dc.description.abstract자외선은 피부 광노화와 밀접한 관련이 있다. 세포 외 기질에는, 콜라겐, 피브로넥틴과 같은 구성 단백질들의 합성을 억제하고 분해작용을 유도하는 matrix metalloproteinases-1 (MMP-1)가 있다. 자외선 B(290 nm-320 nm)에 노출이 되면, 이 MMP-1의 발현 및 분비작용이 증가하고 결국, 광노화가 일어나게 된다. 또한, 활성산소의 생성이 증가되고 세포사멸이 유도된다. 펩토이드는 펩티드모사물질로, 뛰어난 안정성과 활성을 지녀 약리학적으로 널리 활용되고 있지만, 아직 화장품 성분으로는 연구가 활발히 진행되지는 않았다. 그리하여 본 연구에서는, 6량체 펩토이드인 PAL-12의 자외선B에 의하여 유도되는 광노화에 대한 효과를, 사람 섬유아세포(Human dermal fibroblasts)와 3D 인공피부모델인 Keraskin-FTTM 이용하여 확인하였다. 그 결과, PAL-12를 처리하였을 시에는 MMP-1 발현량과 분비량이 감소, 콜라겐과 피브로넥틴의 발현량은 약간 증가하는 것을 확인할 수 있었다. 세포사멸과 활성산소 생성에는 별 영향을 주지 않았다. 그러나 자외선 B에 의해 활성화 되는 상피 성장 인자 수용체(EGFR), 세포 외 신호 조절 인산화 효소(ERK), 활성인자(AP-1)의 구성 단백질인 c-Jun 발현량을 약간 억제하는 것을 확인하였다. 이 결과들을 종합해 보면, PAL-12는 자외선 B에 의해 유도된 광노화를 억제하는 효능을 가지는 것으로 평가된다.;Solar ultraviolet radiation is closely associated with skin photo-aging. UVB (290 nm-320 nm) irradiation causes the production of matrix metalloproteinases-1 (MMP-1), which is related to the degradation of extracellular matrix (ECM) and, suppression of the synthesis of dermal structural proteins, such as collagen and fibronectin. Furthermore, UVB induces reactive oxygen species (ROS) generation and cell necrosis. Peptoids are a class of peptidomimetics whose pharmacological activities are widely known as their remarkable stability and biological activity. However, utilities of peptoids as cosmetic ingredients have not been widely studied. In this study, we demonstrated the effect of PAL-12, a new hexa-peptoid on UVB-induced photo-aging by using human dermal fibroblasts (HDFs) and a 3D We found that UVB-induced MMP-1 mRNA expression was inhibited by the treatment with PAL-12. Also, secretion of MMP-1 was decreased as determined by Enzyme-linked immunosorbent assay (ELISA). The mRNA expression levels of collagen-1 and fibronectin-1 were slightly increased by PAL-12 when compared with that of UVB irradiated control. While cell necrosis and ROS generation were not affected by PAL-12, UVB-induced activation of epidermal growth factor receptors (EGFR), extracellular signal-regulated kinase (ERK) and c-Jun were suppressed by the treatment with PAL-12. These results suggest that PAL-12 might be effective against UVB-induced skin photo-aging.-
