dc.description.abstractSociability is a behavioral disposition of interacting with other conspecific members. Social animals including humans live on social behavior in daily life, and therefore impairment of sociability is problematic in getting and surviving well in social and sociable world. Sociability behavior is often impaired in many psychiatric illnesses including depression, anxiety disorders, schizophrenia, and autism. Despite the behavioral difference between humans and rodents, various rodent models have been introduced to unravel the property and mechanism underlying sociability, but the methodological easiness and usefulness as a behavioral assay needs to be improved further. In the present study, the U-field test, a novel behavioral test, was developed to use to measure sociability of mice. The U-field consists of the two symmetrical rectangular fields that are attained by partitioning the open field (45 x 45 x 40 cm) with a wall to the central point, so that each field has closed and open squares, configurating a “U-shaped field”. A circular grid cage (12 cm in diameter x 33 cm in height) was placed on each side of the closed square. When the closed square is placed with a social target in a circular grid cage, it is called target-zone, whereas the closed square is unanimated, it is called the non-target-zone. Likewise, relevant fields (each including closed and open squares) are named target and non-target fields, respectively. First, the U-field test was used to examine the known behavior, called behavioral preference for novel animals. It was demonstrated that the U-field test was sensitive enough to read animal’s behavioral preference for novel vs. familiar mice. Next, the U-field test was applied to the analysis of stress and depression –related behavior. When both fields were equipped with unanimated cages, normal B6 mice and stressed mice visited both zones evenly, so did they both fields. Normal B6 mice showed a behavioral preference for the target zone containing a social target, while mice treated with daily 2 h restraint for 14 days (2h×14d stress) apparently showed reduced preference to the target zone, without showing reduced locomotion, suggesting mice with 2h×14d stress showed reduced sociability. Treatment of stressed mice with imipramine (an anti-depressant) reversed their suppressed sociability behavior. Thus, the U-field is a useful method to trace behavioral differences between sociable and unsociable mice. Finally, it was demonstrated that the U-field test provides a predictive value that select stress resilient mice from stress susceptible mice. By combining with the tail suspension test, the U-field test was shown to provide a novel resolving power to filter out stress resilient mice from stress susceptible mice. Availability of a proper animal model like the U-field test and its application may facilitate to unravel the neural mechanism underlying social behavior-based higher nervous function, which may include stress, depression, autism, and schizophrenia. ;사교성(sociability)은 다른 개체와의 상호 작용에서 보이는 행동적 기질이다. 사람을 포함한 사회적 동물은 매일 사회적인 행동을 보이며, 이러한 과정에 장애가 발생할 경우, 사회에 적응하고 생존하기에 심각한 문제를 초래하게 되며, 우울증, 불안 장애, 정신분열증, 자폐증 등의 다양한 정신질환에서 사회적 행동이 줄어드는 사례를 볼 수 있다. 사람과 동물 간에 행동학적으로 차이가 있음에도 불구하고, 사회적 행동과 그에 대한 메커니즘을 연구하기 위하여 다양한 설치류 모델들이 소개되고 있다. 하지만, 방법적으로 쉽고 유용한 행동실험을 위해서는 더욱 많은 연구가 필요한 실정이다. 본 연구에서는, 동물의 사회성을 측정하는 U자형 영역 실험(U-field test)을 새롭게 개발하였다. 오픈 필드(open field)의 가운데에 칸막이를 두어, 두 개의 닫힌 구역(zone)과 두 개의 열린 구역으로 구성된 U자 모양의 영역(field). 높이 33 cm와 직경 12 cm의 원통 철망 우리가 각 닫힌 구역에 위치하며, 한 쪽 우리에는 표적이 되는 생쥐(target mouse)를 넣어두고, 다른 한 쪽 우리는 비어둔다. 표적이 들어있는 구역은 표적 구역(target zone), 비어있는 우리가 놓여있는 구역은 비표적 구역(non-target zone)이라 명명한다. 첫째로, 이 U자형 영역 실험은, 피험 생쥐(subject)와 그에 대하여 친숙하지 않은 낯선 생쥐를 이용하여, 동물의 행동학적 선호도를 조사하는 실험에 이용 가능하다. 이러한 사실은, 동물이 U자형 영역 실험에서 낯선 대상과 친숙한 대상에 대하여 특정 선호 행동을 보이는 것을 통해 알 수 있다. 둘째로, U자형 영역 행동실험은 스트레스와 우울증 관련 행동을 분석에도 응용 가능하다. U자형 영역 실험의 한 가운데에 놓여진 평범한 C57BL/6 생쥐는, 자유롭게 양쪽 영역을 돌아다니며, 그 결과, 표적 생쥐가 존재하는 표적 구역을 더욱 선호하는 행동 경향을 보인다. 반면, 하루에 2시간씩 14일 동안 감금 스트레스 처치를 받은 생쥐는 표적 구역을 덜 선호하는 행동을 보였으며, 스트레스를 받은 생쥐에게 항우울제인 이미프라민(imipramine)을 처치하였을 경우에는 감소되었던 사회성 정도가 회복되었다. 반면, 양쪽 영역에 아무 표적 동물도 없는 경우에는, 평범한 생쥐와 스트레스를 받은 생쥐 모두, 두 영역과 구역을 균등한 시간 동안 머무르고 움직였다. 즉, U자형 영역 실험은 사회적인 생쥐와 비사회적인 생쥐의 행동 차이를 분석하고 측정하기에 유용한 도구가 될 수 있다. 마지막으로, U자형 영역 행동실험은 스트레스에 취약한 생쥐(stress susceptible mice)와 스트레스 저항 생쥐(stress resilience mice)를 구분해 낼 수 있다. 또한, 꼬리 매달기 실험(tail suspencion test)과 같은 다른 행동실험 결과와 U자형 영역 실험의 결과를 연계하여, 스트레스에 저항성을 보이는 집단을 선별해 낼 수 있었다. 이러한 접근은 효과적이며 손쉽게 이용이 가능하고, 본 논문에서 처음 시도되었다. U자형 영역 행동실험과 그의 응용은, 고등인지기능이 연계되어있는 스트레스, 우울증과 같은 질환 연구의 올바른 접근을 가능하게 할 것이다.-
