Abstract
The mechanistic target of rapamycin (mTOR) signaling pathway integrates diverse signaling cues and controls cellular functions linked to cell growth, proliferation and metabolism. Notably, mTOR signaling plays a critical role in brain development, and aberrant activation of mTOR signaling has been etiologically implicated in the development of tuberous sclerosis complex (TSC)-associated neurological lesions. To investigate the gene expression changes induced by mTOR hyperactivation in developing central nervous system, RNA-sequencing analysis was conducted with neonatal telencephalon of Tsc1 conditional knockout (CKO), a model mouse for TSC disease. Among the most highly induced was the solute carrier family 7 member 7 (Slc7a3) gene. Experiments carried out in vitro and in utero for functional characterization are presented. In addition, proteomic analysis using LC-MS/MS was performed to compare differentially expressed genes and proteins in the Tsc1 CKO mouse brain. Integration of transcriptomic and proteomic data provided a comprehensive gene expression profile affected by hyperactivated mTOR signaling and novel marker genes associated with neuronal development. In particular, multiple proteins involved in neuronal development were identified and demonstrated to be down-regulated in the forebrain of Tsc1 CKO. Taken together, these results present an enriched molecular basis for functional dissection of mTOR signaling in the developing brain and for understanding pathogenesis of neurological disorder from mTOR dysregulation.;mTOR 신호전달체계는 다양한 세포신호를 통합하여 세포의 성장과 증식, 대사작용과 관련된 여러 가지 기능을 조절하는 중심 조절인자이다. 이러한 작용은 뇌의 발생에도 중요한 역할을 하며, 신경 발생 과정에서의 비정상적인 mTOR 신호전달체계 활성화는 결절성 경화증 (TSC; tuberous sclerosis complex)을 비롯한 신경 질환을 일으키는 병인학적 요인이 된다. 본 연구에서는 중추신경계의 발생과정에서 과잉 활성화된 mTOR 신호전달체계의 영향을 받는 유전자를 찾기 위해 차세대 염기분석법 (NGS; next generation sequencing)을 이용한 전령 RNA (mRNA; messenger RNA) 서열 분석을 수행하였다. 결절성 경화증의 모델 동물인 Tsc1 돌연변이 마우스 (Tsc1fl/flEmx1cre/+)의 전뇌 조직을 대상으로 mRNA 염기분석을 한 결과 solute carrier family 7 member 7 (Slc7a3) 유전자의 발현이 가장 많이 증가하였고, 그 기능을 알아보기 위하여 과발현 실험을 수행하였다. 또한, 액체 크로마토그래피-텐덤 질량분석법 (LC-MS/MS; liquid chromatography-tandem mass spectrometry)을 이용한 단백질체 분석 (proteomic analysis)을 통해 Tsc1 돌연변이 마우스의 전뇌에서 발현량이 변화된 단백질을 찾고 그 것을 유전자 발현 변화 결과와 비교하였다. 이러한 유전체와 단백질체의 통합적 분석으로 과잉 활성화된 mTOR 신호전달체계의 영향을 받는 유전자 발현 프로파일을 얻게 되었으며, 중합효소연쇄반응 (PCR; polymerase chain reaction)과 면역조직형광염색 (immunohistochemistry) 실험으로 신경 발생 과정과 관련된 새로운 마커 유전자를 선별하였다. 따라서 본 연구의 결과는 신경 발생 과정에서의 mTOR 신호전달체계의 기능을 이해하고 나아가 비정상적인 mTOR 신호전달체계 조절에 의한 신경학적 질환을 분자 수준에서 분석할 수 있는 기초적 자료가 될 것이다.
