dc.description.abstractTranslationally controlled tumor protein (TCTP) is a highly conserved protein and widely expressed in all eukaryotic organisms. It is involved in various biological processes such as protein synthesis, cell growth, and microtubule stabilization. Moreover, TCTP has cytokine like activity, anti-apoptotic property, and cell penetrating activity. Along with these functions of TCTP, many recent findings suggest that TCTP is a potential target for cancer. The beneficial effect of digitalis treatment in breast cancer patients has been suggested by epidemiological studies. Ouabain, one of the digitalis used for congestive heart disease and an Na,K-ATPase inhibitor, is also known to reduce breast cancer cell proliferation by triggering Na,K-ATPase mediated signal transduction. In our lab, we discovered that TCTP, an anti-apoptotic protein, inhibits Na,K-ATPase activity in HeLa cell, and that TCTP over-expression in MCF10A cell induces many different signaling pathways associated with tumor progression. In this study, I explored whether ouabain specifically inhibit tumor cells by regulating TCTP level. Using CCK-8 assay, I discovered that ouabain had cytotoxicity effect on HeLa cell when treated at 100 nM. However, there was no cytotoxicity effect on MCF10A cell when treated with same concentration of ouabain. Interestingly, the HeLa cell treated with 100 nM ouabain showed decrease in intracellular TCTP level, where as MCF10A cell treated with the same concentration showed increase in intracellular TCTP level, suggesting that modulating intracellular TCTP level is important is a key factor in ouabain-induced cell death. Furthermore, since TCTP seems to be an important target for cancer, we searched for a TCTP regulating molecule by Insilico screening. By measuring intracellular calcium level in primary cultured vascular smooth muscle cell (VSMC) and cell contractility in cardiomyocytes, the TCTP regulating ability of this Insilico Drug was tested. From calcium imaging system, using confocal microscopy, the relative ratio(340/380 nm) of calcium transient was 16% higher when VSMCs were treated with 100 μM Insilico Drug for 5 minutes compared to control. Moreover, using IonOptix system, the primary cultured cardiomyocytes showed 10.9% contraction when treated with 100 μM Insilico Drug for 5 minutes, while the control showed 8.2% contraction. From this study, it can be suggested that ouabain induced cell death in HeLa cell, but not in MCF10A cell, is due to difference in regulation of intracellular TCTP level. Furthermore, since regulation of TCTP is an important in many diseases, Insilico Drug (ISD1), which seems to regulate TCTP activity, has a potential to be a therapeutic drug with further research;Translationally controlled tumor protein (TCTP) 는 모든 진핵 생물에서 발현되며 단백질합성, 세포 성장, 미세소관 안정화 등 다양한 생물학적 반응에 관여한다. TCTP 는 암세포 및 암 조직에서 과발현 되어있으며 TCTP 과발현 한 쥐에서는 고혈압이 나타나는 등 여러 질병에 관여한다고 보고 된다. 본 연구실에서는 TCTP 가 Na+, K+-ATPase 의 활성을 억제하고 MCF10A 세포에서 과발현 되었을 때 암세포 증가에 관여하는 세포신호를 유발한다는 것을 밝혔다. 최근 역학조사에 따르면 cardiac glycoside 가 유방암에 탁월한 효과가 있다고 한다. Ouabain 은 이런 cardiac glycoside 중에 하나로써 Na+, K+-ATPase를 억제한다. 예전부터 심혈관계 질환에 쓰이던 약물이지만 유방암세포의 증가를 억제한다는 것 또한 최근에 밝혀졌다. Ouabain 과 TCTP 의 상관관계성에 대해 연구하고자 먼저 HeLa, MCF10A 에 다양한 농도의 ouabain 을 처리한 후 cell viability 를 관찰했다. 암 세포인 HeLa 에서는 100 nM 에서부터 세포독성이 나타나는 반면 MCF10A 에서는 1 μM 부터 세포독성이 나타났다. 이 때 세포 내 TCTP level 을 측정하였더니, HeLa 의 경우 1, 10, 50 100 nM ouabain 을 처리하였을 때 dose-dependent 하게 TCTP level 이 감소하는 것을 확인하였다. 반면 MCF10A 에서는 같은 농도의 ouabain 을 처리하여도 TCTP level 의 감소가 일어나지 않았으며 오히려 살짝 증가하는 것으로 나타났다. 따라서 ouabain 의 암세포 특이적 반응은 세포 내 TCTP level 의 변화에 기인 하는 것으로 보인다. 이와 더불어 TCTP 의 activity 를 조절하는 약물을 찾고자, In silico screening 을 통하여 TCTP 와 결합할 수 있는 약물 중 제일 잘 결합할 것 같은 물질을 골라 In Silico Drug 1 (ISD1) 이라 명명하였다. ISD1는 primary cultured vascular smooth muscle cell (VSMC) 에서 1 mM까지 처리하여도 세포독성을 일으키지 않았다. 또한 ISD1을 처리한 VSMC에서는 세포 내 TCTP의 변화 또한 보이지 않았다. 그래서 TCTP의 활성 변화를 보기 위해 세포 내 Ca2+ 농도를 측정하였다. ISD1을 5분간 처리하였을 때 calcium transient 가 control 에 비해 증가하는 것으로 나타났다. 또한 primary cultured cardiomyocyte 에 ISD1을 5분간 처리 후 바로 세포 수축 정도를 확인한 결과 ISD1을 처리하였을 때 control 보다 더 많이 수축하는 것으로 나타났다. 따라서 ISD1은 sarcoplasmic reticulum의 calcium release에 관여하는 것으로 보이며 TCTP와의 연관성은 좀 더 연구가 필요하다.-
