Abstract
Streptococcus pneumoniae (S. pneumoniae) is a commensal bacterium of human upper respiratory tract. In healthy individuals, S. pneumoniae colonizes without any symptoms, but its invasion through lower respiratory tract develops pneumococcal pneumonia and gives high lethality to the old and the weak worldwide. Especially, pneumococcal pneumonia is the number one cause of community-acquired pneumonia (CAP) and major cause of morbidity and mortality. STIP1 homologous and U box-containing protein 1 (STUB1), also known as carboxyl-terminus of Hsc70 interacting protein (CHIP), is a chaperon-dependent E3 ligase in the ubiquitin proteasome system. CHIP by leading target substrates for proteasomal degradation regulates protein stability and thereby regulates various biological functions such as tissue fibrosis, tumor metastasis, and allergic inflammation. However, the role of CHIP on regulating inflammatory response in infectious diseases is not characterized at all. In this study, it is showed that CHIP is highly expressed in lung alveolar epithelial cells and inhibits S. pneumoniae-induced transcriptional activation of Activator protein-1 (AP-1), which is a transcription factor regulating transcription of various genes involved in innate immune and inflammatory responses. For the first time, a mixed-lineage protein kinase 3 (MLK3) was identified as a key signaling molecule mediating S. pneumonia-induced up-regulation of pro-inflammatory cytokines. Activated MLK3 by S. pneumoniae induces phosphorylation of c-Jun N-terminal kinase (JNK) MAPK, and subsequently activates AP-1 and transcription of pro-inflammatory genes. CHIP inhibits S. pneumoniae-induced inflammation by inhibiting MLK3-JNK1/2 MAPK signaling pathway by promoting degradation of MLK3. Moreover, CHIP heterozygous mutant mice showed enhanced alveolar microvascular leakage and hemorrhage, enhanced migration of inflammatory cells into airway, and enhanced expression of pro-inflammatory cytokines compared to those of wild-type (WT) mice following inoculation with S. pneumoniae, which may result in higher mortality in CHIP heterozygous mice. Furthermore, it was also found that activation of CHIP using chemical activator of CHIP, 17-AAG, inhibited S. pneumoniae-induced up-regulation of pro-inflammatory cytokines in mice in vivo. Taken together, these data indicate that CHIP acts as an important endogenous negative regulator of S. pneumoniae-induced inflammation by inhibiting MLK3-JNK1/2 MAPK signaling pathway, and thus modulating CHIP expression might be a novel therapeutic strategy for treating S. pneumoniae infections.;Streptococcus pneumoniae(폐렴구균 또는 폐구균)는 사람의 상부호흡기도에 위치한 공생세균이다. 일반적으로 면역력이 정상적으로 작동하는 건강한 성인에서는 특이적인 임상증상 없이 상재하나 가지 이유로 인하여 숙주의 면역력이 감소하게 되면 하부 호흡기도로 침투하여 폐렴과 같은 침습성 질환을 유발할 수 있다. 폐렴구균은 지역사회획득 폐렴의 가장 흔한 원인으로 면역력이 취약한 환자나 유아 및 노인에게서 높은 치사율을 보이는 대표적인 세균성 호흡기 병원체로 잘 알려져 있다. STIP1 homologous and U box-containing protein 1(STUB1)은 ubiquitin proteasome system의 chaperon-dependent E3 ligase로 carboxyl-terminus of Hsc70 interacting protein(CHIP)이라는 이름으로 더 잘 알려져 있다. CHIP은 표적 단백질에 ubiquitin을 표지 함으로서 프로테아좀에 의한 표적 단백질의 분해를 유도하여 표적물질이 관여하는 다양한 생물학적 현상을 조절한다. 최근 연구에서는 조직의 섬유화, 암세포의 전이, 및 알레르기성 염증반응에서 CHIP이 중요한 역할을 함이 밝혀졌다. 하지만 아직까지 감염성 질병에서의 염증반응에서 CHIP의 역할에 대한 연구는 거의 알려진 바가 없다. 이 연구에서는 CHIP이 폐상피세포에서 발현되며, 폐렴구균에 의해 활성화 되는 전사조절인자인 AP-1의 활성이 CHIP에 의해 조절된다는 것을 확인하였다. 또한 이 실험의 결과, MLK3 인산화효소가 폐렴구균에 의하여 활성화 되며, 활성화된 MLK3는 신호전달 물질인 JNK1/2 MPAK의 인산화를 유도하여 폐렴구균에 의한 염증성 사이토카인의 발현을 조절함을 확인하였다. 또한 CHIP이 MLK3의 분해를 유도하여 폐렴구균에 의한 염증반응을 억제하는 중요한 염증억제 인자로 작용함을 확인하였다. CHIP의 발현이 저하된 CHIP 변이 마우스에서는 폐렴구균에 의한 폐모세혈관투과성과 폐포출혈이 증가하고, 염증성 사이토카인의 발현과 염증세포의 폐포 내 침윤이 증가하였다. 그 결과 폐렴구균에 의한 마우스의 폐사가 CHIP 변이 마우스에서 증가되어 나타남을 확인할 수 있었다. 마지막으로 CHIP 활성제인 17-AAG에 의하여 폐렴구균에 의한 폐조직에서의 염증성 사이토카인의 발현이 현저히 억제됨을 확인하였다. 이러한 발견은 CHIP이 폐렴구균에 의한 염증반응을 조절하는 중요한 세포 내 음성조절인자이며, CHIP의 발현을 조절 함으로서 폐렴구균에 의한 염증 및 이로 인한 치명적인 손상을 치료할 수 있는 치료기술이 개발 가능함을 시사한다.
