Abstract
Bone homeostasis is maintained by the coordinated regulation of bone-forming osteoblasts and bone-resorbing osteoclasts. Imbalances of bone homeostasis cause various bone diseases. Diseases such as osteoporosis and rheumatoid arthritis are characterized by enhanced osteoclastogenesis. In contrast, osteopetrosis is characterized by a lack of osteoclasts and inheritable skeletal diseases. Osteoblasts are derived from bone marrow mesenchymal cells. Osteoclasts are multinucleated giant cells generated from hematopoietic monocyte/macrophage precursor cells by multi-step cell differentiation in response to macrophage-colony stimulating factor (M-CSF), and receptor activator of NF-κB ligand. Bone remodeling process is regulated by several molecules and mechanisms. Here, I report the role of SIRT6 on receptor activation of nuclear factor-κB ligand (RANKL)-induced osteoclast differentiation and the effect of (R)-TEMOSPho in osteoclast precursor cells. This doctoral dissertation is divided into two parts, i.e. SIRT6 and (R)-TEMOSPho. As described below. Part I: SIRT6 are highly conserved NAD+-dependent deacetylases and/or ADP-ribosyltransferases involved in transcriptional silencing, regulating genome stability, cellular metabolism, stress response and longevity. SIRT6 expression is induced in RANKL-induced osteoclastogenesis. Sirt6-deficient mice developed osteopetrosis due to decreased numbers of osteoclasts, indicative of a positive regulatory role for SIRT6. Sirt6-deficient osteoclast precursor cells underwent decreased osteoclastogenesis in response to RANKL, moreover Sirt6-deficient osteoclast precursor cells exhibited a markedly decreased capacity for bone resorption. NFATc1, a master transcription factor for osteoclastogenesis, binds to and activates the Sirt6 promoter. Activating transcription factor 4 (ATF4) expression was reduced in Sirt6-deficient osteoclast precursor cells. Moreover, the reintroduction of SIRT6 in Sirt6-deficient osteoclast precursor cells led to induction of ATF4, and complete restoration of osteoclast differentiation. Our results showed that SIRT6 is a positive regulator of osteoclast differentiation via regulation of ATF4 expression. Part II: (R)-TEMOSPho blocks osteoclast formation, and the resorptive function of mature osteoclasts. The inhibitory effect of (R)-TEMOSPho on osteoclasts was due to a disruption of the actin cytoskeleton, resulting from impaired downstream signaling of c-Fms, a receptor for M-CSF linked to c-Cbl, phosphoinositol-3-kinase (PI3K), Vav3, and Rac1. Moreover, (R)-TEMOSPho blocked inflammation-induced bone destruction by reducing the numbers of osteoclasts produced in mice. Thus, (R)-TEMOSPho may represent a promising new class of antiresorptive drugs for the treatment of bone loss associated with increased osteoclast formation and activity.;포유동물에서 뼈는 몸을 지지하는 역할을 하고 미네랄의 저장고이며 콜라젠과 같은 단백질과 칼슘 포스페이트 형태로 존재하는 칼슘화된 단단하고 가장 큰 조직이다. 또한 뼈의 안쪽 공간에 위치한 조직인 골수에는 많은 면역세포와 골세포를 저장하고 있다. 골수에서는 한 줄기세포로부터 면역세포와 골세포인 파골세포로 분화된다. 골수 에는 면역세포와 골세포가 같이 존재하면서 여러 조절물질들을 공유하기때문에 이것으로 면역세포와 골세포가 밀접한 연관이 있음을 알 수 있다. 특히 조혈모세포로부터 분화되어 파골세포로 최종 분화를 유도하는 다양한 사이토카인과 신호전달 매개체들을 면역 세포의 기능을 조절한다고 알려져 있다. 조골세포는 골수 내에 존재하는 중간엽 줄기세포로부터 분화된다. 골격 계의 뼈 항상성은 뼈를 구성하는 단백질과 미네랄 축적을 유도하여 뼈를 형성하는 조골세포와 뼈의 표면의 단백질 분해 효소 분비 및 산성화에 의해서 뼈를 파괴하는 파골세포의 균형적인 활동을 통해 유지된다. 뼈의 대표적인 질환으로는 골다공증과 골석화증이 있는데 이것은 파골 세포와 조골세포 두 세포간의 활성에 조화의 불균형 즉 뼈의 항상성이 깨어지면서 유발되는 질병이다. RANKL 에 의한 신호전달은 파골 세포가 분화 및 활성에 결정적인 역할을 하는 것으로 알려져 있다. 또한 M-CSF 에 의한 신호전달도 RANKL 신호전달체계와 협력하여 파골 세포 분화에 기여하는 사이토카인 이다. 주요한 두 가지 사이토카인에 의해 신호전달체계가 시작되면서 파골전구세포는 파골세포로 분화되는데 이 과정에 관여하는 많은 물질들이 관여하는데 밝혀지지 않은 물질과 해결되지 않은 신호전달도 많이 있어 계속적인 연구가 필요하다. 본 논문에서는 파골전구세포로 부터 파골세포로 분화되는 과정에서 SIRT6 의 역할과 (R)-TEMOSPho 합성물질이 뼈 질환의 치료제로서 가능성이 있는지를 연구하였다. 논문의 첫 번째 장에서는 NAD+ 의존적 히스톤 탈아세틸화 기능을 가지고 있는 SIRTUIN 의 7가지 단백질 중에서 핵에 존재하는 SIRT6 단백질이 파골세포의 분화과정에서의 역할이 무엇인지 알아보았다. SIRT6 단백질은 RANKL 에 의해서 유도되는 파골세포분화 과정 동안에 단백질의 양이 증가하는 것을 확인하였다. 이는 파골 세포 분화과정에서 중요한 전사인자라고 알려져 있는 NFATc1에 의해서 SIRT6의 전사수준에서 조절됨으로써 SIRT6의 단백질 양을 촉진시키는 것을 확인하였다. SIRT6 단백질은 몸의 여러 장기에서 존재하기 때문에 특이적으로 파골세포로 분화시키는 줄기세포에서만 SIRT6의 유전자를 제거되는 마우스를 제작하여 SIRT6의 자세한 기능을 알아보았다. 만들어진 마우스는 태어날 때부터 SIRT6가 제거된 것이 아닌 태어난 후 9일 이후에 SIRT6 유전자가 제거된 마우스 로서 뼈의 리모델링 과정에서의 SIRT6 역할을 자세히 연구하였다. 만들어진 마우스로 뼈 분석을 한 결과 골석화증으로 판별되었다. 또한 세포수준에서 더 자세히 기능을 연구하기 위해서 SIRT6가 제거된 마우스로부터 유래한 파골전구세포를 사용하여 파골 세포 분화 능력이 감소하는 것을 확인하였으며, 파골세포의 기능을 확인할 수 있는 뼈 흡수 활성도가 감소되는 것을 확인함으로써 SIRT6는 파골세포로 분화되는 과정에 촉진인자로 작용하는 것을 알 수 있었다. 기존에 SIRT6는 히스톤 디아세틸레이즈를 통해서 많은 유전자의 발현을 조절한다고 알려져 있는데 SIRT6가 제거된 마우스로부터 유래한 파골전구세포에서 유전자의 발현변화를 보이는 유전자중 파골 세포 분화에서 알려져 있는 ATF4 유전자를 찾게 되었다. SIRT6 제거된 파골전구세포에서 ATF4 유전자의 발현이 8배 감소되어 있음을 확인하였고 ATF4 를 SIRT6 가 제거된 파골전구세포에 넣어주었을 때 ATF4에 의해서 파골세포의 분화가 증가하는 것을 확인하였다. 또한 SIRT6 가 제거된 파골전구세포에 SIRT6를 다시 넣어주었을 때 파골 세포 분화능력이 다시 증가함을 확인하였고 ATF4 단백질이 증가되는 것을 확인할 수 있었다. ATF4는 C/EBPβ 에 의해서 발현이 억제되고 eIF2α 에 의해서 발현이 촉진된다고 알려져 있는데, SIRT6가 와 붙는 것을 확인 하였다. 이로써 SIRT6는 C/EBPβ에 붙음으로써 ATF4의 발현을 직, 간접적으로 조절함으로써 파골전구세포로 부터 파골 세포로의 분화 과정을 촉진인자로 작용하는 것을 규명하였다. 두 번째 장에서는 녹용추출 지질계 합성 물질인 (R)-TEMOSPho 가 파골전구세포로 부터 파골 세포로의 분화 과정에 어떤 영향을 주며 치료제로 가능성이 있는지를 연구하였다. (R)-TEMOSPho 는 TEHSPho 에서 파생된 합성물질이며 TEHSPho 는 파골세포분화를 억제한다고 알려진 바 있다. 파골 세포는 분화가 시작되면서 단핵세포에서 다핵세포로 바뀌고 다핵세포는 actin ring 을 형성하게 되고 뼈 흡수의 활성을 가지게 되면서 파골세포의 기능을 하게 되다. (R)-TEMOSPho 처리된 파골전구세포는 파골세포의 분화 과정에서 분화는 되지만 정상적인 파골 세포 형태를 가지지 못하며 acitn ring 형성을 못하여 뼈 흡수 활성이 감소됨을 확인하였다. 실제로 마우스 에서 (R)-TEMOSPho의 영향을 보기 위해서 LPS 로 염증을 유도시킨 후에 (R)-TEMOSPho 를 처리하였더니 뼈에 파골세포의 수도 감소하였으며 골 파괴 정도가 회복되어 있음을 확인하였다. 이러한 결과는 인구의 고령화가 진행되면서 가장 대표적인 뼈 질환인 골다공증 에서 (R)-TEMOSPho가 좋은 치료제로서 가능성 있음을 제시한다.
