Abstract
P-glycoprotein (MDR1/ABCB1), the well-known member of the ATP-binding cassette (ABC) family of drug transporters, is in many cases, overexpressed in chemoresistant cancer cells. The acquisition of MDR1/ABCB1-mediated chemoresistance poses a major obstacle to the success of conventional chemotherapeutic agents such as doxorubicin, vinblastine, paclitaxel, vincristine and others. Heat shock factor 1 (HSF1) is a transcription factor involved in the expression of heat shock proteins in response to heat shock or oxidative stresses. It is also widely suggested that HSF1 is involved in carcinogenesis such as metastasis, angiogenesis and chemoresistance. Subsequent studies have demonstrated the relationship between HSF1 and MDR1/ABCB1. However, the exact role of HSF1 on the expression of MDR1/ABCB1 is still undefined. There are reports showing that the overexpression of HSF1 upregulates the expression of MDR1/ABCB1. On the contrary, other reports demonstrate that HSF1 downregulates the expression of MDR1/ABCB1 interrupting binding of nuclear factor κB (NFκB) on DNA, the well-known transcription factor of MDR1/ABCB1. Since the regulation of MDR1/ABCB1 expression is complicated and the interplay between factors involved in the regulation varies widely, there lies more intense need to understand the precise mechanism involved in the regulation. In this study, we investigated the relationship between HSF1 and MDR1/ABCB1 using the non-small cell lung cancer (NSCLC) cell line A549 to provide with another possible explanation for chemoresistance. We compared normal A549 cells with paclitaxel resistant A549-t100 cells and the influence of different forms of HSF1 on the expression of MDR1/ABCB1 was studied with RT-PCR and Western blotting. Our results indicated that knockdown of HSF1 reduced MDR1/ABCB1 expression in paclitaxel-resistant A549-t100 cells. A549-t100 cells showed an unique feature of altered post-translational modifications of HSF1 compared to that in normal A549 cells. Also, we further provide evidence that phosphorylation of Ser303 and Ser307, the characteristic inhibitory posttranslational modification sites, may play an important role in the regulation of MDR1/ABCB1. However, the specific mechanism involved in this regulation needs to be further investigated. ;항암제 내성의 기전에 대한 이해는 미래의 항암제 개발 전략에서의 필수적인 요소로 관련 연구들이 다양하게 진행되고 있다. 대표적인 항암제 내성 기전으로 약물 수송에 관여하는 multi drug resistance 1 (MDR1/ABCB1, P-gp) 단백질의 발현 증가가 알려져 있으며, 이는 암 세포로의 약물 전달 체계에서 결함을 야기한다. 또한, 열충격 혹은 산화적 스트레스에 의해 유도되는 heat shock factor 1 (HSF1)이 암화 과정(carcinogenesis)과 항암제 내성 등에 관여한다는 보고가 있으며, 암세포에서 종종 그 발현이 증가하는 것을 관찰할 수 있다. 많은 연구에서 HSF1과 MDR1/ABCB1의 발현 사이의 관계를 제시하였으나, 상반되는 연구 결과들이 보고 된 바 있다. HSF1이 직접적인 전사인자로 작용하여 MDR1/ABCB1의 발현을 증가시킨다는 보고가 있으며, 이와는 반대로 HSF1이 또 다른 전사인자인 nuclear factor κ B((NFκB)의 작용을 저해하여 MDR1/ABCB1의 발현을 억제한다는 보고가 있다. MDR1/ABCB1 발현 조절은 매우 복잡하며 다양한 전사인자가 복합적으로 작용하여 정확한 작용 기전을 규명하는 데 어려움이 있다. 비소세포폐암은 세계적으로 암 관련 사망의 대표적인 원인으로 꼽히고 있으며, 1차 혹은 2차 치료 요법에서 병용 혹은 단독으로 사용되는 paclitaxel은 항암제 내성을 가지는 대표적인 약물로 알려져 있다. 본 연구에서는 비소세포폐암 모델을 이용하여 HSF1과 MDR1/ABCB1의 발현 사이의 관계를 규명하는 것을 목표로 하였다. Paclitaxel에 대한 내성을 획득한 A549-t100 모델에서 HSF1과 MDR1/ABCB1의 발현이 증가한 것을 확인할 수 있었으며, HSF1의 발현을 억제하였을 때, MDR1/ABCB1의 발현 또한 감소하는 것을 확인하였다. 또한, HSF1의 전사 후 조절 양상이 일발 A549모델과는 다른 양상을 가지는 것을 확인하였다. 일반적으로 HSF1의 Ser303과 Ser307 잔기의 인산화는 HSF1의 전사 개시 능력을 억제하는 것으로 알려져 있으나, 본 실험에서는 A549-t100 내성 모델에서 Ser303과 Ser307의 인산화가 증가한 것을 확인하였으며, MDR1/ABCB1의 발현에 관여하는 것을 확인하였다. 하지만, 구체적인 기전에 대해서는 더 많은 연구를 필요로 한다.
