WNT signaling pathway and regulate cytoskeletal rearrangement. However, how TNIK is involved in cytoskeletal rearrangement is still elusive. Moreover, the cytoskeletal rearrangement is known to be regulated by the noncanonical WNT signaling pathway, especially the Planar Cell Polarity (PCP) pathway. Thus, I investigated possible roles of TNIK in the non-canonical WNT pathway – the Wnt/PCP pathway – in relation to cell migration and the membrane protrusion. I first developed tnik-/- zebrafish mutant and observed abnormal cellular movement during epiboly stages of development and defects at the posterior tissue. Additionally, I found that LASP2, an actin-binding protein which is mainly located at focal adhesions, interacts with TNIK. Knockdown of TNIK by siRNA showed a decrease in cell migration and also resulted in a decreased expression level of both LASP2 and paxillin, a focal adhesion adaptor protein, at the leading edge of colon cancer cell, HCT116. Interestingly, HCT116 cells that TNIK was stably overexpressed resulted in a phenotype of collective cell migration with an increased migration rate as well as the expression level of E-cadherin, indicating the cells retained epithelial characteristics. Collectively, our data suggest possible roles of TNIK in the cytoskeletal rearrangement in the context of the cell migration and the membrane protrusion through LASP2 in colorectal cancer cell, HCT116. ;Traf2와 Nck 단백질과 상호작용하는 인산전달효소인 TNIK은 b-catenin 의존적인 Wnt 신호전달계에서 중요한 활성인자로써 역할을 한다고 알려져 있으며, 세포 골격의 재배열 과정을 조절한다고도 알려져 있다. 더 나아가, 세포 골격의 재배열 과정은 -catenin 비의존적인 Wnt 신호전달계인 planar 세포 극성 전달계에 의해 조절되고 있다. 따라서, 세포 이동과 세포막의 돌출부 형성을 통해 -catenin 비의존적인 Wnt 신호전달계에서의 TNIK의 역할에 대해 연구를 진행하였다. 먼저 tnik-/- 이 결여 된 제브라피쉬 모델을 제작하여 배아 발생 관찰 결과, 피포 과정에서 비정상적인 세포 이동이 관찰 되었으며 꼬리 부분에서도 발생 결함을 관찰하였다. 추가적으로, 세포 골격을 구성하는 actin과 상호작용하며 세포막 최첨단의 focal adhesion에 위치하고 있는 LASP2 단백질이 TNIK과도 상호작용한다는 것을 확인하였다. 대장암 세포주인 HCT116에서 TNIK의 발현이 siRNA에 의해 억제되었을 때, 세포의 이동이 감소하였으며, 동시에 LASP2 와 focal adhesion의 위치하고 있는 paxillin 단백질의 발현도 감소하였다. 흥미롭게도, HCT116 세포주에서 TNIK이 지속적으로 발현이 되었을 때, 세포들이 서로 집단을 형성하여 자라는 것을 관찰하였으며, 상피세포에서 관찰할 수 있는 E-cadherin의 발현량도 증가하였다. 이러한 결과는 대장암 세포에서 TNIK이 LASP2를 통해 세포 골격의 재배열의 관여 가능성을 보여주며, 세포 골격의 재배열을 통해 세포 이동과 세포막의 최첨단 부분 형성에서의 역할 가능성을 시사하고 있다.
