- 폴리카프로락톤(PCL)은 오래전부터 뛰어난 생체적합성을 가진 고분자로 보고되어왔지만 생체 의학 재료로서의 연구는 더디게 진행되어 왔다. PLGA와 같은 고분자에 비해 합성 공정이 간단하여 가격경쟁력을 갖췄음에도 불구하고, 최대 2년이라는 긴 분해 기간을 갖는다는 제한이 있었다.
본 연구에서는 생체 내에서 빠르게 분해되는 옥살레이트에 결합된 폴리카프로락톤(PCL-OX) 다중 블록 공중합체를 합성하였다. 이 고분자는 합성 후 100,000 달톤의 분자량을 갖는 것으로 분석되었으며, 44.7oC의 녹는점과 40%의 결정화도를 갖는 것으로 나타났다.
합성된 고분자는 oil-in-water 단일 에멀젼 방법을 통해 소수성 항암제인 파클리탁셀을 봉입한 마이크로 입자로 만들어졌고, 비교 군인 PLGA 마이크로 입자 또한 같은 방법으로 준비되었다. 생체 외 약물 전달 실험 결과, PCL-OX 마이크로 입자는 PLGA 마이크로 입자보다 느린 약물 방출 결과를 보였다. 그러나 쥐의 피하에 마이크로 입자를 주입한 결과, PCL-OX 마이크로 입자와 PLGA 마이크로 입자의 약물 방출 속도가 28일 동안 비슷한 것으로 나타났다. 이 분해는 체내의 과산화수소에 가속된 것으로 나타났다. 또한, PCL-OX 마이크로 입자는 PLGA 마이크로 입자와 비슷한 생체적합성을 보이며 앞으로 PLGA를 대체할 수 있는 생체 의료용 재료로서의 가능성을 보였다.;Polycarprolactone (PCL) was reported long time ago, however its biomedical application has not extensively been investigated, compared with poly(lactide co-glycolide) (PLGA) due to its too slow degradation profile, even though polymerization process of PCL is much less expensive than PLGA. We are reporting an oxalate connected PCL multiblock copolymer (PCL-OX) as an alternative to PLGA as a fast in vivo degradable PCL. The degradation was accerlerated by ROS. PCL-OX with a molecular weight of 100,000 Daltons exhibited melting point of 44.7 oC and crystallinity of 40%. PCL-OX microspheres containing paclitaxel, a model drug, were prepared by the oil-in water emulsion method and were compared with PLGA microspheres containing the same drug. In vitro release of paclitaxel from the PCL-OX microspheres was slower than from the PLGA microspheres. However, the in vivo application of the PCL-OX microspheres by the subcutaneous injection into rats provides a similar plasma drug concentration to PLGA microspheres over 28 days. The PCL-OX microspheres showed a similar tissue compatibility to PLGA microspheres in the subcutaneous layer of rats with a marginal tissue irritation over 28 days. This paper suggests that the PCL-OX is a useful biomaterial that improved the slow degradation problems of PCL, and thus it opens other biomedical applications.
