dc.description.abstractParkinson’s disease (PD) is a common progressive neurodegenerative disease, pathologically characterized by the selective loss of dopaminergic neurons in the substantia nigra (SN). Although the exact mechanism remains unknown, oxidative stress has been suggested as a major underlying cause of PD. The Oxi-gene family was originally discovered by microarray analysis in SN4741 cells, dopaminergic cell line. Oxidative stress altered the expression of the Oxi-gene family, including Oxi-α and Oxi-β. Oxidative stress reduced and increased Oxi-α and Oxi-β expression, respectively. Here, I demonstrated that Oxi-α and Oxi-β exerted opposing effects on autophagy, a catabolic process involved the cellular degradation of protein aggregates and organelles. Recent studies have revealed that abnormal autophagy occurs in neurodegenerative diseases, including PD. However, the specific regulatory mechanisms of autophagy remain largely unknown. In this study, Oxi-α and Oxi-β proteins were identified as novel autophagy regulators in pathological and pharmacological models of PD. I employed two models of autophagy to characterize the mechanism of Oxi-α under oxidative stress. First, it was shown that Oxi-α protein expression was significantly reduced during oxidative stress-induced cell death. In accordance with reduced Oxi-α protein levels, autophagy was activated during oxidative stress. Overexpression of Oxi-α activates mammalian target of rapamycin (mTOR) phosphorylation and subsequently suppressed autophagosome formation. Second, in the insulin-challenged model, which mimics the physiological condition in which autophagy is reduced, expression of Oxi-α was dramatically increased, suggesting the anti-autophagic effect of Oxi-α. Molecular and structural analyses of Oxi-α protein have revealed that Oxi-α directly bound to G protein α subunit i3 (Gαi3), which plays a crucial role in the anti-autophagic action of insulin. Furthermore, 3-D protein structure analysis demonstrated that Oxi-α protein possessed a hydrophobic pocket that specifically bound to phosphatidylethanolamine (PE), which is an essential lipid for autophagosome formation. Interestingly, a mutant Oxi-α protein which lacked binding affinity for PE also significantly decreased the binding affinity to Gαi3. Moreover, overexpression of the mutant Oxi-α increased autophagy and decreased the anti-autophagic effect of insulin. For mechanistic studies of Oxi-β, I used 6-hydroxydopamine (6-OHDA) and staurosporine models. Oxidative stress increased Oxi-β expression, and comparable results were observed in both the 6-OHDA and staurosporine models. Treatment with 6-OHDA increased autophagy along with upregulation of Oxi-β protein expression. Oxi-β protein overexpression induced autophagy as demonstrated by lowered mTOR phosphorylation and increased numbers of autophagosomes. Gene-chip analysis led me to identify Fbxw5 as being significantly upregulated in parallel to Oxi-β protein overexpression. Moreover, Fbxw5 directly bound to Oxi-β. Fbxw5 is a tuberous sclerosis 2 (TSC2) binding protein that leads to TSC2 ubiquitination by a CUL4 E3 ligase. Increased Oxi-β protein competed with TSC2 to bind Fbxw5 and suppressed TSC2 ubiquitination and degradation. TSC2 accumulation inhibited mTOR phosphorylation and increased autophagy. Next, treatment of staurosporine, as an apoptosis inducer, induced Oxi-β expression and activated mTOR-dependent autophagy. Moreover, staurosporine induced Parkin-PINK1-dependent mitophagy. Staurosporine induced more severe effects on PINK1 null cells, in which mitophagy was inactivated. The autophagy inhibitor bafilomycin A1 increased susceptibility to cell death in the presence of staurosporine treatment. In conclusion, the results of the present study suggest that Oxi-α and Oxi-β act as an autophagy regulators in cellular models of PD. These results may contribute to the development of a therapeutic strategy targeting of oxidative stress-induced autophagy in the pathogenesis of PD.;파킨슨 병은 수명이 길어짐에 따라 그 환자수가 급격히 늘어나고 있는 대표적인 신경퇴행성 질환 중의 하나이다. 아직까지 그 정확한 기전은 알려지지 않았으나 산화 스트레스가 가장 큰 원인중의 하나로 제시되어 오고 있다. 파킨슨 병의 대표적 병리학적 특징으로서 뇌의 흑색질에 존재하는 도파민 신경의 특징적인 사멸을 들 수 있는데, 본 연구에서는 순수한 도파민 세포주인 SN4741에서 산화 스트레스에 의해 특이적으로 조절되는 Oxi-α와 Oxi-β의 분자적 길항작용기전에 대해 논하였다. SN4741 세포주에 산화스트레스 유도 물질 (H2O2)을 처리하게 되면, apoptosis와 오토파지가 유도되면서 Oxi-α의 발현은 현저히 떨어지게 되고 Oxi-β는 상승하게 되는데, 흥미롭게도 이 두 개의 유전자는 오토파지라는 세포 내 기전을 반대로 조절하는 기능을 가진 것이 밝혀졌다. 오토파지란 세포의 ‘자가포식시스템’ 이라 불리며 세포가 절식되었을 때나 스트레스를 받을 때 활성화 됨이 알려져 있다. 오토파지는 매우 복잡한 신호전달을 통해 조절되며 평상시에도 일정한 수준으로 그 활성을 유지하여 세포 항상성 유지에 중요한 역할을 하는 기전이다. 최근 연구에서 이 오토파지 시스템은 세포 사멸과 밀접한 연관이 있고, 비정상적인 오토파지 활성이 여러 병리학적 모델에서 관찰되었다. 그러나 아직까지는 정확한 분자적 조절기전은 확립되어 있지 않으며, 특히 산화스트레스에 의한 오토파지 이상이 유도되는 기전은 미 규명 상태이다. 이러한 관점에서 Oxi-α와 Oxi-β가 도파민 신경 내의 산화스트레스에 의한 오토파지 이상유발 및 세포사멸과의 연관성 연구는 파킨슨병 기전연구에 기여를 할 수 있으리라 사료된다. Oxi-α분자의 작용 기전 연구를 위해 두 가지 모델을 사용하였는데 첫째로 산화스트레스 모델을 사용하였고, 둘째로 인슐린 모델을 사용하였다. 산화스트레스 모델에서 Oxi-α는 산화스트레스로 인한 세포사멸이 유도되었을 때 그 발현이 감소하였고, 이는 오토파지의 증가와 함께 관찰되었다. Oxi-α단백질을 과발현 시켰을 때는 오토파지를 조절하는 대표적인 표적 단백질인 mTOR의 인산화가 증가되었고, 오토파지의 감소를 유발하였다. 흥미롭게도 오토파지가 감소하는 생리학적 조건인 인슐린 모델에서는 세포 내 Oxi-α단백질 발현 증가가 관찰되었다. 주목할 만한 결과로서 Oxi-α와 활성화된 G protein α subunit i3 (Gαi3)이 직접 결합하는데, Gαi3는 인슐린의 항 오토파지 효과에 필수적인 요소로 알려져 있다. 또한 Oxi-α단백질의 3차 구조분석을 통해 phosphatidylethanolamine (PE)을 특이적으로 수용할 수 있는 hydrophobic 구조를 확인하였는데 이 PE는 오토파고좀이 형성될 때 필수적인 지질로 알려져 있다. Mutation을 통해 PE가 결합할 수 없는 mutant Oxi-α를 제작하여 과발현 시킨 결과 Oxi-α의 오토파지 저해기능이 사라지고, Gαi3와의 결합 역시 현저하게 감소되는 것을 확인할 수 있었다. 이 같은 결과를 종합하여 볼 때 Oxi-α는 Gαi3와 밀접하게 연게하여 인슐린의 오토파지 조절 기능을 포함하여 여러 항오토파지 생리기전에 관여하는 단백질임을 알 수 있다. 앞서 언급한 Oxi-α분자 또한 오토파지 조절 기전 연구를 위해서 두 가지 모델을 사용하였는데 첫째는 대표적 파킨슨병 동물 모델인 6-hydroxydopamine (6-OHDA)를 사용하였고 또 다른 한가지는 staurosporine 모델을 사용하였다. 산화스트레스를 통한 세포사멸이 유발되었을 때 Oxi-β는 Oxi-α와 반대로 그 발현이 현저하게 증가하였는데, 6-OHDA 모델에서도 동일한 유전자 발현 양상이 뚜렷하게 관찰되었다. 6-OHDA가 세포에 투여되었을 때 Oxi-β발현이 증가함에 따라 오토파지 역시 증가되는 것을 확인할 수 있었고, 또한 Oxi-β단백질이 과발현 되었을 때 오토파지가 증가됨을 확인할 수 있었다. Oxi-β단백질이 과발현 되었을 때 발현이 조절되는 유전자들을 Gene chip analysis 를 통해 분리해 본 결과 Fbxw5 단백질이 가장 큰 폭으로 증가하였으며 흥미롭게도 이 Fbxw5 단백질은 Oxi-β 단백질과 직접 결합함을 면역침강법을 통해 확인할 수 있었다. Fbxw5 단백질은 CUL4 E3 ligase complex의 TSC2 binding receptor로서 작용하여 TSC2 단백질을 유비퀴틴화 시켜 분해하는 역할을 하는데, Oxi-β가 증가하게 되면 TSC2와 경쟁하여 TSC2 단백질 분해 작용의 저해 및 축적을 유발하였다. 따라서 Oxi-β단백질은 Fbxw5 단백질과 결합함으로써 TSC2 단백질을 증가시키고 mTOR 의 인산화를 억제시켜 오토파지의 증가를 조절하는 새로운 오토파지 regulator임을 이 논문에서 증명하였다. Oxi-β기전연구의 두 번째 모델로 사용된 Staurosporine은 기존에 apoptosis를 유발시키는 대표적인 약물로서 사용되었고, 오토파지와의 연관성이 자세히 알려져 있지 않았다. 그러나 이를 세포에 투여하였을 때 Oxi-β의 발현이 현저하게 증가되는데, 이를 토대로 staurosporine으로 세포사멸을 유도할 때 변화되는 단백질을 분석 해 본 결과, staurosporine은 오토파지와 미토파지 (mitophagy)를 매우 현저하게 증가시킴을 관찰할 수 있었다. 주목 할만 한 결과로서, staurosporine은 미토파지가 유전적으로 억제된 PINK1 K.O 세포에서 더 급격한 세포사멸을 일으켰으며, 오토파지 및 미토파지를 억제하는 약물인 bafilomycin A1과 함께 투여되었을 때도 세포사멸은 증가하였다. 위 연구를 통해, Oxi-α와 Oxi-β는 각각 반대로 오토파지를 조절하는 유전자로써 이는 정교하게 오토파지 시스템을 조절할 수 있는 분자적 타깃이 될 수 있으며, 파킨슨병의 병리학적 현상에서 나타나는 오토파지 장애를 치료하기 위한 새로운 치료제 개발에 Oxi-α와 Oxi-β기전 연구가 사용 될 수 있음을 시사한다.-
