Abstract
Excessive reactive oxygen species (ROS) have been recognized as a provoking cause of numerous diseases and disorders including atherosclerosis. During normal redox metabolism, aerobic organisms are constantly exposed to ROS. Therefore, removal of ROS is crucial for maintenance of cellular homeostasis and metabolism. Peroxiredoxin 5 (Prdx5), an essential antioxidant enzyme, is known to regulate not only global oxidative stress but also pro-inflammatory responses. Previous studies reported that other Prdx isoforms play a protective role against atherosclerosis. However, the precise role of Prdx5 in the pathogenesis of atherosclerosis remains unknown. To identify the relationship between Prdx5 and atherosclerosis, Prdx5-/-ApoE-/- mice were generated by crossing Prdx5-/- mice with ApoE-/- mice. Prdx5-/-ApoE-/- mice exhibited an increase in the atherosclerotic plaque formation and necrotic core volume, demonstrating that Prdx5 deficiency accelerates atherogenesis. To understand the molecular mechanism that Prdx5 deficiency aggravates atherosclerosis, the study was conducted in three aspects: metabolic syndrome, lipid metabolism, and inflammation. Interestingly, Prdx5 deficiency did not affect metabolic characteristics and overall lipid metabolism. However, the transcription levels of Srebp-1a and -2, mediators linking lipid metabolism to innate immune responses, were increased in the liver of Prdx5-/-ApoE-/- mice. Furthermore, the transcription levels of most pro-inflammatory cytokines, chemokines, and adhesion molecules were enhanced, while the transcription levels of anti-inflammatory cytokines were reduced in the aorta of Prdx5-/-ApoE-/- mice. These results suggested that deficiency of Prdx5 could alleviate atherosclerosis by activating an inflammatory response. Taken together, these results verified the ability of Prdx5 to suppress atherogenic responses and suggest its potential usefulness as an effective anti-atherogenic therapy.;호기성 유기체는 정상적인 산화 환원 대사 과정에서 끊임없이 활성산소종에 노출된다. 과다한 활성산소종은 동맥 경화를 비롯한 수많은 질병과 장애의 유발 원인으로 알려져 있다. 따라서 항산화 효소에 의한 과도한 활성산소종의 제거는 세포의 항상성과 신진 대사에 필수적이다. 동맥경화는 만성적인 염증질환으로 동맥경화 초기에는 혈액 안의 저밀도 지질 단백질이 혈관의 내피에 유입되어 다양한 염증 인자들을 활성화시키게 된다. 퍼옥시레독신 (Peroxiredoxin, Prdx)의 여섯 가지 동형 단백질 중 가장 최근에 발견된 퍼옥시레독신-5 (Prdx5) 유전자는 마우스의 생체 내에서 항산화 작용뿐만 아니라 전 염증 반응을 조절하는 것으로 알려져 있다. 현재까지 보고된 선행 연구결과에 따르면 Prdx1, 2를 포함한 다른 Prdx 동형단백질들이 결핍되었을 때 대동맥 내에서 염증반응이 유발되고 동맥경화가 촉진된다는 기존 보고가 있다. 지금까지 보고된 연구 사례에 따르면, Prdx5 유전자가 결핍된 세포의 경우 과산화수소, 방사선조사나 약물에 의한 DNA 손상과 세포 사멸이 증가된다는 in vitro 연구를 통해 알려져 있으나, 실제 in vivo 상에서 Prdx5 유전자의 기능에 대한 연구는 거의 보고된 바가 없다. 또한, 동맥경화의 발병과 그 과정에서 Prdx5 유전자의 기능을 입증한 보고 역시 아직 없다. 이를 토대로 강력한 항산화효소인 Prdx5이 동맥경화 상황에서 긍정적인 기능을 할 것으로 기대했다. 따라서 본 연구에서는 동맥경화 내에서 Prdx5의 역할을 확인하기 위해, Prdx5가 결핍된 마우스를 동맥경화 모델 마우스인 ApoE가 결핍된 마우스와 교배시켜 Prdx5/ApoE 이중유전자결핍 마우스를 제작하였고, 동맥경화 표현형 분석을 진행하였다. 예상대로, Prdx5/ApoE 이중유전자결핍 마우스의 대동맥과 대동맥궁에서 병변의 크기가 유의적으로 증가한 것을 확인하였다. 또한, 대동맥 궁에서 괴사 면적이 증가된다는 것을 알아냈다. 즉, 이는 Prdx5의 결핍이 동맥경화를 촉진할 것으로 생각했던 가설을 입증했다. 이 결과를 바탕으로 Prdx5의 결핍이 어떠한 과정을 통해 동맥경화증을 촉진시키는 것을 알아내기위해, 대사 증후군, 지질 대사 이상 및 염증반응 측면에서 추가적인 연구를 진행했다. 그 결과 Prdx5의 결핍이 대사질환이나 전반적인 지질 대사에는 영향을 미치지 않는다는 것을 확인했다. 하지만 흥미롭게도, 지질 대사와 선천적 면역반응을 연결시키는 매개체인 Srebp-1a와 -2의 전사 수준이 Prdx5/ApoE 이중유전자결핍 마우스의 간에서 증가한 것을 확인했다. 이를 토대로 동맥경화 병변 주변에 존재하는 면역세포들의 활성 정도를 확인하고자, 대동맥의 동맥경화 병변 내에서 면역세포들이 분비하는 사이토카인 (cytokine)과 케모카인 (chemokine) 그리고 부착분자 (adhesion molecules)들의 전령RNA 발현을 확인하였다. 그 결과, Prdx5/ApoE 이중유전자결핍 마우스의 대동맥에서 대부분의 프로-염증성 사이토카인, 케모카인 및 접착 분자의 전사 수준이 증가되었고, 항-염증성 사이토카인의 수준은 변화가 없거나 감소한 것을 알아냈다. 이는 Prdx5의 결핍이 염증 반응을 활성화시킴으로써 동맥경화를 촉진함을 시사한다. 본 연구 결과를 종합해보면, Prdx5 유전자의 결핍이 만성 염증 질환인 동맥경화의 발생과정에서 신진대사나 지질대사의 조절이 아닌 염증반응의 유도에 관여하여 병변을 증가시키고 동맥경화를 촉진시킨다는 사실을 증명하였다. 이와 관련된 분자적 메커니즘의 규명은 앞으로 동맥경화의 치료에 있어서 새로운 장을 열어 줄 것으로 기대되고, 향후 새로운 동맥경화의 치료제 개발에 있어 원천적인 기술을 제공 할 것으로 사료된다.
