- Abstract
- Glioblastoma multiforme (GBM), is most common primary brain tumor in adults. Featured characteristic of GBM is diffuse infiltration of tumor cells into normal brain parenchyma, rapid growth, a high degree of infiltration of microglia and macrophage, and presence of necrosis. Microglia/microphages are frequently found in glioma and extensively infiltrate GBM tissue up to 30% of total tumor mass. However, little is known about the effect of necrotic cells on microglia infiltration in GBM and the tumor-infiltrating microglia-induced factors in GBMs. In this study, to address whether necrosis or necrosis-exposed GBM cells affect the degree of microglia/macrophage infiltration, migration and invasion/infiltration assays were performed. The presence of necrotic cells promoted the migration/infiltration of microglia, and CRT-MG cells exposed to necrotic cells further enhanced the migration and infiltration of HMO6 microglial cells. Treatment of necrotic cells induced mRNA and protein expression of chemokines such as CCL2/MCP-1 and CCL20/MIP-3α in CRT-MG cells. In particular, CCL2/MCP-1 and CCL20/MIP-3α were significantly increased in necrotic cells-treated CRT-MG cells. Necrotic cells induced DNA binding of the transcription factors NF-κB and AP-1 to the CCL2/MCP-1 and CCL20/MIP-3α promoters, leading to increased CCL2/MCP-1 and CCL20/MIP-3α mRNA expression and protein expression. These results provide the evidence that necrotic cells induce the expression of CCL2/MCP-1 and CCL20/MIP-3α in glioblastoma cells through activation of NF-κB and AP-1 and facilitates the infiltration of microglia into tumor tissues.;교모세포종은 성인의 뇌에서 가장 흔히 발병하는 원발성 종양이다. 교모세포종은 정상 뇌조직에 분산되어 침윤되고 빠르게 성장하며 높은 정도의 소교세포 및 대식세포의 침윤, 그리고 괴사 세포의 존재가 특징이다. 소교세포/대식세포는 교모세포종 내에서 발견되고 전체 종양 질량의 30%까지 광범위하게 침윤되어있다. 하지만, 교모세포종 세포가 괴사 세포에 노출되었을 때 소교세포 침윤에 대한 영향과 종양으로 침윤하는 소교세포를 유도하는 인자에 대한 연구는 아직 없다. 이 연구에서는 괴사 세포 또는 괴사 세포에 노출된 교모세포종 세포가 소교세포/대식세포의 침윤에 미치는 영향을 확인하기 위해 이동 및 침윤 분석을 통해 괴사 세포가 소교세포의 이동/침윤을 촉진하고 괴사 세포에 노출 된 교모세포종이 HMO6 소교세포의 이동과 침윤을 더 많이 증가시키는 것을 확인하였다. 교모세포종에 괴사 세포를 처리한 경우 CCL2/MCP-1 과 CCL20/MIP-3α 과 같은 케모카인의 mRNA 와 단백질 발현을 유도하였다. 특히 CCL2/MCP-1 과 CCL20/MIP-3α 은 괴사 세포에 노출된 교모세포종에서 상당히 증가하였다. 괴사 세포는 CCL2/MCP-1 과 CCL20/MIP-3α promoter 에 대한 전사인자인 NF-κB 와 AP-1 의 DNA 결합을 유도하였고, 이를 통해 CCL2/MCP-1 과 CCL20/MIP-3α 의 mRNA 와 단백질 발현이 증가하였다. 본 연구를 통해, 괴사세포에 노출된 교모세포종에서 NF-κB 와 AP-1 의 활성을 통해 CCL2/MCP-1 과 CCL20/MIP-3α 의 발현을 유도하고 이는 종양 조직으로 소교세포 침윤을 촉진시킴을 확인하였다.
