Abstract
본 논문은 실시간으로 생체 내 가스를 측정할 수 있는 전류측정방식의 전기화학적 미세 센서를 개발하는 기술들 및 이를 이용한 생물학적 응용연구에 대해 기술하고 있으며 이는 센서의 종류에 따라 크게 두 주제로 나뉜다. 제 1장에서는 일산화탄소, 일산화질소와 산소를 동시 측정할 수 있는 삼중 미세센서의 제작하는 기술과 이를 응용한 생물학적 연구에 대해 기술한다. 제 2장에서는 황화수소를 단독으로 측정할 수는 센서의 제작방법과 발전방향에 대해 다룬다. 제 1장에서는 기존의 삼중 센서 타입을 분리하여 각 전극에서 단독으로 측정할 수 있는 일산화탄소/일산화질소/산소 삼중센서로 개발한 내용을 다룬다. 각 작업전극의 표면은 표적 기체에 따라 전착되는 금속과 막을 구분하여 개별적으로 개질된다. 표적 물질에 대해 높은 감응도와 선택성을 갖도록 인가 전위를 결정하여 센서를 최적화한다. 고성능의 전극을 이용하여 쥐의 신장 단면에서 실험을 진행하여 피질과 수질에서의 각 기체의 농도 분포를 확인한다. 제 2장에서는 황화수소를 측정하는 센서를 개발하고 발전시킨다. 센서의 기본형태는 제 1장의 일산화탄소 측정 전극과 유사하다. 금을 전착한 작업전극을 다른 두 종류의 막(trimethylchlorosilane과 광경화된 소수성 고분자)을 각각 입혀서 황화수소를 단독으로 측정하는 센서를 개발한다. 두 종류의 센서 모두 황화수소에 감응도를 보이며, 이후 진행한 생물학적 방해종 실험을 통해 막의 구성을 조절하여 선택성 개선이 필요함을 확인한다.;This thesis is composed of two chapters: Chapter I deals with a subject on the fabrication and biological application of a CO/NO/O2 triple amperometric microsensor with high performance. Chapter II describes the fabrication and development of a H2S single sensor. In chapter I, an amperometric CO/NO/O2 triple microsensor was fabricated and its high performances were demonstrated, such as high sensitivity, selectivity and stability with fast response time. In-vitro experiments with rat kidney was conducted for validating the sensor feasibility. The dynamic concentration changes of CO, NO and O2 levels were attained for the cross section of rat kidney by the lateral scanning analysis and location-dependent analysis. The tendency of detected concentrations at medulla and cortex was definitely correspondent with the results of previous researches. The triple sensor was successfully fabricated with high performances: especially the ability to detect a single target gas separately at each working electrode and to be used for qualitative and quantitative measurements. In chapter II, an amperometic H2S single microsensor was newly developed. The fundamental step for the fabrication was similar to that of CO sensor of the triple sensor in chapter I. The sensor was prepared with two different membrane coatings: one is trimethylchlorosilane, and the other is a polymer of photocurable fluorinated oligomer. Both sensors had sensitivity and selectivity to H2S over CO and NO, but not for all interferents tested. The results of two sensor types showed the possibility to detect only H2S with high sensitivity. Further fine optimization study is needed to overcome the current limitation of the sensor.
