Abstract
Syndecan은 세포 표면에 존재하는 heparan sulfate proteoglycan의 한 종류로써, 세포 밖 환경 변화에 대응하여 세포 외기질의 변화 및 세포 내부의 세포 골격의 변화를 조절하는 결합 수용체이다. 이러한 syndecan의 역할은 세포 이동이 활발한 암 세포에서 쉽게 확인할 수 있으며, 특히 syndecan-2는 대장암의 발병과 관련이 높은 것으로 알려져 있다. Syndecan-2는 정상적인 상피 세포에서는 발현되지 않고, 암화 과정에서 암 활성 특이적으로 세포 표면에 증가하여 대장암 세포의 암 활성을 조절한다고 밝혀졌다. 또한, 대장암 세포 표면에 증가 된 syndecan-2는 matrix metalloproteinase-7 (MMP-7) 의 발현을 증가 시키고, 직접적으로 MMP-7과 결합할 수 있음이 선행 연구에서 밝혀졌다. 한편, 최근 syndecan에 관한 연구는 세포 표면 수용체로써의 역할 뿐 아니라 완전한 세포외도메인 형태로 잘리는 절단 (shedding)에 관한 연구가 활발히 진행되고 있다. 이에 본 논문에서는 대장암 세포에서 syndecan-2 가 MMP-7에 의해 절단 되고, shedding 된 syndecan-2조각의 암 활성 역할을 확인함으로써 syndecan-2가 세포 표면 수용체로써 뿐 만 아니라 잘려진 조각의 형태로 대장암의 암 활성을 조절할 수 있음을 규명하였다. 다양한 대장암 세포주의 배양액을 모아 slot blotting을 한 결과 syndecan-2 조각이 세포 밖으로 유출되어 배양액에 존재함을 밝혔다. 또한, 대장암 세포에 syndecan-2를 과 발현 시킨 뒤, 배양액을 얻어 syndecan-2 항체를 이용한 면역침강을 시행하여 syndecan-2 조각을 재 확인 후, DEAE 이온교환수지를 이용하여 정제한 결과 세포 배양액으로 유출 된 syndecan-2 조각은 약 16 KDa임을 증명하였다. 나아가syndecan-2의 절단 현상이 MMP-7에 의존적인지 확인 한 결과, 대장암 세포에서 syndecan-2 는 MMP-2, -9이 아닌 MMP-7에 의해 절단이 되며 이는 MMP-7처리의 농도 및 시간 의존적임을 밝혔다. 또한, MALDI-TOF 분석 결과 MMP-7은 syndecan-2세포외도메인 류신149번 아미노산 위치를 절단하며, syndecan-2의 당 사슬의 유무에 관계 없이sydecan-2의 core protein을 절단할 수 있음을 밝혔다. 나아가 세포 배양액으로 유출 된 syndecan-2 조각이 대장암의 활성에 어떠한 역할을 하는 지 증명하였다. 먼저, syndecan-2절단 부위 돌연변이체를 제작하여 MMP-7에 의한 절단이 현저히 감소됨을 확인하였고, 이를 세포에 과 발현 시켰을 때 마찬가지로 배양액으로 유출되는 syndecan-2 조각이 감소됨을 확인하였다. 또한, syndecan-2 절단의 감소는 대장암 세포주 HCT116과 HT29의 증식에는 영향을 미치지 못하지만, 이동성과 콜로니 형성능을 현저하게 감소시켰으며, 이는B16F10 마우스 멜라노마 세포주를 이용한 폐 전이능 동물 모델을 이용하여서도 확인하였다. 이러한 결과는 절단 된 syndecan-2 조각이 암 세포의 이동성과 암 활성능을 촉진시킬 수 있음을 의미한다. 또한, 세포 배양액으로부터 정제 한 절단 된 syndecan-2 조각과 재조합 syndecan-2 조각을 대장암 세포에 처리하였을 경우, syndecan-2 조각이 직접적으로 대장암의 암 활성을 증가시킬 수 있음을 밝혔다. 나아가 절단 된 syndecan-2 조각의 암 활성 부위를 조사한 결과 C-말단 부위의 L79TSAAPKVETTTLNIQ94 아미노산이 암 활성을 조절하는 필수 부위임을 증명하였다. 이를 펩타이드로 합성한 뒤, 대장암 세포에 처리하였을 때, 대장암 세포의 활성이 충분히 증가됨을 확인하였으며 이를 간 및 폐 전이 마우스 모델과 피하 종양 모델을 이용하여 확인 한 결과 합성 펩타이드는 암 세포의 전이 및 원발성 고형암의 성장을 촉진 시킬 수 있음을 확인하였다. 한편, syndecan-2 조각은 대장암 환자의 혈장에 분비되며, 이는 범종양 마커인 CEA와 달리 초기 대장암에서도 관찰이 가능하였다. 또한, 대장암 환자의 암의 진행 정도와 혈장 syndecan-2 조각의 농도는 서로 상관관계가 성립하였으며, 혈장에 분비 된 syndecan-2 조각 역시 대장암 세포의 암 활성을 조절할 수 있음을 증명하였다. 이는 절단 된 syndecan-2 조각이 새로운 표지인자가 될 수 있으며, 초기 관찰을 통해 조기 진단이 가능할 수 있음을 시사한다. 또한, 대장암 환자 혈중의 syndecan-2 조각은 대장암 치료의 새로운 타겟으로 작용할 수 있을 가능성을 제시한다. 결론적으로, 절단 된 syndecan-2조각은 대장암 세포의 암 활성을 조절하고 새로운 암 진단 마커로써의 기능을 가짐으로써 대장암의 치료 및 조기 진단으로써의 중요한 역할을 수행할 수 있음을 제시한다. 이와 관련하여, 본 연구에서 사용한 syndecan-2 항체를 혈중 syndecan-2 조각의 검출 및 제거에 응용한다면, 대장암의 진단 및 치료제 개발을 효율적으로 도모할 것으로 기대한다. ;Syndecan-2, one of the cell surface heparansulfate proteoglycan, is known to regulate colon cancer cell activities by functioning as a docking receptor for matrix metalloproteinase-7 (MMP-7) at the cell surface. However, the exact regulatory mechanism of which syndecan-2 mediates regulation of colon cancer remains unknown. Since MMP can cleave proteolytically various cell surface receptors, pro-tumorigenic functions of syndecan-2 were investigated in colon cancer cells, related with MMP-7 function. First of all, activated MMP-7-mediated by syndecan-2 could cleave the extracellular domain of syndecan-2 in itself. In vitro digestion studies showed that MMP-7 cleaved syndecan-2 at the N-terminal of Leu149 residue, resulting in accumulation of syndecan-2 in culture media from colon cancer cells. In addition, increased serum levels of the soluble syndecan-2 have been analyzed for colon cancer patients and animal models compared with normal donors. Furthermore, the serum levels of shed syndecan-2 had positive correlation with the colon cancer progression. More importantly, shed syndecan-2 itself enhances colon cancer cell activities. Substitution of Leu149 to Ile does not only hamper syndecan-2 shedding by MMP-7, but also significantly reduces shed syndecan-2-induced migration and anchorage-independent growth. In addition, depletion of shed syndecan-2 also abolished these cancer-associated effects. Especially, a fragment L79TSAAPKVETTTLNIQ94 derived from the shed syndecan-2 extracellular domain is the core component for tumorigenecity of colon cancer. Taken together, these results provide shed syndecan-2 plays an essential role in the promoting tumorigenic activities in colon cancer cells and serum-shed syndecan-2 could be a promising colon cancer diagnostic marker. These suggest a valid therapeutic approach for colon cancer.
