Abstract
Synthetic hair dye is widely used by millions of people. However, hair dyes contain some skin sensitizing and irritating ingredients, which vary by their degree of skin sensitizing and irritating potency. To provide further understanding about skin allergies after using hair dyes, researchers have evaluated the sensitization potency of hair dyes. However, little information is available on the irritation effects of hair dyes(Lloyd, Ligget, Kynoch, & Davies, 1977). Skin sensitization and irritation reactions are similar in the outcome and manifestation. The skin sensitization and irritation can contribute to causing adverse effects of hair dyes on the skin. Thus, both the skin sensitization and irritation potency of hair dyes must be considered in order to avoid skin adverse effects. Here we aimed to provide a comprehensive evaluation on the skin sensitization and irritation of hair dyes using alternative animal test methods. In this study, 9 hair dye compounds permitted for use by the E.U. were initially selected. Those 9 substances were tested to evaluate the skin sensitization potency using Direct Peptide Reactivity Assay (DPRA), and the KeratinoSens™ assay. Eight substances were classified as sensitizers and m-Aminophenol (MAP), as the only one non-sensitizer according to DPRA. However, the KeratinoSens™ assay predicted only Resorcinol (RS) only as a non-sensitizer. Based on the skin sensitization data, we selected 5 of among the 9 tested dye compounds and conducted the skin irritation experiments with or without 6% of hydrogen peroxide solution on the two reconstructed human epidermis models, EpiDermTM and Keraskin™. A well-known sensitizer, p-Phenylenediamine (PPD) significantly decreased tissue viability in both skin models only in the presence of hydrogen peroxide. Furthermore, RS and 2-Methylresorcinol (MRP-N), which were predicted as a non-sensitizer and a low-DPRA–reactivity-sensitizer, manifested tissue-toxicity, which was synergistically increased by hydrogen peroxide. In conclusion, we provide an integrated evaluation of the skin sensitization and irritation of hair dye compounds, which would help to understand the dermal toxicity of hair dyes.;많은 소비자 들이 주기적으로 산화적 염모제를 사용합니다. 하지만 염모제 속에는 피부 감작성 및 자극성 물질이 존재합니다. 피부 감작성과 자극성 반응에는 개념상의 차이는 존재하지만 비슷한 부분이 많아 동시에 일어날 수도 있습니다. 따라서 염모제의 피부 안전성 평가에 있어서 감작성과 자극성 모두 고려해야 합니다. 특히 다양한 알러지성 피부염은 염모제 물질의 감작성에 의해 일어나는데 관련 연구는 상대적으로 많이 존재합니다. 하지만 염모제 물질의 피부 자극성 평가에 대한 자료는 부족합니다 따라서 본 연구는 동물대체법을 활용해 염모제 속 다양한 물질의 감작성과 자극성 자료를 통합적으로 제공하기 위해 진행되었습니다. 우선 E.U. 에서 허용한 9가지 염모제 물질을 선택 후 Direct Peptide Reactivity Assay (DPRA)와 KeratinoSensTM assay를 통해 감작성 정도를 측정하였습니다. DPRA에서는 m-Aminophenol (MAP)만, 그리고 KeratinoSensTM assay에서는 Resorcinol (RS)만 비감작성 물질로 분류되었습니다. 이러한 감작성 결과에 근거해 위의 9가지 물질 중 5가지 물질만 선택 후 EpiDerm™ 과 Keraskin™ 두 인공피부를 활용해 피부 자극성을 측정하였습니다. 실제 염색 과정 시 필수 물질인 과산화수소를 염모제 물질과 섞어 실질적 자극성을 평가하였습니다. 그 결과 2-Methylresorcinol (MRP-N), RS 그리고 p-Phenylenediamine (PPD)와 과산화수소를 함께 처리한 군에서 그 자극성이 증가한 것을 확인하였습니다. 따라서 감작성과 자극성은 항상 일치하지 않는다는 것을 확인했으며 과산화수소는 염모제 속 물질을 산화시켜 피부 자극성을 증가시킵니다. 추후 염모제 개발 시 감작성과 자극성을 동시에 고려하고, 과산화수소와의 반응성이 적은 물질을 선택하여 안전한 염모제 개발에 도움을 주고자 했습니다.
