Abstract
Hibiscus is commonly consumed as beverage which contains variety of bioactive compounds and chlorogenic acid is one of them. Chlorogenic acids (CGAs) is formed by ester bonding of caffeic acid and quinic acid and it is well known as antioxidants which has effect on degenerative diseases such as cancer, and diabate. CGAs also has isomers named neochlorogenic acid, and cryptochlorogenic acid which are esterified on different site of caffeic acid. Subcritical water extraction (SWE) technique is eco-friendly and highly efficient extraction because the major solvent for SWE is water not organic solvent and extraction time is very brief in comparison to conventional extraction technique. SWE extraction conditions for Hibiscus were as follows: extraction time of 10, 20, 30, and 40 min; extraction temperature of 110, 130, 150, 170, and 190°C. SWE was performed using Dionex Accelerated Solvent Extractor (ASE 350, Dionex) and then extracts were analyzed using HPLC. The maximum concentration for chlorogenic acid, neochlorogenic acid, cryptochlorogenic acid was observed at different extraction condition respectively. The optimal extraction condition of chlorogenic acid was 190°C-10 min, of neochlorogenic acid was 110°C-10 min, and of cryptochlorogenic acid was 130°C-40 min. In addition, we observed varying extraction trend with extraction temperature and time and we also found that the conformation of the residues and the structural stability determined by binding position was related to the varying extraction yield with extraction conditions, To the best our knowledge, there was no study dealing with the correlation between steric and structural stability and optimization of subcritical water extraction. In this study, we optimized SWE extraction condition for chlorogenic acid and its isomers, and the relationship between different optimal extraction condition for three isomers was discussed.;히비스커스는 주로 음료나 민간요법으로 소비되어 왔던 고부가가치의 식물 원료이다. 수많은 선행연구를 통하여 항암, 항산화, 항바이러스 등의 건강기능성을 가지고 있음이 확인되면서 최근에는 정제, 티백 등의 다양한 형태로 가공되거나 화장품의 원료로서도 사용되며 그 범위가 확장되고 있다. 특히, 히비스커스에 함유되어 있는 클로로겐산류는 항산화 기능을 가지고 있다고 잘 알려져 있는 화합물의 한 종류이다. 뿐만 아니라 암, 당뇨병과 같은 퇴행성 질환에도 영향을 나타내는 항산화제로서 잘 알려져 있다. 클로로겐산류는 카페인산과 퀸산의 에스테르 결합에 의해 형성되며, 퀸산의 각기 다른 위치에 커피산이 에스테르화 된 클로로겐산, 네오클로로겐산, 크립토클로로겐산은 서로 이성질체 관계이다. 본 연구는 아임계수 추출 기술을 이용하여 히비스커스로부터 세 종류의 클로로겐산류에 대한 추출 조건을 최적화하여 추출 수율을 증대시키고자 하였다. 뿐만 아니라, 추출 조건에 따른 세 물질의 추출 경향을 관찰하고, 이성질체의 물리화학적 특성과 추출 조건 간의 상관관계를 파악하고자 하였다. 아임계수 추출은 Dionex Accelerated Solvent Extractor (ASE 350, Dionex)를 사용하여 수행하였으며 추출 시간은 10, 20, 30, 40분으로, 추출 온도는 110, 130, 150, 170, 190°C으로 총 20개의 조건을 설정하였다. 추출물은 동결건조 한 뒤 50%(v/v)의 메탄올에 녹여 HPLC 분석하였다. 그 결과, 클로로겐산, 네오클로로겐산 및 크립토클로로겐산의 최적 추출 조건은 각각 190°C/10분, 110°C/10분, 130°C/40분이었다. 세 물질 간에 최적의 추출 조건 및 추출 경향이 다르게 나타나는 원인은 세 이성질체마다 다르게 가지는 수산화 잔기의 입체구조와 아실기의 결합 위치에 따른 안정성의 차이, 화합물의 이성질화 및 분해 등에서 기인한 것으로 분석된다. 이에 더해, 실험을 통해 메탄올 용매 추출의 최적 조건으로 도출된 50%(v/v)/6시간에 비교하였을 때, 수율 측면에서 2~3배와 시간 측면에서 6~24배 효율적임을 확인하였다. 본 연구는 결과적으로 히비스커스로부터 세 종류의 클로로겐산류의 추출 효율을 극대화하는 데에 아임계수 추출 기술이 효과적임을 입증하였으며, 더 나아가 식물 및 식품 소재로부터 기능성 물질의 추출 수율을 증대시킬 수 있는 친환경적이고 효율적인 신 가공 기술로서의 가능성을 확인하였다. 뿐만 아니라, 이성질체의 물리화학적인 특성이 추출 효율과 연관되어 있음을 확인하였다. 따라서 아임계수 추출 기술을 활용한 기능성 물질의 추출 극대화를 통해 고부가가치의 식품 소재 개발 등의 산업적 활용 가능성을 기대한다. 또한 앞으로의 아임계수 추출 기술에 관련된 연구를 수행함에 있어서 목표 물질의 입체구조를 파악하고 이를 적용하여 연구의 효율을 높일 수 있음을 기대한다.
