Abstract
In chapter Ⅰ, the bis-binaphthol-pyrene derivative 1 selectively senses Zn2+ by undergoing a reversible fluorescent emission enhancement at 550 nm. Moreover, upon addition of Zn2+, the magnitude of the peak in the CD spectrum of 1 at 289 nm increases and the intensity of the peak at 272 nm decreases. The distinctive fluorescence and ratiometric CD spectral changes are attributed to a change in the dihedral angle between the binaphthol groups in 1 caused by Zn2+ binding. Probe 1 was employed to image Zn2+ in cells. In addition, the 550 nm emitting 1-Zn2+complex was utilized for enantioselective recognition of D-scyllo-inositol 1,2-bisphosphate (D-scyllo-I(1,2)P2) versus L-scyllo-I(1,2)P2. Specifically, addition of D-scyllo-I(1,2)P2 (2 eqiv.) completely quenches emission of the 1-Zn2+ complex at 550 nm and a relatively smaller quenching effect occurs upon addition of L-scyllo-I(1,2)P2. In chapter Ⅱ, we designed and synthesized probe 2 and 3, BINOL derivative containing two phenol and imine moieties, for detecting metal ions. In the presence of Zn2+, probe 2 and 3 undergo a highly selective enhancement of the broad intramolecular excimer fluorescence band at 576 nm. These changes are attributed to a formation of an intermolecular charge transfer complex promoted by binding of Zn2+. Additionally, we observed that probe 2 can be used to image Zn2+ in HeLa cells. In chapter Ⅲ, probe 4 which is a simple bis-pyrene derivative bearing pyridine unit undergoes a selective enhancement of the fluorescence band at 460 nm in the presence of Hg2+. These changes are attributed to a formation of the static excimer between probe 4 and Hg2+ with 1:1 stoichiometry. In contrast, Al3+ and Fe3+ induce only relatively small enhancement of the monomeric pyrene emission bands and a small decrease in the intensity of dynamic excimer band. Furthermore, another utility of probe 4 was examined in this study. Interestingly, among various reactive oxygen species, only ∙OH promotes a large fluorescent quenching of probe 4 at 475 nm. As a consequence of this notable observation, our group is scheduled to implement the further experiment to find the quenching mechanism between probe 4 and ∙OH.;1 장에서는 카이랄 인식을 위한 비스-나프톨-파이렌(bis-naphthol-pyrene) 유도체인 탐침 1을 디자인하고 합성하였다. 탐침 1은 550 nm 에서 가역적인 형광 방출 증가를 통해 아연 이온을 선택적으로 감지한다. 또한, 아연 이온의 첨가 시, 탐침 1의 원 편광 이색성(Circular Dichroism, CD) 스펙트럼에서 289 nm 의 피크 세기가 증가하고 272 nm 에서 피크의 강도가 감소한다. 이러한 독특한 형광과 CD 스펙트럼 상의 변화는 아연 이온과의 결합에 의한 탐침 1의 바이나프톨 그룹 사이의 2 면각의 변화에 기인한다. 탐침 1은 세포에서 아연 이온의 영상화를 위해서도 사용될 수 있다. 또한, 550 nm 에서 발광하는 탐침 1-Zn2+ 복합체는 실로-이노시톨 1,2-비스포스페이트(scyllo-inositol 1,2-bisphosphate)의 거울상 이성질체를 선택적으로 인식할 수 있다. 구체적으로, 실로-이노시톨 1,2-비스포스페이트의 D 이성질체를 첨가하면 550 nm 에서의 탐침 1-Zn2+ 착물의 형광 방출을 완전히 소광시키는 반면, L 이성질체를 첨가하면 상대적으로 작은 소광 효과를 나타낸다. 2 장에서는 금속 이온 검출을 위한 두 개의 페놀 그룹과 이민 그룹을 함유한 바이놀 (1,1’-bi(2-naphthol), BINOL) 유도체 2 와 3 을 디자인하고 합성하였다. 탐침 2 와 3 은 다양한 금속 이온 중 아연 이온과 선택적으로 상호 작용하여 576 nm 에서 형광 강도 증가를 나타낸다. 이러한 형광 세기 변화는 아연 이온과의 결합에 의해 촉진되는 분자간 전하 이동 착물의 형성에 기인한다. 추가적으로, HeLa 세포에서 아연 이온을 영상화하는 데 탐침 2 가 사용될 수 있음을 관찰하였다. 본 연구단은 카이랄 시스템에 많이 응용되는 바이놀의 특성에 착안하여 탐침 2, 3 – 아연 이온 착물과 반응하여 선택적인 형광 변화를 나타내는 카이랄 분자를 찾기 위해 추가적인 연구를 진행할 예정이다. 3 장에서는 피리딘 단위를 갖는 비스-파이렌(bis-pyrene) 유도체인 단순한 구조의 탐침 4가 수용액에서 다양한 금속 이온들 중 수은 이온에서만 선택적인 형광 증가를 나타냄을 관찰하였다. 이러한 변화는 탐침 4 와 수은 이온 사이에 1:1의 화학량론으로 정적 엑시머(static excimer)가 형성되는 것에 기인한다. 대조적으로, 알루미늄 이온과 철(Ⅲ) 이온은 단량체 파이렌 방출 밴드의 상대적으로 작은 증가 및 동적 엑시머(dynamic excimer) 밴드의 작은 감소만을 유도한다. 이에 더해, 본 연구에서는 탐침 4 의 또 다른 유용성을 조사하였다. 흥미롭게도, 다양한 활성 산소 종 중에서 히드록실 라디칼(∙OH)을 첨가하였을 때에만 475 nm 에서 탐침 4 의 큰 형광 소광을 촉진하였다. 이러한 흥미로운 결과에 주목하여, 본 연구단은 탐침 4 와 히드록실 라디칼 사이의 소광 메커니즘을 찾기 위한 추가적인 실험을 실행할 예정이다.
